Anti-inflammatory and Antinociceptive Activities of the Hydroalcoholic Extract and the Volatile Fraction of Southeastern Brazilian Brown Propolis

Background: Lichens which are betide to mangroves are termed as Manglicolous Lichens (ML). As these ML are habituated under stress conditions, they are screened for unique metabolites and biological activities. Objective: The study aimed to establish the chemical and biological profile of ML, Graphis ajarekarii. Methods: The Ethyl Acetate Extract of G. ajarekarii (EAE) was subjected to chromatographic techniques and the obtained isolates were characterized by spectroscopic analysis. The hydroalcoholic extract of G. ajarekarii (AE), EAE, isolates and Hydroalcoholic Extract of host (HE) were evaluated for fibrinolytic (fibrin clot method), in vitro (protein denaturation method) and in vivo (formalin-induced rat paw oedema assay), anti-inflammatory and cytotoxicity (MTT assay) activities. Results: Chemical investigation of the EAE led to the isolation of two known compounds namely atranorin (1) and ribenone (2), which were confirmed by spectral data. The AE and EAE gradually lysed the fibrin clot with 94.54 and 65.07%, respectively, at 24 h. The AE inhibited protein denaturation of about 88.06%, while the standard (Indomethacin) with 93.62%. Similarly, the in vivo antiinflammatory analysis of AE (200 mg/mL) showed potent reduction of rat paw oedema than the standard, whereas EAE and 1 depicted moderate depletion. In addition, the AE revealed prominence inhibition on MCF-7, DU145 and K-562 with IC50 values of 69.5, 42.5 and 38 µg/mL, respectively, whereas the HE exhibited mild inhibitory profile against fibrin clot, inflammation and cancer. Conclusion: From the results, it can be concluded that the G. ajarekarii has an aptitude to act against coagulation, inflammation and cancer cells.

Oenothera biennis L. (OB), also commonly known as evening primrose, belongs to the Onagraceae family and has the best studied biological activity of all the members in the family. In therapy, the most frequently used type of extracts are from the aerial part, which are the fatty oils obtained from the seeds and have a wide range of medicinal properties. The aim of this study was to evaluate the phytochemical composition and biological activity of OB hydroalcoholic extract and to provide directions for the antimicrobial effect, antiproliferative and pro-apoptotic potential against A375 melanoma cell line, and anti-angiogenic and anti-inflammatory capacity. The main polyphenols and flavonoids identified were gallic acid, caffeic acid, epicatechin, coumaric acid, ferulic acid, rutin and rosmarinic acid. The total phenolic content was 631.496 µgGAE/mL of extract and the antioxidant activity was 7258.67 μmolTrolox/g of extract. The tested extract had a mild bacteriostatic effect on the tested bacterial strains. It was bactericidal only against Candida spp. and S. aureus. In the set of experimental conditions, the OB extract only manifested significant antiproliferative and pro-apoptotic activity against the A375 human melanoma cell line at the highest tested concentration, namely 60 μg/mL. The migration potential of A375 cells was hampered by the OB extract in a concentration-dependent manner. Furthermore, at the highest tested concentration, the OB extract altered the mitochondrial function in vitro, while reducing the angiogenic reaction, hindering compact tumor formation in the chorioallantoic membrane assay. Moreover, the OB extract elicited an anti-inflammatory effect on the experimental animal model of ear inflammation.

Objective: The aim of this study was to evaluate anti-inflammatory activity and cytotoxicity in vitro of hydroalcoholic extract of Bidens andicola.Methods: B. andicola hydroalcoholic extract was obtained from aerial parts of B. andicola, following a standardized methodology. Briefly, aerial parts of B. andicola were extracted with ethanol 70% v/v and defatted with n-hexane, hydroalcoholic fraction was concentrated under controlled conditions in a rotary evaporator, and finally the residue was freeze-drying to obtain the hydroalcoholic extract of B. andicola. Anti-inflammatory activity and cytotoxicity assays were carried out using in vitro isolated neutrophils model using stable water-soluble tetrazolium salts.Results and Conclusions: The in vitro anti-inflammatory assay on isolated neutrophils demonstrated that the hydroalcoholic extract showed antiinflammatoryactivity compared to aspirin, with inflammatory inhibition percent values of 80.138±0.729 to hydroalcoholic extract of B. andicola and 82.117±0.762 to aspirin, each tested in five replicates at the concentration of 200 ppm of hydroalcoholic extract or reference.

The phytochemical composition of Lactuca sativa L. var. crispa, commonly known as crisphead lettuce, from Morocco has been re-investigated for the presence of oxyprenylated cinnamic acid and coumarin derivatives. In the mean time also the anti-oxidant and anti-inflammatory effects of extracts from leaves obtained by the use of a Soxhlet apparatus have been assessed. The extracts had a high content of polyphenols with remarkable differences ranging from 0.19 to 0.38 mg gallic acid eq/mg, while the content of flavonoids ranged from 3.75 to 9.64 mg catechin eq/g. The hydroalcoholic extract with a polarity index of 5.8 displayed the best anti-oxidant, radical scavenging, and anti-inflammatory activities. The same extracts were screened by reverse-phase HPLC-UV/V analysis for the presence of biologically active oxyprenylated phenylpropanoids. Only 4'-geranyloxyferulic acid was recorded in detectable amounts. Results described herein enforce the nutraceutical value and depict a potential chemopreventive dietary feeding role for crisphead lettuce.
