Anaerobic digestion of wood vinegar wastewater using domesticated sludge: focusing on the relationship between organic degradation and microbial communities (archaea, bacteria, and fungi)

Olive oil and pig productions are important industries in Portugal that generate large volumes of wastewater with high organic load and toxicity, raising environmental concerns. The principal objective of this study is to energetically valorize these organic effluents—piggery effluent and olive mill wastewater—through the anaerobic digestion to the biogas/methane production, by means of the effluent complementarity concept. Several mixtures of piggery effluent were tested, with an increasing percentage of olive mill wastewater. The best performance was obtained for samples of piggery effluent alone and in admixture with 30% of OMW, which provided the same volume of biogas (0.8 L, 70% CH4), 63/75% COD removal, and 434/489 L CH4/kg SVin, respectively. The validation of the process was assessed by molecular evaluation through Next Generation Sequencing (NGS) of the 16S rRNA gene. The structure of the microbial communities for both samples, throughout the anaerobic process, was characterized by the predominance of bacterial populations belonging to the phylum Firmicutes, mainly Clostridiales, with Bacteroidetes being the subdominant populations. Archaea populations belonging to the genus Methanosarcina became predominant throughout anaerobic digestion, confirming the formation of methane mainly from acetate, in line with the greatest removal of volatile fatty acids (VFAs) in these samples.

Black soldier fly, Hermetia illucens (L.) (Diptera: Stratiomyidae) can be used for recycling a wide variety of organic wastes. However, these wastes are also suitable for colonisation by pest fly species, including the blow fly Lucilia sericata (Meigen) (Diptera: Calliphoridae), and by pathogenic microorganisms. We tested effects of the presence of black soldier fly larvae (BSFL) on recruitment and retention of blow fly larvae, as well as on composition and dynamics of microbial communities in finfish-based substrates. Substrates inhabited by BSFL contained approximately one-tenth of the L. sericata pupae found in the control substrate even when BSFL were removed prior to the introduction of gravid L. sericata females. Furthermore, almost three-quarters of L. sericata released into substrates previously colonised by BSFL emigrated within 24 h, while no such emigration was detected for the control substrates. Unlike previous studies, bacterial community was heavily dominated by Gram-positive species. Overall bacterial abundance on BSFL substrates declined approximately two-fold compared to the control substrate, while fungal abundance increased. However, surviving microbial communities were still robust and diverse, and continued to be dominated by Gram-positive bacteria. On the other hand, Gram-negative Shigella sonnei, which is a common cause of diarrhoea in humans, was more than 200 times less abundant in BSFL substrates compared to the control throughout the experiment. Repellent and antibiotic properties observed in this study are advantageous for using BSFL in remediation of finfish wastes. However, additional treatment of remaining residue may still be necessary to eliminate all biological contamination.

Biogenic amines (BAs) and nitrites are both considered harmful compounds for customer health, and are closely correlated with the microorganisms in fermented mustard (FM). In this study, BAs and nitrite contents in fifteen FM samples from different brands were analyzed. The concentrations of cadaverine in one sample and of histamine in one sample were above the toxic level. Moreover, five FM samples contained a high level of nitrite, exceeding the maximum residue limit (20 mg/kg) suggested by the National Food Safety Standard. Then, this study investigated bacterial and fungal communities by high-throughput sequencing analysis. Firmicutes and Basidiomycota were identified as the major bacteria and fungi phylum, respectively. The correlations among microorganisms, BAs and nitrite were analyzed. Typtamine showed a positive correlation with Lactobacillus and Pseudomonas. Cadaverine and nitrite is positively correlated with Leuconostoc. Furthermore, thirteen strains were selected from the samples to evaluate the accumulation and degradation properties of their BAs and nitrite. The results indicated that the Lactobacillus isolates, including L. plantarum GZ-2 and L. brevis SC-2, can significantly reduce BAs and nitrite in FM model experiments. This study not only assessed the contents of BAs and nitrite in FM samples, but also provided potential starter cultures for BAs and nitrite control in the FM products industry.

Abstract The oral microbiota is the direct precursor of dental caries and periodontitis, which are the most common microbial-induced diseases worldwide. The distinct microenvironment at the oral barrier breeds unique microbial communities, which are regulated by host factors (inflammation or dietary sugars)1. Increasing evidence indicates that dysbiosis of oral microbial communities is associated with many human diseases2-5. Our studies demonstrated that human disease could induce different alterations in salivary protein glycopatterns6. However, the relationship between salivary protein glycopatterns and oral microbial communities is unknown. Here, we report that altered salivary protein glycopatterns, namely, fucosylated or sialylated structures induced by gastric cancer (GC) or type 2 diabetes mellitus (T2DM), respectively, are also drivers of dysbiosis of oral microbial communities and ultimately dental caries and periodontitis. The fucosylated neoglycoproteins and sialic acid (SA) α2-3 galactose (Gal) structure can inhibit the growth or/and adhesion of Aggregatibacter segnis and Candida albicans from the oral cavity of patients with GC and T2DM, respectively. These findings provide a novel theory that dynamic communities of oral microbiota are regulated naturally by host salivary protein glycopatterns, having important implications for developing new carbohydrate drugs for oral and body health.
