Effects of acclimation temperature on cortisol and oxygen consumption in Atlantic salmon (Salmo salar) post-smolt exposed to acute stress

The selected temperature for Atlantic salmon and rainbow trout, as determined in a horizontal gradient, increases with acclimation temperature over the acclimation range 5–20 C for salmon and 10–20 C for trout. The final preferendum for salmon is about 17 C. The results for rainbow trout suggest that the type of gradient used, i.e. vertical or horizontal, has a marked influence on the experimentally determined relation between acclimation temperature and selected temperature.

Weighted mean hourly rates of oxygen consumption in embryonated ova of Atlantic salmon, Salmo salar L., during embryogenesis, were reduced significantly by levels of dissolved oxygen below air saturation and by a temperature of 5 °C, relative to those for ova incubated at 10 °C. Total oxygen consumption during embryogenesis also was reduced significantly by the lower levels of dissolved oxygen, but not by temperature. The decrease in the pace of embryogenesis in the lots of ova at 5 °C extended the developmental time so that the lower rate of oxygen uptake was offset. Thus, within each level of dissolved oxygen there was no appreciable difference in the products of time units and units of oxygen uptake. At 5 °C, 100% air saturation, mean hourly uptake was 0.0141 mg O2/ovum, and total uptake was 28.153 mg O2/ovum. At 10 °C, 100% air saturation, these values were 0.0270 mg O2/ovum, and 27.974 mg O2/ovum, respectively. Values for ova incubated at 50 and 30% air saturation were correspondingly lower.

In this first study examining the thermal tolerance of adult Arctic char (Salvelinus alpinus) acclimated to seawater, we measured their critical thermal maximum (CTMax) and several cardiorespiratory parameters (oxygen consumption (MO2), heart rate (fH), stroke volume (SV), cardiac output (Q), ventilatory frequency (VF), opercular pressure (PO), and ventilatory effort (VE)) when exposed to a temperature increase of 2 °C·h−1. Further, we directly compared these results with those obtained for the eurythermal Atlantic salmon (Salmo salar) under identical conditions. There was no significant difference in cardiorespiratory values between the two species at their acclimation temperature (9.5–10 °C). In contrast, the slope of the MO2–temperature relationship was lower (by 27%) in the char as compared with that in the salmon, and the char had significantly lower values for maximum fH (by 13%), maximum MO2 (by 35%), absolute metabolic scope (by 39%), and CTMax (approximately 23 versus 26.5 °C, respectively). Although not a focus of the study, preliminary data suggest that interspecific differences in mitochondrial respiration (oxidative phosphorylation), and its temperature sensitivity, may partially explain the difference in thermal tolerance between the two species. These results provide considerable insights into why Atlantic salmon are displacing Arctic char in the current era of accelerated climate change.

Spontaneous activity, oxygen consumption, and excretion of ammonia by juvenile Atlantic salmon (Salmo salar) were measured over 17 wk as the appearance of the fish changed from early smolt to smolt and then postsmolt. A substantial change in routine oxygen consumption from 66 to 148 mL∙kg−1∙h−1 was primarily correlated with the increase in activity. An initial increase in ammonia excretion from about 5 to about 14 mL∙kg−1∙h−1 was largely correlated with the decline in condition factor from 0.99 to 0.79 suggesting that increased metabolic needs caused by the increased activity were not being met by the amount of food ingested. This is supported by the secondary increase in condition factor (C.F. = 0.93) and decrease in ammonia excretion (to about 7 mg∙kg−1∙h−1) that occurred after the meal size was increased.

We assessed the metabolic response of juvenile Atlantic salmon (Salmo salar; JAS) originating from two rivers with different natural thermal regimes to different acclimation temperature (15 or 20 °C) and diel temperature fluctuation (constant: ±0.5 °C; fluctuating: ±2.5 °C). Diel temperature fluctuation (15 ± 2.5 °C) near the thermal optimum (16 °C) for the species did not influence standard metabolic rate (SMR) compared with JAS acclimated to a constant temperature of 15 °C. Diel temperature fluctuation at 20 ± 2.5 °C increased SMR of JAS from the warmer river by 33.7% compared with the same fish acclimated to a constant temperature of 20 °C. SMR of JAS from the cooler river held at fluctuating conditions had SMR that were 8% lower than SMR at constant conditions. The results suggest that the mean temperature to which JAS is exposed may affect their responses to diel temperature fluctuation and that this response may vary between populations originating from rivers with different natural thermal regimes. Results were used to develop the first empirical SMR model for JAS subjected to diel temperature fluctuation using fish mass (3–36 g wet) and temperature (12.5–22.5 °C) as explanatory variables.
