ABSTRACT The cattle tick, Rhipicephalus (Boophilus) microplus, is a major threat to the improvement of cattle production in tropical and subtropical countries worldwide. Bos indicus cattle are naturally more resistant to infestation with the cattle tick than are Bos taurus breeds, although considerable variation in resistance occurs within and between breeds. It is not known which genes contribute to the resistant phenotype, nor have immune parameters involved in resistance to R. microplus been fully described for the bovine host. This study was undertaken to determine whether selected cellular and antibody parameters of the peripheral circulation differed between tick-resistant Bos indicus and tick-susceptible Bos taurus cattle following a period of tick infestations. This study demonstrated significant differences between the two breeds with respect to the percentage of cellular subsets comprising the peripheral blood mononuclear cell population, cytokine expression by peripheral blood leukocytes, and levels of tick-specific immunoglobulin G1 (IgG1) antibodies measured in the peripheral circulation. In addition to these parameters, the Affymetrix bovine genome microarray was used to analyze gene expression by peripheral blood leukocytes of these animals. The results demonstrate that the Bos indicus cattle developed a stabilized T-cell-mediated response to tick infestation evidenced by their cellular profile and leukocyte cytokine spectrum. The Bos taurus cattle demonstrated cellular and gene expression profiles consistent with a sustained innate, inflammatory response to infestation, although high tick-specific IgG1 titers suggest that these animals have also developed a T-cell response to infestation.

The numbers of cattle ticks, Boophi1us microplus, were counted on cattle, with different Bos indicus (zebu) content in central and southern Queensland, from November 1977 to May 1982. Concurrently, measurements were made of the survival rates of parasitic stages of the tick and of the success of free-living stages in producing larval progeny. These measurements were related to the tick burdens on the cattle in order to test current understanding of the ecology of the tick.Tick numbers increased exponentially with decreasing Bos indicus content. Tick numbers built up very rapidly on the Bos taurus cattle and resulted in deaths early in the first season, so the herd was replaced with animals of 25% zebu content. The pure zebu animals carried an average of five ticks per animal per day in central Queensland and only one in southern Queensland. The 50% zebu herds carried 65 ticks per day in central Queensland and 1 l in southern Queensland, compared with 79 and 37 on the 25% zebu herds at the two locations. During their brief time in the experiment the B. taurus animals carried 465 and 302 ticks per day in central and southern Queensland respectively.The observed tick infestation levels were estimated to cause annual losses of liveweight gain of 1, 14 and 17 kg in zebu, 50% zebu and 25% zebu animals respectively in central Queensland. Comparative figures for southern Queensland were 0, 2 and 8 kg. Compensatory liveweight gain would have reduced these losses by up to 6 kg. These data relate to animals which were breeding most of the time and hence were less resistant than dry cattle. The results indicate that B. microplus has been reduced to a marginal problem on animals with 50% zebu genes in the region. It is therefore difficull to justify any control measures in southern areas, particularly when they could increase the risk of babesiosis. Owners of B. taurus cattle will continue to be at risk of major losses in productivity.Changes in the tick burden on the herd5 were explained mostly by changes in survival rates of parasitic ticks in central Queensland and by changes in availability of larvae on the pastures in southern Queensland.

The ability of cattle to resist tick infestations is partly genetically determined. In order to better define the nature of Bos taurus resistance to the cattle tick Boophilus microplus, skin gene expression was studied using a cattle skin derived cDNA microarray. Expression profiles were determined in skin biopsies sampled from three highly tick resistant animals (HR) and two animals with lower tick resistance (LR) at time 0, immediately before challenge, and again 24 h after challenge. The analysis of the resulting expression data addressed two biological questions: first, for any animal exposed to ticks, which genes are differentially expressed in the 24 h following challenge; and second, which genes are differentially expressed between animals of high and low resistance at 24 h after challenge? In total, 214 genes were found to be differentially expressed in response to larval challenge across all the animals. Seventy-two genes were upregulated and 76 were downregulated at 24 h after challenge. Genes with significantly altered gene expression levels following tick infestation were predominantly keratin genes or mitochondrial genes, as well as odorant binding protein (OBP) and Bos taurus major allergen BDA20. In addition, we identified 66 genes with differential expression between HR and LR animals at 24 h. Of these, genes representing the extracellular matrix and immunoglobulin gene expression pathways were overrepresented. Three differentially expressed genes, OBP, Bos taurus major allergen BDA20 and dendritic cell protein HFL-B5 were further analysed by quantitative reverse transcription PCR (qRT-PCR). The qRT-PCR assay results closely mirrored the expression profiles found in the microarray experiment.

O objetivo do trabalho foi analisar os métodos de controle do carrapato Boophilus microplus realizados em três fazendas representativas dos sistemas de produção de leite da Microrregião Fisiográfica Fluminense do Grande Rio, Rio de Janeiro, levando-se em consideração o manejo das fazendas, o grau de sangue Bos taurus e Bos indicus dos rebanhos, os fatores climáticos e a prevalência estacional do carrapato. Para efeito de avaliação, foi utilizada a contagem periódica de fêmeas ingurgitadas medindo entre 4,5 e 8mm, no antímero direito de 20% das vacas em lactação de cada fazenda, durante um ano. A diferença no manejo das pastagens, a composição genética dos rebanhos e as condições climáticas influenciaram a prevalência estacional de B. microplus. A maior lotação animal por hectare, o elevado "stand" vegetativo das pastagens e o maior grau de sangue B. taurus contribuíram para as maiores infestações de carrapatos nas fazendas. O controle de B. microplus realizado pelos proprietários teve importância secundária em relação as outras atitudes de manejo dos rebanhos. Ficou evidenciado o uso excessivo e ineficiente de produtos químicos para o controle de B. microplus nas fazendas. Para implantação de medidas de controle estratégico do B. Microplus, fazem-se necessários esforços para a transferência e adoção dos resultados de pesquisas disponíveis aos produtores rurais.

Four purebred Brahman and four beef Shorthorn cattle which had not previously been exposed to Boophilus microplus were infested four times with 20,000 B. microplus larvae. On first infestation, the yield of engorged female ticks on all animals was about 25% of the larvae applied. Aftcr three further infestations, the mean yield of engorged females on the Brahmans decreased to 7.5% whereas there was no decrease in the yield of ticks on the Shorthorns. On the Brahman cattle. development of larvae to engorged females took 1–2 days longer and the engorged females weighed less. However, there was no difference in the reproductive index of female ticks engorged on either Brahmans or Shorthorns. Increases in rectal temperatures occurred in all cattle 15–17 days after the first infestation and in some cattle on days 6–7 as well as on days 15–17 after reinfestation. It was concluded that resistance to B. microplus is an acquired phenomenon in Brahman cattle.

The effects of cattle tick infestations on liveweight and fertility of Droughtmaster (1/2 Bos indicus) cows and on calf weaning weights were determined over 3 years. Tick populations on the control (non-dipped) group of cattle fluctuated, with mean annual tick counts/side being 9.5, 8.9 and 13.6 for years 1, 2 and 3 respectively, while the treated group of cattle, which were dipped every 21 days, were free of ticks. Tick counts were not related to fertility or liveweight change in pregnant-lactating cows or to calf growtb or weaning weights. Treatment for ticks significantly (P<0.05) affected liveweight change in pregnant-lactating cows on only a few occasions, and annual liveweight changes were not significantly influenced by treatment. When lactating cow pregnancy rates were low (< 30% for control cows), treatment for ticks increased the pregnancy rate by about 100% in 2 of the 3 years, these differences being significant only in the last year. Calves in the treated group were born significantly earlier in the first year and had significantly lighter birth weights in the third year. Treated calves grew faster to weaning and had higher weaning weights (mean difference 17.9 kg) than control calves but differences were significant in the first and third years only.

The cryopreservation of in vitro-produced (IVP) bovine embryos is one of the most challenging areas of the assisted reproductive biotechnologies. The aim of the present study was to evaluate the global gene expression pattern of Bos indicus (Nellore) and Bos taurus (Simmental) IVP embryos after vitrification. Follicular aspiration was performed on Nellore (n = 14) and Simmental (n = 14) cows, and oocytes (n = 840 and 450; respectively) were submitted to in vitro maturation and in vitro fertilization. Presumptive zygotes were denuded and cultured in SOFaa with 0.5% BSA and 2.5% FCS during 7 days under standard culture conditions. Blastocysts (grade 1 and 2) were vitrified, warmed, and cultured for an additional 12 h under the same conditions. Nellore (n = 8) and Simmental (n = 8) IVP blastocysts considered viable after vitrification, with re-expanded blastocoel, were submitted to total RNA extraction (PicoPure, Arcturus, Applied Biosystems®, Foster Dity, CA, USA), DNAse I treatment (Qiagen®, Valencia, CA, USA), and amplification (RiboAmp, Applied Biosystems®). Fragmented cRNA were obtained through 3′IVT Express Kit (Affymetrix®, Santa Clara, CA, USA) to perform the hybridization using GeneChip Bovine Genome Array (Affymetrix®). Microarray data analysis was performed using the FlexArray 1.6.1.1 software. Genes with at least a 1.5-fold change and a P-value of less than 0.05 were considered differentially expressed. Of the 1278 genes differentially expressed between Bos taurus and Bos indicus vitrified embryos, 1108 were annotated, with 1193 genes up-regulated and 85 genes down-regulated in Bos taurus compared with Bos indicus IVP vitrified embryos. Differentially expressed genes were associated with the functional networks of cell cycle, cellular movement and DNA replication, recombination and repair; RNA post-transcriptional modifications; gene expression, protein synthesis; RNA damage and repair; cellular function and maintenance; and cell death and survival. The top 6 canonical pathways generated by Ingenuity Pathway Analysis® with the differentially expressed genes were ELF2 signalling, oxidative phosphorylation, tricarboxylic acid cycle, protein ubiquitination pathway, mTOR signalling, and IGF-1 signalling. In conclusion, Bos taurus IVP embryos seem to trigger different cellular response mechanisms to the vitrification stress in comparison with Bos indicus IVP embryos. Differential response is mainly represented by different expression profiles of genes regulating important canonical pathways involved in cellular response to stress that could be related with the higher post-cryopreservation survival capacity observed in Bos taurus embryos.Research was supported by FAPESP, CNPq, FAPERGS, and LNBio – National Laboratory of Biosciences/MCT.
