Abstract Origanum vulgare has been of great interest in academia and pharma industry due to its antioxidant, antifungal and antitumor properties. The present study aimed to find the anti-MRSA potential and in vivo toxicity assessments of O. vulgare. O. vulgare extract was used to monitor anti-MRSA activity in mice. Following MRSA established infection in mice (Mus musculus), treatment with O. vulgare was continued for 7 days. Autopsies were performed and re-isolation, gross lesion scoring and bacterial load in various organs were measured. Additionally, blood sample was analysed for hematological assays. Toxicity assessment of O. vulgare potential as medicine was done at 200 mg/kg and 400 mg/kg by evaluating liver and kidney functions. Bacterial load and gross lesion in lungs and heart were significantly low compared to positive control following O. vulgare treatment. Likewise, O. vulgare treated groups had hematological, neutrophil and TLC values similar to control groups. Increased AST, ALP and total bilirubin alongwith marked hepatocellular degeneration and distortion around the central vein, inflammatory cell infiltration, and cytoplasmic vacuolization of hepatic cells was observed at higher dose. It is concluded that crude extract of O. vulgare may contain beneficial secondary metabolites and in future may be explored for curing infectious diseases.

Shaqīqa-i-Muzmin (chronic migraine) is a type of migraine that is usually caused by cold humours (bārid akhlāṭ), specifically by phlegm (balgham) and to lesser extent by black bile (sauda). The aim of this study was to evaluate the effect of Nutul-i-Ḥār (hot irrigation) in the treatment of chronic migraine. Nutul (irrigation) therapy is widely and successfully used in diseases of head as described in Unānī system of medicine. This open, single-arm, exploratory clinical study was conducted in 30 patients of chronic migraine selected by convenient sampling method. One litre decoction prepared with 12 g each of Astragalus hamosus L. (Iklilul malik), Matricaria chamomilla L. (Babuna), Artemisia absinthium L. (Afsanteen), Origanum vulgare L. (Marznjosh) and Trigonella foenum graecum L. (Hulba) was poured over painful side of head for 45 minutes on every alternate day for a period of 30 days (15 sittings). The patients, thereafter, were followed untill 90th day of the study for various outcome measures comprising headache intensity, headache frequency, Migraine Disability Assessment Scale (MIDAS), and rescue medication. The reduction in headache frequency, MIDAS score and use of rescue medication was significant (p < 0.001) after the treatment. Reduction in headache intensity at 30th day, 60th day and 90th day was significant as compared with baseline values (p < 0.001) but not statistically significant at 90th day with respect to 60th day (p > 0.05). Statistical analysis was done using parametric (paired t-test) and non-parametric tests (Wilcoxan sign ranked test, Freidman with Dunn’s multiple comparison tests). Hot irrigation with medicated decoction was found effective in the treatment of chronic migraine.

Fermentation processes in the rumen of ruminants determine how much final metabolites and their derivatives will be formed, which are necessary for the full development of the organism, the level of productivity, and also affect the level of formation of endogenous substances, namely, greenhouse gas emissions. These criteria lead us to the search for new feed products that improve the metabolic processes of the rumen and the digestive system as a whole, so phyto-substances can serve as an alternative. The article presents the results of in vitro study of the influence of Salviae folia, Scutellaria baicalensis, Oríganum vulgáre on formation of methane, synthesis of volatile fatty acids and nitrogen, as the main indicator parameters of the enzymatic activity of the rumen of ruminants. It was found that when using phyto- substances: Salviae folia and Scutellaria baicalensis, more acetic and propionic acid was formed, Oríganum vulgare in various dosages shifted towards propionic and valeric acid. Formation of a larger amount of microbial protein (P≤0.05) with use of Salviae folia, Scutellaria baicalensis, Oríganum vulgáre in various dosages was established. Methane production decreased with use of Oríganum vulgáre.

The goal of the study was to establish a procedure for improving the efficiency of supercritical carbon dioxide (scCO2) extraction of Origanum vulgare L. spp. hirtum (Greek oregano) and enhancing the quality of obtained extracts. Microwave and enzymatic pretreatments of the plant material were applied prior to the scCO2 extraction. It was determined that the microwave pretreatment with irradiation power 360 W during 2 min accelerated the extraction of lipophilic compounds and provided a twofold higher extraction yield compared to the control. Moreover, this pretreatment also led to an increase in oxygenated monoterpenes content and the most dominant component carvacrol, as well as the extracts’ antioxidant activity. The enzymatic pretreatment caused a significant increase in the extraction yield and the attainment of the extract with the most potent antioxidant properties. Coupling the pretreatments with scCO2 extraction improves the process of obtaining high value lipophilic products of oregano in terms of utilization of the plant material, acceleration of the extraction with the possibility to adjust its selectivity and quality of extracts, and enhancement of biological activity.

Oregano (Origanum vulgare L.) is a well-known traditional medicine and a cooking spice. Recent practice has also applied the essential oil from oregano (OEO) in poultry due to its great potential for an antibiotic alternative. Our objective was to evaluate the potential effects of OEO (with carvacrol and thymol as the main active ingredient) on preventing necrotic enteritis (NE) caused by Clostridium perfringens (Cp) in chickens. In the feeding trial, a total of 450 one-day-old commercial Arbor Acres broilers were randomly assigned in 5 experimental groups during a 26-day production period (d19 to d 26 was the Cp challenge stage), and each group consisted of 6 replicate pens (15 birds each pen). All treatments were: basal diet (control group); basal diet and Cp challenge (model group); Cp challenge and 10 mg/kg enramycin (positive control group); Cp challenge and 200 mg/kg OEO product (OEO low dosage group, OEOL); Cp challenge and 300 mg/kg OEO product (OEO high dosage group, OEOH). OEO feed supplement at both dosages had significant effects on increasing the body weight gain (BWG) and reversing the dropped feed intake (FI) induced by Cp challenge. Histopathological changes in the ileums of broiler chickens with NE induced by Cp were alleviated by OEO, which was mutually confirmed by the intestinal lesion scores. Dosage did not influence the protective effect of OEO on intestinal lesion scores. Furthermore, OEO was found to have limited effects on tight junction-related gene expressions (Occludin and ZO-1). The broilers of the OEOL and OEOH groups significantly decreased the expression of TNF-α mRNA in the ileum and only the OEOH group was found to inhibit the IFN-γ expression of IFN- induced by Cp challenge. Finally, despite the fact that in vitro antibacterial effects by OEO were observed, considering its high minimum inhibitory concentration (MIC) value, we inferred that the protective effects by OEO against Cp challenge were not attributable to its direct antibacterial effects. We proposed OEO as a promising substitute for antibiotics against NE induced by Cp during poultry production.

Since the registry of common oregano (Origanum vulgare L.) cultivars does not involve regionalization, a comprehensive study of cultivars bred by different institutions in the intended cultivation region is valuable and relevant. The objective of the research was to assess the possibility of using various indices of ecological adaptability originally developed for grain crops for their use in the most adapted genotypes’ selection (breeding samples and cultivars) of Origanum vulgare L. to the temperate climate of the Crimean Peninsula. The research was carried out in the piedmont zone of Crimea from 2016 to 2019. The study material consisted of breeding samples No. 10 and No. 82 from the collection of FSBSI “Research Institute of Agriculture of Crimea”, as well as cultivars Zima, Raduga, and Slavnitsa selected by the All-Russian Scientific Research Institute of Medicinal and Aromatic Plants (ARSRIMAP). Genotype had the greatest influence on yield of fresh oregano material (43%) with the influence of the weather conditions of the year being 2%. On the contrary, meteorological conditions had a much greater effect on the essential oil accumulation and its areal yield, which were 30% and 25%, respectively. In terms of the coefficient of ecological variation of fresh yield, sample No. 82 and Slavnitsa cultivar were the best (11.47–16.7%). The local genotypes No. 10 and No. 82 varied less by the essential oil content and its yield. The genotype effect value was greater than 0 in the Raduga cultivar and local genotype No. 82 for the yield, but only in No. 82 genotype for the other two characteristics. Cultivars Zima and Raduga were classified as intensive (bi > 1) by the environmental flexibility of fresh yield, while local genotype No. 82 and Slavnitsa cultivar formed the group of intensive ones by essential oil content and essential oil yield. Local genotypes No. 10 and No. 82 were better than the introduced cultivars in terms of essential oil content homeostability and essential oil yield (Hom = 1.91–2.18). Thus, local genotypes proved to be more adapted to the region’s conditions in terms of essential oil accumulation. However, they were inferior to the registered cultivars of ARSRIMAP breeding in terms of fresh yield.

El objetivo del estudio fue evaluar el efecto de la adición de aceites esenciales de orégano (AEO) en la dieta alimenticia sobre el comportamiento de la morfometría duodenal de pollos de engorde Cobb 500. Se utilizaron 400 pollos distribuidos en cinco tratamientos: suplementación con AEO (100, 200, 300 ppm) y dos tratamientos control (orégano comercial [250 ppm] y antibiótico promotor de crecimiento [300 ppm]), durante seis semanas, alimentados a voluntad con dietas formuladas semanalmente. El estudio morfométrico se realizó con ayuda de una cámara acoplada al microscópico mediante el software AmScope 3.0, objetivo de 4x, y campo de visión de 2000 µm2 de área. La variable ancho proximal de las vellosidades fue significativamente mayor (183 µm) en el tratamiento AEO 300 ppm a los 21 días de edad y la variable profundidad proximal fue significativamente mayor (256 µm) en el tratamiento AEO 300 ppm a los 42 días (p<0.05). Se concluye que la adición de AEO en dosis de 300 ppm afectan positivamente la morfometría duodenal de pollos Cobb 500.
