AbstractThis study aims developing and evaluate a protocol of semen cryopreservation of the lane snapper Lutjanus synagris. Firstly, sperm motility rate, motility time, density and spermatocrit were appraised to characterize the sperm quality of the lane snapper. The effect of three extenders with distinct ionic compositions and pH values combined with seven concentrations of cryoprotector dimethylsulfoxide (0; 2.5; 5.0; 7.5; 10.0; 12.5 e 15.0%), five cooling rates (110, 90, 60, 45 e 30°C –min), nine equilibration time (1; 2,5; 5; 10; 15; 20; 25; 30 e 60 minutes) e five dilutions ratio (1:1; 1:3; 1:6; 1:10 e 1:20) on the sperm motility rate and motility time were analyzed. Fertilization test was accomplished to evaluate the viability of the cryopreserved sperm. The higher sperm motility rate and motility time (P<0.05) was achieved by combining extender with pH 8.2 with 10% concentration of dimethylsulfoxide and cooling rate 60°C –min, 1 minute of equilibration time and 1:3 (v/v) dilution ratio. The use of cryopreserved sperm presented fertilization rates >60% validating the present protocol for lane snapper. The cryoconserved sperm of lane snapper is a viable alternative, being possible to maintain appropriate sperm viability.

<p class="Pa7">Cryopreservation of Curimba semen (Prochilodus lineatus) is ecological and commercial importance. The objective of this study was to evaluate the effect of different concentrations (2, 5, 8 and 11%) of dimethyl sulfoxide (DMSO) diluted in Betsville Thawing Solution (BTS) on the quality of post-thaw semen Curimba. We analyzed the rate and period motility, sperm viability, membrane integrity and DNA, mitochondrial functionality, and fertilization and hatching rate. The plasma membrane and DNA integrity of a DMSO concentration of 11% obtained better results than the concentration of 5% (p &lt;0.05). However, treatment of 5% DMSO resulted in a longer latency and a higher fertilization rate and hatching, in other sperm quality equal to that of fresh semen. The results of this study indicate that 5% DMSO is ideal for cryopreservation of semen Curimba.</p>

Semen cryopreservation allows crucial management of animal genetic diversity. However, the freeze-thaw process causes biochemical and physical alterations, impairing sperm energy-dependent functions. Currently, many chemicals are added to the media to enhance frozen-thawed sperm quality during artificial insemination. The aims of this study was to determine the effects of Metformin (Metf) on fresh chicken sperm motility and ability to perform acrosome reaction, and to evaluate Metf’s effects on the functions of cryopreserved sperm. Chicken semen was diluted and incubated at 35°C in media supplemented with or without different doses of 5’-AMP-Activated Protein Kinase (AMPK) activator, Metf (0,5 to 5 mM). We then looked for the concentration improving the most sperm quality to use it in the cryopreservation media used for chicken sperm. Our results show that 1 mM Metf is the concentration giving the best results regarding sperm quality. AMPKα phosphorylation, viability, acrosome reaction ability (AR), and various motility parameters, were negatively affected by the freeze-thaw process, and that Metf partially restored them. Sperm quality improved (mean increased by 23% for motility, by 10% for viability) as well as AMPKα phosphorylation (mean increased by 30%). Moreover, fluorescent intensity levels of phospho-AMPK were also stronger with Metf than in the control. These results show that the presence of Metf in fresh semen has a positive impact on the quality of sperm and helps reducing the gradual decline in sperm motility caused by cryopreservation by partially restoring several essential sperm functions, and thus leads to a better overall quality of cryopreserved sperm.

In recent years, in Ukraine, as well as around the world, some local breeds have been on the verge of extinction, resulting in irretrievable loss of valuable genes, reduction of genetic diversity. In order to avoid these problems, it is necessary to work on the conservation of animal genetic resources. The creation of banks for long-term storage of biological material is one of the methods of preserving the gene pool of local, small and indigenous breeds of animals. Sperm quality is one of the main factors that determines the success of insemination of heifers and cows. Comprehensive analysis of sperm includes a large number of qualitative and quantitative indicators, the accuracy of which is influenced by a number of objective and subjective factors. Currently, microscopic analysis of sperm motility and survival is used in production conditions. Such approaches are easy to implement, but their results can be subjective. Research using a computer-based sperm fertility analysis system is becoming a priority today. With the help of CASA-Sperm Vision it is possible to carry out a morphological analysis already during the study of sperm motility. The purpose of our research was to analyze the quality of deep-frozen sperm of sires of Lebedinian breed and original brown cattle of Germany in order to attract such genetic material in the conservation of local Lebedinian breed cattle. Materials and methods. In the study used cryopreserved sperm doses of five sires of Lebedinian breed and three sires of the original brown cattle of Germany. Researches of qualitative, quantitative and dynamic characteristics of sperm of bulls were carried out at the laboratory of biotechnology of reproduction of Institute of Animal Breeding and Genetics nd. a. M.V.Zubets of NAAS and production laboratory "Ukrainian genetic company" using computer analysis of sperm motility Sperm Analysis System Version 12 IVOS. It was determined that the average dose of cryopreserved sperm of bulls of the Lebedinian breed was 0.25 ml, and of the sires of the original brown cattle of Germany -0.21 ml. It was found that the sires of the original brown cattle of Germany had the best indicators of total sperm motility by 21.7% compared to the sires of the Lebedinian breed (55.7%). They also noted a greater number of sperm with straight-forward movement of 16.5%. (40.2% of the Lebedinian breed sires). The average concentration of sperm in one milliliter was 10.5 times higher in animals of the Lebedinian breed (compared to the sires of the original brown cattle of Germany (331 million/ml). Comparing the indicators of the average speed of the sperm on the average trajectory (VAP), it should be noted that the minimum value was 85.3 microns/sec, the maximum value for this indicator – 141.7 microns/sec. The results of the VSL study showed that the average value of this indicator of sperm in the studied sperm doses of sires of the Lebedinian breed was 102 microns/sec, of the original brown cattle of Germany – 75. Obtained opposite to the direction coefficients of the correlation depending on the origin, between total sperm motility and average sperm speed for the average trajectory, the average speed of the sperm on the real trajectory, the average deviation of the sperm, the degree of straightness of the directed movement of sperm. Summary. 1. The complex of researches provided an objective analysis of qualitative and quantitative indicators of cryopreserved sperm production of the bulls of original brown cattle of Germany and the Lebedinian breed. It was found that the quality of the studied sperm doses met the requirements of "Instructions for Artificial Insemination of Cows and Heifers" (Order of the Ministry of Agrarian Policy of Ukraine dated August 1, 2001 № 230). 2. The comparative analysis of sperm production of the original brown cattle of Germany and the Lebedinian breed for its long-term storage was carried out. It is established that the sperm of sires of the original brown cattle of Germany exhibit higher dynamic characteristics of movement, while the sperm of sires of the Lebedinian breed were inferior to them in these indicators. 3. The possibility of insemination of females with cryopreserved sperm of the studied bulls whose sperm production has been stored for more than 30 years has been proved.

Aim. To evaluate the activity of cryopreserved boar ejaculated spermatozoa under different thawing protocols to optimize biotechnological approaches in pig reproduction based on the genetic material of the Institute of Animal Breeding and Genetics nd. a. M.V. Zubets of National Academy of Agrarian Science of Ukraine. Methods. Cryopreserved ejaculated boars' sperm was thawed in three different ways. Biotechnological, cryobiological and morphological methods were used to assess the viability of the sperm. Results. We noted the individual feature of semen quality of the studied boars, which affects its suitability for cryopreservation and quality indicators after thawing. It was found that in liquefied boar sperm (sperm activity on average is 86.7%) after its centrifugation and three-hour equilibration at + 4 °C sperm activity decreased by an average of 25.0%. In order to improve the quality of cryopreserved sperm, it was thawed under different conditions. The highest activity of spermatozoa in thawed sperm of boar No. 225 of Myrhorod pig was at the level of 25.0% at the thawing temperature + 70 °C, and the thermal resistance and heat resistance were 70.0% and 60.0%, respectively. Conclusions. During this study was noted that there are peculiarities of boar semen at breed and individual level, which influence its ability to tolerate cryopreservation.
