Abstract Background Melatonin played an essential role in numerous vital life processes of animals and captured the interests of plant biologists because of its potent role in plants as well. As far as its possible contribution to photoperiodic processes, melatonin is believed to act as a growth regulator and a direct free radical scavenger/indirect antioxidant. The objective of this study to identify a precise melatonin concentration for a particular application method to improve plant growth requires identification and clarification. Methods This work establishes unique findings by optimizing melatonin concentration in alleviating the detrimental effects of drought stress in maize. Maize plants were subjected to drought stress (40–45% FC) after treatments of melatonin soil drenching at different concentrations (50, 100, and 150 µM) to consider the changes of growth attribute, chlorophyll contents, photosynthetic rate, relative water content (RWC), chloroplast ultrastructure, endogenous hormonal mechanism, and grain yield. Results Our results showed that the application of melatonin treatments remarkably improved the plant growth attributes, chlorophyll contents, photosynthetic rate, RWC, hormonal mechanism, and grain yield plant−1 under drought conditions at a variable rate. Conclusion Our current findings hereby confirmed the mitigating potential of melatonin application 100 µM for drought stress by maintaining plant growth, hormone content, and grain yield of maize. We conclude that the application of melatonin to maize is effective in reducing drought stress tolerance. Graphical Abstract

Background: This study was conducted during the 2020/2021 cropping season at Setif Agricultural Experimental Station, it aims to assess the efficiency of using numerical image analysis (NIA) in the selection of durum wheat genotypes in semi-arid areas. Methods: The genetic materials used in this study consist of 11 advanced lines and 4 genotypes of which 3 are local landraces used as control to evaluate their performance, the genotypes tested were sown in a randomized block design (RDB) with three replications. each plot consisted of 6 lines of 10 m long spaced of 0.2 m witdth makes 12 m2 as plot area. Result: Analysis of variance showed that all the parameters measured numerically (senescence and total reflectance) had a very high genotypic significance. The chlorophyll content at full heading showed a very highly significant genotypic effect. Thousand kernels weight, number of spikes per meter square, number of days to heading and plant height had a significant genotypic effect. The correlation study showed that all senescence parameters were significantly correlated. A significant and negative correlation was observed between chlorophyll contents; average of velocity and total reflectance. Grain yield was highly and significantly correlated with thousand kernels weight and number of spikes per meter square. Number of spikes per meter square was significantly and positively correlated with average of velocity and negatively correlated with sum of temperatures at mid-senescence. Number of days to heading was significantly and negatively correlated with senescence average and maximum of senescence average. A significant correlation was observed between plant height and sum of temperatures at mid-senescence.

The southwestern mountains of Hainan Island are distributed in the southernmost tropical karst landscape of China, and the unique hydrological structure and frequent solifluction droughts lead to double water stress for local plants. Highly heterogeneous water environments affect the water–use characteristics of plants. Plants develop local adaptative mechanisms in response to changes in the external environment. In this paper, hydrogen–oxygen and carbon stable isotope technology, and physiological index measurements were applied to determine the leaf traits, water–use efficiency, and photosynthetic characteristics of Impatiens hainanensis leaves in dry and foggy seasons, hoping to expound the adaptation mechanism of I. hainanensis leaves to the water dynamics in dry and foggy seasons. In dry and foggy seasons (November 2018 to April 2019), the leaves of I. hainanensis at low and medium altitudes have the following combination of traits: larger leaf dry weights, leaf areas, and specific leaf areas; smaller leaf thicknesses and leaf dry matter contents; and higher chlorophyll contents. In comparison, the leaves of I. hainanensis at high altitudes have the following combination of traits: smaller leaf dry weights, leaf areas, and specific leaf areas; larger leaf thicknesses and leaf dry matter contents; and lower chlorophyll contents. The leaves of I. hainanensis can absorb fog water through their leaves. When the leaves are sprayed with distilled water, the water potential is low, the water potential value gradually increases, and the leaves have a higher rate of water absorption. The leaves of I. hainanensis at low and medium altitudes have the following water–use characteristics: high photosynthesis, high transpiration, and low water–use efficiency. At high altitudes, the Pn of I. hainanensis decreases by 8.43% relative to at low altitudes and by 7.84% relative to at middle altitudes; the Tr decreased by 4.21% relative to at low altitudes and by 3.38% relative to at middle altitude; the WUE increased by 16.61% relative to at low altitudes and increased by 40.79% relative to at middle altitudes. The leaves of I. hainanensis at high altitudes have the following water–use characteristics: low photosynthesis, low transpiration, and high water–use efficiency. I. hainanensis develop different physiological mechanisms of water adaptation by weighing the traits of the leaves and their use of light and water to obtain resources during dry and foggy seasons.

Chlorophyll fluorescence (ChlF) is a non-invasive technique that can be potentially used in postharvest research to gain useful information on early responses to postharvest stresses. This study was conducted to validate the application of ChlF transient analysis in determining the postharvest changes in photosynthetic apparatus in three ornamental foliage species, i.e., Cordyline fruticosa ‘Willy’s Gold’ and ‘Rubra’, Dracaena sanderiana ‘White’, and Nephrolepis exaltata. Salicylic acid (100 and 300 mg·L−1), glucose (10 g·L−1), and their combinations were used as holding solutions with control treatment (distilled water) at room temperature (25±2°C). Vase life was evaluated using OJIP analysis. OJIP parameters, i.e., specific energy fluxes per reaction center (ABS/RC, TR/RC, ET/RC, and DI/RC), flux ratios (maximum quantum yield of primary photochemistry-φPo), electron transport efficiency (ψo), and quantum yield of electron transport (φEo), and performance index (PI) were recorded every other day, using a fluorometer (FluorPen 100). Leaf chlorophyll contents of all species and anthocyanin contents of two cordyline cultivars were determined. Data were subjected to ANOVA in a completely randomized design. Mean separation was done by DMRT (p ≤ 0.05). Clear variations in ChlF were observed in every foliage species with the time. OJIP analysis showed species-depended variations. The higher ABS/RC and DI/RC were recorded for D. sanderiana and N. exaltata compared to the PI of those species. At the end of the experiment, the chlorophyll contents were decreased, while anthocyanin contents were increased. Consequently, chlorophyll fluorescence changes in photosynthetic apparatus can be used for the prediction of the postharvest stresses and longevity of cut foliage.

The effect of freeze-drying and gamma irradiation at 0.5, 1, and 2 kGy on the physicochemical composition (moisture, fat, ash, mineral, and chlorophyll contents), microstructure, total phenolic content, and antioxidant capacity of stevia leaves was investigated in the present study. The results obtained indicated that freeze-drying and gamma irradiation treatments caused significant reduction ( p  ≤ 0.05) of moisture and fat contents in comparison with those of commercial leaves, while ash content was not significantly affected. Mineral composition was analysed. Among the analysed elements, potassium and iron levels were higher in the freeze-dried and irradiated samples, respectively. The microstructure was analysed using a scanning electron microscope. Micrographs revealed that a higher porous size structure was obtained by freeze-drying, and degradation of cell wall structure was more clearly visualized by irradiation at 2 kGy. However, the main functional groups were stable as confirmed by Fourier transform infrared spectroscopy analysis. The effects on chlorophyll content, phenolic profile, and antioxidant properties were evaluated before and after ten months of storage. In terms of chlorophyll contents, the freeze-dried leaves exhibited the highest content. Chlorophylls a and b decreased when storage progressed for freeze-dried leaves as well as for gamma-irradiated leaves. Both preservation methods gave significant advantages in increasing the total phenolic content and DPPH scavenging activity. Moreover, a significant increase of bioactive compounds and antioxidant activity was observed as the gamma irradiation dose increased. In addition, the storage time increased the amounts of polyphenols and DPPH scavenging activity. After 10 months of storage, gamma-irradiated leaves had the highest total phenolic content as well as the DPPH scavenging activity followed by freeze-dried leaves. The results indicate that freeze-drying and gamma irradiation at the studied doses could be effective postharvest methods for preservation of stevia leaf quality.

Root diseases are one of the main forest nursery problems that have a significant impact on forest production which are caused by Fusarium solani. Rhizobacteria from healthy forest soils were isolated and screened in streak method to select antagonistic strains against F. solani. Two isolates showed high antagonistic activity and molecularly identified as Paenibacillus sp. and Pseudomonas sp. The capability of the Paenibacillus sp. and Pseudomonas sp. were tested in greenhouse plastic containers experiments against F. solani. Soil bacterization with Paenibacillus sp. and Pseudomonas sp. significantly protected thuja seedlings from F. solani compared to the untreated control seedlings. The containers added by Paenibacillus sp. and pseudomonas sp. are also showed plant growth promotion including shoot length, root length, dry and wet weights of the seedlings as well as the chlorophyll contents of the thuja seedlings compared to the untreated control plants. In this research it has been showing that the rhizobacterial treatments have potential to decrease the effect of fungal disease severity, promoting the plant growth and also helps plants to maintain a good health.

Environmental stresses may alter the nutritional profile and economic value of crops. Chemical fertilizers and phytohormones are major sources which can enhance the canola production under stressful conditions. Physio-biochemical responses of canola altered remarkably with the use of nitrogen/phosphorus/potassium (N/P/K) fertilizers and plant growth regulators (PGRs) under drought stress. The major aim of current study was to evaluate nutritional quality and physio-biochemical modulation in canola (Brassica napus L.) from early growth to seed stage with NPK and PGRs in different water regimes. To monitor biochemical and physiological processes in canola, two season field experiment was conducted as spilt plot under randomized complete block design (RCBD) with four treatments (Control, Chemical fertilizers [N (90 kg/ha), P and K (45 kg ha-1)], PGRs; indole acetic acid (IAA) 15g ha-1, gibberellic acid (GA3) 15g ha-1 and the combination of NPK and PGRs] under different irrigations regimes (60, 100, 120, 150 mm evaporations). Water stress enhanced peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), polyphenol oxidase (PPO), soluble sugar, malondialdehyde (MDA), proline contents as well as leaf temperature while substantially reduced leaf water contents (21%), stomatal conductance (50%), chlorophyll contents (10–67%), membrane stability index (24%) and grain yield (30%) of canola. However, the combined application of NPK and PGR further increased the enzymatic antioxidant pool, soluble sugars, along with recovery of leaf water contents, chlorophyll contents, stomatal conductance and membrane stability index but decreased the proline contents and leaf temperature at different rate of evaporation. There is positive interaction of applied elicitors to the water stress in canola except leaf area. The outcomes depicted that the combination of NPK with PGRs improved the various morpho-physiological as well as biochemical parameters and reduced the pressure of chemical fertilizers cost about 60%. It had also reduced the deleterious effect of water limitation on the physiology and grain yield and oil contents of canola in field experiments.

In recent years, the delay in sowing has become a major obstacle to high wheat yield in Jiangsu Province, one of the major wheat producing areas in China; hence, it is necessary to screen wheat varieties are resilient for late sowing. This study aimed to provide an effective, fast, and non-destructive monitoring method of soil plant analysis development (SPAD) values, which can represent leaf chlorophyll contents, for late-sown winter wheat variety screening. This study acquired multispectral images using an unmanned aerial vehicle (UAV) at the overwintering stage of winter wheat growth, and further processed these images to extract reflectance of five single spectral bands and calculated 26 spectral vegetation indices. Based on these 31 variables, this study combined three variable selection methods (i.e., recursive feature elimination (RFE), random forest (RF), and Pearson correlation coefficient (r)) with four machine learning algorithms (i.e., random forest regression (RFR), linear kernel-based support vector regression (SVR), radial basis function (RBF) kernel-based SVR, and sigmoid kernel-based SVR), resulted in seven SVR models (i.e., RFE-SVR_linear, RF-SVR_linear, RF-SVR_RBF, RF-SVR_sigmoid, r-SVR_linear, r-SVR_RBF, and r-SVR_sigmoid) and three RFR models (i.e., RFE-RFR, RF-RFR, and r-RFR). The performances of the 10 machine learning models were evaluated and compared with each other according to the achieved coefficient of determination (R2), residual prediction deviation (RPD), root mean square error (RMSE), and relative RMSE (RRMSE) in SPAD estimation. Of the 10 models, the best one was the RF-SVR_sigmoid model, which was the combination of the RF variable selection method and the sigmoid kernel-based SVR algorithm. It achieved high accuracy in estimating SPAD values of the wheat canopy (R2 = 0.754, RPD = 2.017, RMSE = 1.716 and RRMSE = 4.504%). The newly developed UAV- and machine learning-based model provided a promising and real time method to monitor chlorophyll contents at the overwintering stage, which can benefit late-sown winter wheat variety screening.

Early leaf senescence phenotype in soybean could be helpful to shorten the maturation period and prevent green stem disorder. From a high-density mutation library, we identified two early leaf senescence soybean mutant lines, els1-1 (early leaf senescence 1) and els1-2. The chlorophyll contents of both els1-1 and els1-2 were low in pre-senescent leaves. They degraded rapidly in senescent leaves, revealing that ELS1 is involved in chlorophyll biosynthesis during leaf development and chlorophyll degradation during leaf senescence. The causal mutations in els1 were identified by next-generation sequencing-based bulked segregant analysis. ELS1 encodes the ortholog of the Arabidopsis CaaX-like protease BCM1, which is localized in chloroplasts. Soybean ELS1 was highly expressed in green tissue, especially in mature leaves. The accumulation of photosystem I core proteins and light-harvesting proteins in els1 was low even in pre-senescent leaves, and their degradation was accelerated during leaf senescence. These results suggest that soybean ELS1 is involved in both chlorophyll synthesis and degradation, consistent with the findings in Arabidopsis BCM1. The gene els1, characterized by early leaf senescence and subsequent early maturation, does not affect the flowering time. Hence, the early leaf senescence trait regulated by els1 helps shorten the harvesting period because of early maturation characteristics. The els1-1 allele with weakly impaired function of ELS1 has only a small effect on agricultural traits and could contribute to practical breeding.

Background Polyploidy plays an important role in plant breeding and has widespread effects on photosynthetic capacity. To determine the photosynthetic capacity of the tetraploid variety Acer buergerianum Miq. ‘Xingwang’, we compared the gas exchange parameters, chloroplast structure, chlorophyll contents, and chlorophyll fluorescence parameters between the tetraploid Acer buergerianum ‘Xingwang’ and the diploid ‘S4’. To evaluate the effects of genome duplication on the photosynthetic capacity of Acer buergerianum ‘Xingwang’, the transcriptomes of the autotetraploid ‘Xingwang’ and the diploid ‘S4’ of A. buergerianum were compared. Methods The ploidy of Acer buergerianum ‘Xingwang’ was identified by flow cytometry and the chromosome counting method. An LI-6800 portable photosynthesis system analyzer was used to assess the gas exchange parameters of the tetraploid variety ‘Xingwang’ and diploid variety ‘S4’ of A. buergerianum. We used a BioMate 3S ultraviolet-visible spectrophotometer and portable modulated fluorometer to measure the chlorophyll contents and chlorophyll fluorescence parameters, respectively, of ‘Xingwang’ and ‘S4’. Illumina high-throughput sequencing technology was used to identify the differences in the genes involved in the photosynthetic differences and determine their expression characteristics. Results The single-cell DNA content and chromosome number of the tetraploid ‘Xingwang’ were twice those found in the normal diploid ‘S4’. In terms of gas exchange parameters, the change in stomatal conductance, change in intercellular CO2 concentration, transpiration rate and net photosynthetic rate of ‘Xingwang’ were higher than those of the diploid ‘S4’. The chlorophyll contents, the maximal photochemical efficiency of PSII and the potential photochemical efficiency of PSII in ‘Xingwang’ were higher than those of ‘S4’. The chloroplasts of ‘Xingwang’ contained thicker thylakoid lamellae. By the use of Illumina sequencing technology, a total of 51,807 unigenes were obtained; they had an average length of 1,487 nt, and the average N50 was 2,034 nt. The lengths of most of the unigenes obtained ranged from 200–300 bp, with an average value of 5,262, followed by those longer than 3,000 bp, with an average value of 4,791. The data revealed numerous differences in gene expression between the two transcriptomes. In total, 24,221 differentially expressed genes were screened, and the percentage of differentially expressed genes was as high as 46.75% (24,224/51,807), of which 10,474 genes were upregulated and 13,747 genes were downregulated. We analyzed the key genes in the photosynthesis pathway and the porphyrin and chlorophyll metabolism pathway; the upregulation of HemB may promote an increase in the chlorophyll contents of ‘Xingwang’, and the upregulation of related genes in PSII and PSI may enhance the light harvesting of ‘Xingwang’, increasing its light energy conversion efficiency.
