Abstract Background The biomanufacturing of d-glucaric acid has attracted increasing interest because it is one of the top value-added chemicals produced from biomass. Saccharomyces cerevisiae is regarded as an excellent host for d-glucaric acid production. Results The opi1 gene was knocked out because of its negative regulation on myo-inositol synthesis, which is the limiting step of d-glucaric acid production by S. cerevisiae. We then constructed the biosynthesis pathway of d-glucaric acid in S. cerevisiae INVSc1 opi1Δ and obtained two engineered strains, LGA-1 and LGA-C, producing record-breaking titers of d-glucaric acid: 9.53 ± 0.46 g/L and 11.21 ± 0.63 g/L d-glucaric acid from 30 g/L glucose and 10.8 g/L myo-inositol in fed-batch fermentation mode, respectively. However, LGA-1 was preferable because of its genetic stability and its superior performance in practical applications. There have been no reports on d-glucaric acid production from lignocellulose. Therefore, the biorefinery processes, including separated hydrolysis and fermentation (SHF), simultaneous saccharification and fermentation (SSF) and consolidated bioprocessing (CBP) were investigated and compared. CBP using an artificial microbial consortium composed of Trichoderma reesei (T. reesei) Rut-C30 and S. cerevisiae LGA-1 was found to have relatively high d-glucaric acid titers and yields after 7 d of fermentation, 0.54 ± 0.12 g/L d-glucaric acid from 15 g/L Avicel and 0.45 ± 0.06 g/L d-glucaric acid from 15 g/L steam-exploded corn stover (SECS), respectively. In an attempt to design the microbial consortium for more efficient CBP, the team consisting of T. reesei Rut-C30 and S. cerevisiae LGA-1 was found to be the best, with excellent work distribution and collaboration. Conclusions Two engineered S. cerevisiae strains, LGA-1 and LGA-C, with high titers of d-glucaric acid were obtained. This indicated that S. cerevisiae INVSc1 is an excellent host for d-glucaric acid production. Lignocellulose is a preferable substrate over myo-inositol. SHF, SSF, and CBP were studied, and CBP using an artificial microbial consortium of T. reesei Rut-C30 and S. cerevisiae LGA-1 was found to be promising because of its relatively high titer and yield. T. reesei Rut-C30 and S. cerevisiae LGA-1were proven to be the best teammates for CBP. Further work should be done to improve the efficiency of this microbial consortium for d-glucaric acid production from lignocellulose.

The Canadian Government has introduced a work-sharing program in which lay offs are avoided by reducing the work week and using unemployment insurance funds to pay workers short-time compensation. Compared to the lay-off alternative, there appear to be economic benefits to work-sharing for both management and employees. Reaction to the scheme has been generally positive at the union local level and the firm level, but it has been negative at the national level of both labour and management. These divergent views can be explained mainly as a result of short-run versus long-run perspectives. Managers at the firm level see the immediate benefit of improved labour relations and the avoidance of the costs of hiring and training replacements for laid-off workers who do not respond when recalled. The national business leaders are more concerned with work incentive and efficiency aspects of work-sharing.

The article focuses on working time policies introduced in Belgium during the period 1975-1990. As a country with early mass-unemployment, the magnitude of the unfolding Labour market problems fostered a specific set of responsive strategies. The initial trajectory of Belgian working time policies was centered around cutting standard weekly working hours in order to enhance Labour market effects. In the course of a marked issue transformation, work sharing objectives were substituted by the notion of temporal flexibility which focused primarily on concerns for and changes in the economie performance of individual firms. The author outlines various structural features of the Belgian socio-economic system and argues that these profoundly affected the goals identified with working time policies as much as the actor constellations endorsing the respective measures.

Centrifugal compressors operating at varying rotational speeds, such as in helicopters or turbochargers, can experience forced response failure modes. The response of the compressors can be triggered by aerodynamic flow non-uniformities, such as with diffuser-impeller interaction or with inlet distortions. The work presented here addresses experimental investigations of forced response in centrifugal compressors with inlet distortions. This research is part of an ongoing effort to develop related experimental techniques and to provide data for validation of computational tools. In this work measurements of blade surface pressure and aerodynamic work distribution were addressed. A series of pressure sensors were designed and installed on rotating impeller blades and simultaneous measurements with blade-mounted strain gauges were performed under engine representative conditions. To the best knowledge of the authors, this is the first publication which presents comprehensive experimental unsteady pressure measurements during forced response for highspeed radial compressors. Experimental data were obtained for both resonance and off-resonance conditions with uniquely tailored inlet distortion. This paper covers aspects relating to the design of fast response pressure sensors and their installation on thin impeller blades. Additionally, sensor properties are outlined with a focus on calibration and measurement uncertainty estimations. The second part of this paper presents unsteady pressure results taken for a number of inlet distortion cases. It will be shown that the intended excitation order due to inlet flow distortion is of comparable magnitude to the second and third harmonics which are consistently observed in all measurements. Finally, an experimental method will be outlined that enables the measurement aerodynamic work on the blade surface during resonant crossing. This approach quantifies the energy exchange between the blade and the flow in terms of cyclic work along the blade surface. The phase angle between the unsteady pressure and the blade movement will be shown to determine the direction of energy transfer between the blade and the fluid.

Daily rainfall data for twenty years in arable farming areas are analysed with respect to four standards of drainage and for three lengths of schedule of spring work. Distribution and frequency in time of available work days are interpreted in terms of lateness of sowing and of barley yield. Formulae are established to calculate average yield loss in terms of drainage standard and work schedule, enabling estimates to be made of the effect of planned improvements.

In ad hoc networks, the communication is usually made through multiple hops by establishing an environment of cooperation and coordination among self-operated nodes. Such nodes typically operate with a set of finite and scarce energy, processing, bandwidth, and storage resources. Due to the cooperative environment in such networks, nodes may consume additional resources by giving relaying services to other nodes. This aspect in such networks coined the situation of noncooperative behavior by some or all the nodes. Moreover, nodes sometimes do not cooperate with others due to their social likeness or their mobility. Noncooperative or selfish nodes can last for a longer time by preserving their resources for their own operations. However, such nodes can degrade the network's overall performance in terms of lower data gathering and information exchange rates, unbalanced work distribution, and higher end-to-end delays. This work surveys the main roots for motivating nodes to adapt selfish behavior and the solutions for handling such nodes. Different schemes are introduced to handle selfish nodes in wireless ad hoc networks. Various types of routing techniques have been introduced to target different types of ad hoc networks having support for keeping misbehaving or selfish nodes. The major solutions for such scenarios can be trust-, punishment-, and stimulation-based mechanisms. Some key protocols are simulated and analyzed for getting their performance metrics to compare their effectiveness.
