Cacti fruits are known to possess antioxidant and antiproliferative activities among other health benefits. The following paper evaluated the antioxidant capacity and bioactivity of five clarified juices from different cacti fruits (Stenocereus spp., Opuntia spp. and M. geomettizans) on four cancer cell lines as well as one normal cell line. Their antioxidant compositions were measured by three different protocols. Their phenolic compositions were quantified through high performance liquid chromatography and the percentages of cell proliferation of fibroblasts as well as breast, prostate, colorectal, and liver cancer cell lines were evaluated though in vitro assays. The results were further processed by principal component analysis. The clarified juice from M. geomettizans fruit showed the highest concentration of total phenolic compounds and induced cell death in liver and colorectal cancer cells lines as well as fibroblasts. The clarified juice extracted from yellow Opuntia ficus-indica fruit displayed antioxidant activity as well as a selective cytotoxic effect on a liver cancer cell line with no toxic effect on fibroblasts. In conclusion, the work supplies evidence on the antioxidant and antiproliferative activities that cacti juices possess, presenting potential as cancer cell proliferation preventing agents.

Objective: In recent years, herbal plants have been got more attention due to their diverse presence of phytochemicals and its biological properties. Hepatocellular carcinoma is one of the major worldwide problems primarily caused by hepatitis B and C virus infection. End-stage liver cancer treatment options are limited, thus requiring expensive liver transplantation which is not available in many countries. Methods: In the present study, the Mucuna pruriens seed extract was analyzed for phytochemicals, antioxidant activity, total phenols, and total flavonoids content. The seed extract was further analyzed for its anticancer activity by culturing liver cancer cell line. The above protocols were done by standard methods. Results: The seed extract of M. pruriens revealed more number of phytochemicals in different organic solvents. 1,1- Diphenyl- 2- picrylhydrozyl scavenging activity of plant extract was more in ethanol extract (98.051±0.547) among all other solvents. The total phenols and flavonoids content in ethanol extract were 46.442±0.353 mg gallic acid equivalent/g and 2.254±0.647 mg - quercetin equivalent /g, respectively. IC50 value of 3-(4, 5-dimethythiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) analysis of Hep-2 cell line was 150 (μg/ml).Conclusion: The present study revealed about the phytochemical contents and antioxidant potential of M. pruriens seeds. Further, the MTT analysis proved that the seed extract was effective against cancer cells and also used to treat many diseases.

Abstract Background: The prognosis of hepatocellular carcinoma (HCC) is closely related to immunity and inflammation, but the value of using immune and inflammation-related genes as predicting the prognosis of HCC requires further research. Methods: The Hepatocellular Carcinomar mRNA data was downloaded in the TCGA and ICGC database. The R package "limma" was used to analyze the differential expression of genes (DEGs) irelated to immune and inflammatory .Univariate Cox analysis screen for immune and inflammation related genes with prognostic value, then construction and verification of the prognostic model in Hepatocellular Carcinomar. The correlation between risk score with tumor immune immersion and immune cell function was assessed through tumor microensure and immune response analysis. NCI-60 cell line to explore the relationship between prognostic gene expression and drug sensitivity. Results: We evaluated 8 immune and inflammatory-related genes to build a prognostic risk prediction model, riskscore is an independent risk factor affecting prognosis, closely related to histological grading and clinical staging. The immune of adCs, macrophages, Tfh cells, Treg cells and Th1 cells higher in the tumor microenvironment leads to poor prognosis of liver cancer. Using data from the NCI-60 cell line, DNASE1L3 high expression may increased resistance of liver cancer cells to bovine platinum, solafinil and bovine platinum. The expression of SLC7A11 can increase the sensitivity of liver cancer to arsenic trioxide (ATO). Simultaneously constructing models and tumor microenvironment and drug resistance may provide effective and safe strategies for HCC chemotherapy and immunotherapy. Conclusion:Our study screened eight immune and inflammation-related genes play an important role in HCC tumor immunity and can be used to predict the prognosis of HCC.
