A two-fold integrated research study was conducted; firstly, to understand the effects of copper (Cu) and zinc (Zn) on the growth and oxidative stress in Nile tilapia, Oreochromis niloticus; secondly, to study the beneficial effects of the duckweed Lemna minor L. as a heavy metal remover in wastewater. Experiments were conducted in mesocosms with and without duckweed. Tilapia fingerlings were exposed to Cu (0.004 and 0.02 mg L−1) and Zn (0.5 and 1.5 mg L−1) and fish fed for four weeks. We evaluated the fish growth performance, the hepatic DNA structure using comet assay, the expression of antioxidative genes (superoxide dismutase, SOD; catalase, CAT; glutathione peroxidase, GPx and glutathione-S-transferase, GST) and GPx and GST enzymatic activity. The results showed that Zn exhibited more pronounced toxic effects than Cu. A low dose of Cu did not influence the growth whereas higher doses of Cu and Zn significantly reduced the growth rate of tilapia compared to the control, but the addition of duckweed prevented weight loss. Furthermore, in the presence of a high dose of Cu and Zn, DNA damage decreased, antioxidant gene expressions and enzymatic activities increased. In conclusion, the results suggest that duckweed and Nile tilapia can be suitable candidates in metal remediation wastewater assessment programs.

Reservoir Gajah Mungkur of Wonogiri is an artificially aquatic environment with susceptibility of heavy metal contamination, so through the process of bioaccumulation, the fish tissue can be contaminated. This research were to identify concentration of heavy metals copper (Cu) and protein in the tissue of tilapia (Oreochromis niloticus Linn.), bred in floating cage breeders at the reservoir Gajah Mungkur. It was also tend to observe the relationship between heavy metal concentrations of Cu with protein content of tilapia tissue. Determination of sampling points was applied by a purposive sampling method. The concentration of both heavy metal and the measured protein was done descriptively, by comparing data obtained with fresh water quality standards according to PPRI No. 82 years 2001 and Decree DG POM No. 03725/B/SK/VII/89. Relations were analyzed by both correlation. The result showed that the concentration of Cu in the waters of reservoir Gajah Mungkur in the scale of 0,0108 mg/L was still below the threshold of fresh water quality standar. The parameters of temperature, pH, and DO on the results of the research were still in the range of values recommended for aquaculture activities. Cu concentration within tilapia tissues samples at 2,746 mg/kg were above the threshold of metal contamination in foods, especially processed meat, while the average protein content of tilapia value amounted to 18,07%. The higher the Cu content within the tissue, the lower the protein content of fish meat (r = 0,780).

ABSTRAK Timbal merupakan logam berat yang bersifat toksik dan biasanya bersumber dari industri cat, baterai dan percetakan. Sungai Tenggang berdekatan dengan Lingkungan Industri Kecil (LIK) dan pemukiman sehingga meningkatkan limbah ke dalam perairan termasuk logam berat. Hal tersebut menjadi alasan mengapa penelitian mengenai kandungan logam berat timbal pada ikan nila (Oreochromis niloticus) di Sungai tenggang, Semarang, Jawa Tengah ini dilaksanakan. Penelitian ini bertujuan untuk mengetahui kandungan logam berat timbal pada air dan ikan nila, mengetahui nilai biokonsentrasi faktor ikan nila serta mengetahui batas konsumsi mingguan daging ikan nila. Penelitian ini dilaksanakan pada bulan April 2019. Metode yang digunakan adalah metode deskriptif dengan pendekatan secara kuantitatif serta sampling menggunakan metode purposive sampling. Hasil perhitungan konsentrasi logam berat Pb dalam air rata-rata sebesar 0,155 mg/l. Konsentrasi logam berat Pb dalam daging ikan nila rata-rata 2,35 mg/kg. Hasil perhitungan bioconcentration factor (BCF) berkisar antara 12,03 – 17,17.  Hasil perhitungan berat maksimal konsumsi ikan nila mingguan diperoleh rata-rata sebesar 0,63 kg/minggu. Berdasarkan hasil yang telah diperoleh dapat disimpulkan bahwa kadar konsentrasi logam berat timbal pada air dan ikan nila di Sungai Tenggang tergolong cukup tinggi serta sudah melebihi ambang batas baku mutu yang telah ditentukan. Angka BCF logam berat timbal pada ikan nila termasuk dalam kategori tingkat akumulatif rendah (BCF < 100). ABSTRACT Plumbum is heavy metal that is toxic and source from paint industry, battery and printing. Tenggang river near with small industrial environment (LIK) and settlement so that increasing the waste entering to river including heavy metal waste. This is the reason for this research do. This study aims to determine the heavy metal Pb of water and Nile tilapia meat, bioconcentration factor of nile and knowing the limit of weekly consumption of  nile tilapia meat (MTI). The study was conducted in April 2019. The method used was a descriptive method with the determination of sampling using purposive sampling. The result of heavy metal Pb concentration in water averaged by 0,155 mg/l. Concentration of heavy metal Pb in Nile tilapia (Oreochromis niloticus) meat averaged by 2,35 mg/kg. The calculation result of bioconcentration factor (BCF) is range between 12,03 – 17,17. The maximum limit result of weekly fish consumption is averaged by 0,63 kg/minggu. Based on the results obtained it can be concluded that concentration of heavy metal Pb in water and nile tilapia tenggang river quite high and has reached above the permisible limit. BCF number of heavy metal in nile tilapia fall into the category of low accumulative levels (BCF < 100).

This study aims to apply vaccination against bacterial disease. The purpose of vaccination is to trigger the immune respone both non-specific and specific of fish against bacteria Motile Aeromonas Septicaemia (MAS) caused by Aeromonas hydrophila. The vaccination for fish with different ages: 2-3 weeks and 5 weeks were done using immersion method. The fish were re-vaccinated (booster) after two weeks of the first vaccination. The survival rate was < 50 % for juveniles 2-3 weeks and > 50% for juveniles 5 weeks. Survival rate for juveniles 5 weeks was higher than juveniles of 2-3 weeks. This shows that organs of juveniles of 5 weeks were more complete than the 2-3 weeks juveniles. The age of fish is one of the important factors for successfully vaccination. Penelitian ini bertujuan untuk melakukan penerapan vaksinasi terhadap penyakit bakterial. Vaksinasi ditujukan untuk merangsang respon kekebalan non- spesifik dan spesifik pada tubuh ikan terhadap penyakit Motile Aeromonas Septicaemia (MAS) yang disebabkan oleh bakteri Aeromonas hydrophila. Vaksin diberikan pada benih ikan nila yang berbeda umur yaitu 2-3 minggu dan 5 minggu dengan menggunakan metode perendaman. Ikan divaksinasi ulang (booster) setelah 2 (dua) minggu dari vaksinasi yang pertama. Prosentase kelangsungan hidup < 50 % untuk benih umur 2-3 minggu dan > 50% untuk benih umur 5 minggu. Jumlah kematian benih umur 2-3 minggu lebih tinggi dari 5 minggu. Hal ini menunjukkan bahwa fungsi organ benih umur 5 minggu telah lebih lengkap dari pada benih umur 2-3 minggu. Umur ikan merupakan salah satu faktor penting penentu keberhasilan suatu kegiatan vaksinasi.
