Observed and expected combined effects of clinical mastitis and low body condition on pregnancy loss in dairy cows

AbstractDairy cows in confinement and pasture-based feeding systems were compared across 4 spring-calving and 3 fall-calving replicates for differences in reproduction, mastitis, and body condition scores. Each feeding system and replicate included both Jersey and Holstein cows. Cows in confinement were fed a total mixed ration with maize silage as the base forage along with a variety of concentrates. Cows on pasture were routinely supplemented with concentrates and were provided baled grass and/or legume silage when pastures were limiting. Breeding periods were for 75 days in spring or fall. Reproductive performance did not differ significantly due to feeding system but Jerseys had higher conception rates (59.6 vs. 49.5%) and overall pregnancy rates (78.0 vs. 57.9%) than Holsteins. Cows in confinement had significantly higher rates of clinical mastitis and culling for mastitis than cows on pasture. Within both feeding systems, Jerseys had less mastitis than Holsteins. Average body condition scores were higher for confinement cows than pasture cows except for one replicate. Jerseys maintained higher average body condition scores than Holsteins in all but one replicate. In summary, pastured cows produced less milk and had less mastitis but lower body condition scores than confinement cows. Holsteins produced more milk but were less likely to rebreed, had more mastitis, higher culling rates, and maintained lower body condition scores than Jerseys.

The aim of this study was to define the time-related period of intramammary infections and its relation to risk factors for intramammary infections and clinical mastitis at cow and quarter levels. In total, 269 German Holstein Frisian dairy cows on three farms in Northern and Eastern Germany were included in this study. Quarter milk samples were collected at dry-off, 3 ± 1 days after calving and 17 ± 3 days after calving, for cytomicrobiological examination. Risk factors at quarter- and cow-level associated with intramammary infections and clinical mastitis were recorded during the trial period. Data were analyzed using logistic regression procedures and odds ratios were calculated. Calving for the second time increased the odds of clinical mastitis during the first 100 days of lactation compared to cows calving for the third time or more. A high milk yield after calving was a risk factor for new infections, with environmental pathogens 17 ± 3 days postpartum. A body condition score after calving less than 3.5 was associated with a decreased risk of having an intra-mammary infection (IMI) with non-aureus staphylococci and coryneforms 3 ± 1 days postpartum and consistent body condition between dry-off and early lactation decreased the risk of intramammary infections after calving. The absence of a ring of hyperkeratosis at the teat apex shown at dry-off was associated with a lower risk of intramammary infections with environmental pathogens 17 ± 3 days postpartum. This study shows the important influence of the dry period and early lactation on intramammary infections and clinical mastitis postpartum in dairy cows. Udder quarters may have eliminated pathogens during the dry period in 43.6% of cases in this study. Additionally, new infections occurred during early lactation, so 5.1% more quarters were infected 17 ± 3 days compared to 3 ± 1 days postpartum. New infections can be traced to non-aureus staphylococci and Staphylococcus aureus from dry-off up until 3 ± 1 days postpartum, and to non-aureus staphylococci, Staphylococcus aureus and Streptococcus uberis, after calving. In total, 88.7% of the infected quarters showed new infections with another pathogen species 3 ± 1 days postpartum than at dry-off, and 89.2% of the quarters 17 ± 3 days postpartum than 3 ± 1 days postpartum. In conclusion, the early lactation has just as important an influence on intramammary infections postpartum in dairy cows as the dry period. There is the possibility that udder quarters eliminate pathogens during the early lactation, especially during the dry period. However, there is also the danger that new infections manifest, with a large proportion of new infections occurring after calving. Thus, additional control strategies are of great importance to prevent new infections occurring during early lactation as well as during the dry period to reduce negative effects on milk yield and culling hazards in dairy cows by minimizing the associated risk factors.

Abstract Pregnancy loss in high producing dairy cows is significant and negatively affects profitability of dairy farms. It is not clear the extent of pregnancy loss prior to a subsequent estrus or first pregnancy diagnosis. It is clear that percent of cows losing pregnancies are greatest following 1st pregnancy diagnoses then decrease in a time-related fashion throughout gestation. Non-disease related factors that are associated with pregnancy losses in dairy cows include parity, timing of AI relative to ovulation, circulating concentrations of progesterone during growth of the ovulatory follicle(s), double ovulations, unilateral twins, service sire, extent of change in body condition during early lactation, and serum levels of pregnancy associated glycoproteins at 24 or 28 d post-AI. Metabolic changes related to high milk production resulted in a greater chance for double ovulations. A growing body of literature indicates that cows with unilateral double ovulations and unilateral twins have a greater chance for pregnancy loss during the 1st 90 d of pregnancy compared to cows with single ovulations or bilateral twins. It also appears there is a preferential timing of loss in cows with double vs. single ovulations. Levels of circulating concentrations of progesterone during follicular wave development affect follicular development of cows. In a study that controlled progesterone during follicular development, low levels of progesterone caused a greater percentage of cows to have double ovulations and increased chances for pregnancy loss after 35 d post-AI. Lastly, it appears that cows that lose body condition during the 1st 30 DIM have a greater chance of pregnancy loss between 35 and 69 d post-AI compared with cows that maintained or gained body condition during that period. These data lead towards a greater understanding of the potential causes of pregnancy loss in dairy cows and can be instrumental in the development of reproductive technologies to reduce these losses.

Individual and combined effects of several isomers of retinoic acid (RA) and 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) on interferon-gamma (IFN-gamma) secretion by blood mononuclear leukocytes (MNL) from nulliparous and postparturient Holstein cattle were evaluated in vitro. In the first experiment, effects on incubation period (24 to 72 hours) and time of supplementation (0 to 32 hours) with all-trans, 9-cis, 13-cis-, and 9,13-dicis-RAs (0 to 100 nM) on IFN-gamma secretion by pokeweed mitogen (PWM)-stimulated (0 and 10 mug/ml) MNL from nulliparous cattle were evaluated. In the second experiment, MNL from postparturient cows (bled at 0, 2, 4, and 16 days postpartum) were stimulated with PWM (0 and 10 mug/ml) in the presence of RA isomers (9-cis- or 9,13-dicis-RA; 0 to 100 nM), 1,25-(OH)2D3 (0 to 100 nM), or with combinations of these metabolites. The results show that individual isomers of RA had no effect on IFN-gamma secretion by PWM-stimulated MNL from nulliparous or postparturient cows. Furthermore 1,25-dihydroxyvitamin D3 inhibited IFN-gamma secretion by MNL from nulliparous and postparturient dairy cows; however, the degree of inhibition was greater when 9-cis- and 9,13-dicis-RA were also present in the cultures. Finally mononuclear leukocytes from postparturient dairy cows produced substantially less IFN-gamma than did MNL from nulliparous cattle. It is concluded that retinoic acids individually did not affect the capacity of leukocytes from dairy cattle to secrete IFN-gamma. This result is in marked contrast to studies in monogastric species indicating that RAs inhibit IFN-gamma secretion by peripheral blood T cells. Inhibition of IFN-gamma secretion by 1,25-(OH)2D3 was potentiated by 9-cis- and 9,13-di-cis-retinoics acids, suggesting that an excess of dietary vitamins A and D may compromise further the naturally immunosuppressed postparturient dairy cow. Additional research is necessary to determine if the combined effects of these metabolites on IFN-gamma secretion represent an increased susceptibility of the dairy cow to infectious diseases during the periparturient period. Lower secretion of IFN-gamma by MNL from postpartutient dairy cows, relative to nulliparous cattle, suggests that recently-calved cows are naturally immunosuppressed.

 Background: Bovine mastitis causes major economic losses for milk producers by reducing the quantity and the quality of the milk or even leading to the complete loss of the mammary gland secretory capacity. During the transition period, dairy cows are susceptible to infectious diseases; therefore, markers that allow early identification of cows in higher risk of developing diseases are especially useful at this time. Therefore, the aim of this study was to evaluate serum markers in the pre and postpartum of multiparous dairy cows with clinical mastitis and with health condition in the postpartum period in a semi-extensive management system.Materials, Methods & Results: Thirty-Six Holstein cows were monitored daily during milking until 59 days postpartum and were categorized according to the pre-milking strip cup test into clinical mastitis (mastitis group (MG)) and absence of symptoms (control group (CG)) that were negative to the test, representing the health cows. All cows were reared as one group and maintained in a semi-extensive pasture-based system. Blood samples were collected weekly after morning milking via venipuncture of the coccinea vein into tubes without anticoagulant and grouped for prepartum (-21 to 0 days from calving), early postpartum (0 to 30 days from calving), and late postpartum (30 to 59 days from calving) periods. Milk production was recorded daily. The serum markers albumin, aspartate aminotransferase (AST), phosphorus, gamma-glutamyltransferase (GGT) and non-esterified fatty acids (NEFA) were measured. Statistical analyses were performed using SAS®. The cases of clinical mastitis occurred on average at 37.2 ± 4.9 days postpartum. Health cows (CG) had higher milk production compared to the mastitis group (MG) only in the late postpartum period (P < 0.05). There was no difference among groups for albumin and NEFA concentrations in all periods evaluated (P > 0.05). In the early postpartum period the AST activity was higher in CG than in MG (P = 0.02). The GGT enzyme tended to be more concentrated in the CG than in the MG during the early (P = 0.06) and late (P = 0.08) postpartum periods. Late postpartum phosphorus concentration was lower for MG than CG (P = 0.04). In the prepartum and early postpartum periods, there was no difference among groups for phosphorus concentration (P > 0.05).Discussion: A decrease in milk production in MG compared to CG observed in late postpartum period was due to the inci­dence of mastitis observed around 37 days postpartum. Cows that presented clinical mastitis in the postpartum period did not differ in the blood concentration of NEFA in the prepartum period. In the late postpartum period higher concentration of phosphorus was observed in the CG than in MG, indicating that animals affected by mastitis may be in the weakest energy status. Regarding liver health, the concentration of AST was higher in the recent postpartum period for CG, in disagree­ment with previous studies that related AST to tissue injury caused by mastitis. The GGT enzyme tended to had higher concentrations in CG than MG during the whole postpartum period and may be related to increased hepatic metabolism due to higher production. There were no changes in albumin levels among healthy and mastitis cows, indicating that this marker can not be used to predict clinical mastitis. There were no metabolic alterations in the prepartum period related to the occurrence of postpartum mastitis in multiparous cows in a semi-extensive management system.Keywords: AST, dairy cows, NEFA.

Mastitis is one of the diseases with the highest incidence in dairy cows, causing huge economic losses to the dairy industry all over the world. The aim of the study was to characterize mastitic milk metabolome through untargeted nuclear magnetic resonance spectroscopy (1H-NMR). Taking advantage of the high reproducibility of 1H-NMR, we had the opportunity to provide quantitative information for all the metabolites identified. Fifty-four molecules were characterized, sorted mainly into the chemical groups, namely amino acids, peptides and analogues, carbohydrates and derivates, organic acids and derivates, nucleosides, nucleotides and analogues. Combined with serum metabolomic investigations, several pathways were addressed to explain the mechanisms of milk metabolome variation affected by clinical mastitis, such as tricarboxylic acid cycle (TCA cycle) and phenylalanine, tyrosine and tryptophan biosynthesis. These results provide a further understanding of milk metabolome altered by clinical mastitis, which can be used as a reference for the further milk metabolome investigations.
