Detection of Mycobacterium avium subsp. paratuberculosis in intestinal and lymph node tissues of water buffaloes (Bubalus bubalis) by PCR and bacterial culture

The study was undertaken to assess the incidence of subclinical and clinical paratuberculosis in slaughtered buffaloes of Malwa region of Madhya Pradesh. They were unproductive buffaloes (1- 10 years old) slaughtered in Mhow and Indore examined for subclinical and clinical Mycobacterium avium subsp. paratuberculosis (MAP) infection. Study of gross lesions of intestine and respective mesenteric lymph nodes (MLNs) showed overall 87.33% (131/150) incidence of paratuberculosis. Out of these 131 cases of paratuberculosis, 18.21% (24/131) buffaloes suffered from subclinical and 81.68% (107/131) from clinical paratuberculosis, whereas impression smear examination revealed clinical and subclinical paratuberculosis up to 25.77 and 74.33% in intestine and 13.51 and 86.49% in MLNs, respectively.

Background: The occurrence of economic losses in buffaloes may be related to reproductive problems such as chlamydiosis caused by the bacteria Chlamydia abortus considered as a zoonotic agent; and digestive problems highlighting the infection by Mycobacterium avium subsp. paratuberculosis (Map), responsible for paratuberculosis. There is a little information about these diseases in buffaloes, therefore the aim of this study was to determine the occurrence of anti-Chlamydia abortus and anti- Mycobacterium avium subsp. paratuberculosis (Map) antibodies in water buffaloes in the state of Pernambuco, Brazil.Materials, Methods & Results: The 262 bubaline sera belonging to the serum bank of the Infectious Diseases Laboratory (LIDIC) of the Federal Rural University of Pernambuco (UFRPE) were analyzed. The samples were from nine properties distributed in the municipalities of Agreste and Zona da Mata of the state of Pernambuco. For the detection of anti-Chlamydia abortus and anti-Map antibodies was used a technique of Enzyme immunoabsorption Assay (ELISA) of the IDEXX® by following the manufacturer's instructions. Regarding the detection of anti-Chlamydia abortus antibodies, it was observed that 47.70% (125/262) of the samples were positive. All properties showed at least one positive animal for the investigation of anti-Chlamydia abortus antibodies. It was also verified the occurrence of 7.25% of suspected animals for the investigation of anti-C. abortus antibodies. No positives animals were observed for Map in the bubaline analyzed.Discussion: The occurrence of anti-C. abortus antibodies in buffaloes in the region can be explained by the fact that properties with buffalo breeding has sanitary management that allows the contact between animals, thus increasing the risk of transmission of the agent. In addition, the variation found from 35% to 68.75% may be associated with divergences to the type of management and breeding system used in each property. The percentage of suspected animals may suggest that the number of positive animals is higher or that there were non-specific reactions with other species of Chlamydia, but it is not possible to determine the seroconversion without the accomplishment of paired serology. Because it is a zoonosis, C. abortus may be a risk to the health of the population involved, since the transmission of the bacteria to humans can occur by contact with secretions and excretions of these animals. No positive animals were found for the occurrence of anti-Map antibodies, however, there are reports of the infection identifying animals with clinical signs or properties with a history of the disease, and this may be related to the differences in each property in relation to the history and sanitary management. Moreover, the ELISA test may not be sensitive to the agent depending on the phase of the infection, because if the response is predominantly cellular, the number of antibodies is diminished, making the serological diagnosis difficult. The use of more sensitive tests for the bubaline species can also favor the diagnosis of the infection. Although the occurrence of anti-Map antibodies in the analyzed samples was not detected, it is important to carry out routine epidemiological studies, since the disease has already been registered in water buffaloes in the state of Pernambuco. Considering the occurrence of anti-Chlamydia abortus antibodies it is suggested to conduct studies in order to isolate the agent and verify its real importance in the reproductive disorders of the bubaline species, since this agent causes reproductive losses and has a zoonotic character.

A total of thirty sheep with clinical symptomatology of paratuberculosis (Johne’s disease) were subjected to four diagnostic techniques: histopathological examination, bacteriological culture (in faeces and tissues), polymerase chain reaction (PCR) (in blood, tissue and faecal samples) and antibody responses (ELISA). Twenty-one (70.0%) animals showed histological lesions. Bacterial culture of both faeces and tissue revealed that 2 (6.7%) and 6 (20.0%) of the 30 sheep were infected, respectively. Mycobacterium avium subsp. 24 paratuberculosis (Map) was identified in 4 animals via PCR of faeces (13.3%), and in 19 (63.3%) by PCR in tissues. PCR in blood revealed 7 (23.3%) infected animals. Three (10.0%) animals showed antibodies against Map. A greater number of positive animals were detected by histopathological examination and PCR in tissues than by culture or ELISA. This study confirmed the clinical findings and results suggest that histopathology, PCR in tissues and in blood can improve the detection of Map in physically suspected animals and should be considered useful tools in the diagnosis of Map in suspected sheep.

Resumo: Com o objetivo de realizar um estudo dos teores de cobre (Cu), zinco (Zn) e ferro (Fe) em búfalas com paratuberculose (PTB) foram utilizadas 13 búfalas, das raças Murrah, Mediterrâneo e seus mestiços acima de três anos de idade, pertencentes a duas propriedades localizadas nos municípios de São Luiz e São Mateus, no Estado do Maranhão. Os animais foram selecionados de acordo com a presença de sinais clínicos sugestivos de paratuberculose, caracterizados por estado nutricional regular a ruim, diarreia crônica líquida a semi-líquida, desidratação, edema submandibular, anestro prolongado, mastites e verminose gastrintestinal. Foi realizada biópsia retal em todos os animais, para detecção de Mycobacterium avium subsp. paratuberculosis (Map) por meio da qPCR, e exames histopatológicos (HE e Ziehl-Neelsen). No Grupo1 sete animais foram positivos para presença de Map, e no Grupo 2 seis foram negativos. Todos os búfalos foram eutanasiados e necropsiados para coleta de diversos tecidos. Parte dos fragmentos foram fixados em formol a 10% para histopatologia e fragmentos de tecido hepático foram congelados para as dosagens dos microminerais (Cu, Zn e Fe). À necropsia todos os animais positivos para PTB apresentavam linfonodos mesentéricos de coloração castanha sugestiva de hemossiderose. Adicionalmente, em um animal foram observados pequenos pontos de cor marrom distribuídos difusamente na mucosa do intestino delgado. Na histopatologia foi observada hemossiderose moderada a acentuada no baço dos animais do Grupo 1. Na dosagem dos microminerais todos os animais com PTB apresentaram níveis abaixo dos valores de referência para Cu e Zn. Observou-se que a média dos teores de Cu dos búfalos com PTB foi 18,0ppm e de Zn 68,6ppm. No Grupo 2 a média dos teores de Cu foi 113,7ppm e de Zn 110,0ppm. Os teores de Fe em ambos os grupos foram elevados (>670ppm). Baseado nos achados clínico-patológicos e nas dosagens de minerais realizadas neste estudo, conclui-se que na região estudada, a PTB agravou o quadro clínico de animais com deficiência de Cu e Zn. Em áreas menos deficientes desses minerais sugere-se que a doença seja capaz de induzir quadros de deficiência mineral secundária.
