ABSTRACT Extracellular Penicillium fellutanumexo-β-d-galactofuranosidase, with a mass of 70 kDa, was purified to apparent homogeneity. The enzyme was used to investigate the influence of phosphodiesters of the peptidophosphogalactomannans pP2GMii and pP25GMii(containing 2 and 25 phosphodiester residues, respectively, per mol of polymer) on the kinetic parameters of galactofuranosyl hydrolysis of these two polymers, of 1-O-methyl-β-d-galactofuranoside, and of two galactofuranooligosaccharides. The enzyme did not hydrolyze phosphorylated galactose residues of pP2GMii or pP25GMii. Thek cat/Km value for pP25GMii is 1.7 × 103M−1 s−1, that for 1-O-methyl-β-d-galactofuranoside is 1.1 × 104 M−1 s−1, that for pP2GMii is 1.7 × 10 4M−1 s−1, and those for 5-O-β-d-galactofuranooligosaccharides with degrees of polymerization of 3.4 and 5.5 are 1.7 × 105 and 4.1 × 105 M−1s−1, respectively. Variability in thek cat/Km values is due primarily to differences in Km values; thek −1/k 1 ratio likely provides the most influence on Km. k cat increases as the degree of polymerization of galactofuranosyl residues increases. Most of the galactofuranosyl and phosphocholine residues were removed by day 8 in vivo from pPxGMii added to day 3 cultures initiated in medium containing 2 mM phosphate but not from those initially containing 20 mM phosphate. The filtrates from day 9 cultures initiated in 2 mM inorganic phosphate in modified Raulin-Thom medium contained 0.2 mM inorganic phosphate and 2.2 U of galactofuranosidase ml−1h−1. No galactofuranosidase activity but 15 mM inorganic phosphate was found in filtrates from day 9 cultures initiated in 20 mM phosphate. In vivo the rate of galactofuranosyl hydrolysis of pPxGMii and of related polymers is proportional to thek cat/Km value of each polymer. The kinetic data show that thek cat/Km value increases as the number of phosphodiesters of pPxGMiidecreases, also resulting in an increase in the activity of exo-β-d-galactofuranosidase.

The kinetics of the non-enzymatic hydrolysis of adenosine diphosphate in aqueous solution have been studied at pH 3.5 to 10.5 and temperatures from 80° to 95 °C. The reaction has been followed by measuring colorimetrically the inorganic phosphate liberated according to the over-all reaction[Formula: see text]The reaction has been found to be first order with respect to ADP concentration and to be catalyzed by hydrogen ions. From rate studies at pH 8.0 an activation energy of 24.2 kcal./mole was derived. A mechanism is proposed to account for the observed facts and the mechanism for the hydrolysis of adenosine triphosphate is also discussed.

A variety of systems use nucleoside triphosphate hydrolysis to control or provide energy for biological processes, mediated through protein-protein interactions. The nature of this coupling may vary, but often there is a degree of similarity. In this paper, two systems are compared: actomyosin in muscle and p21ras in a signal transduction pathway as yet undefined. The mechanism of the nucleotide triphosphate hydrolysis and the consequent changes in the protein-nucleotide complex have been investigated, to understand how the coupling to biological function is achieved. The basal nucleoside triphosphatase mechanisms are compared and the roles of proteins that activate the hydrolysis, actin and GAP, are discussed. The cleavage process was probed by stereochemical techniques to determine the basic mechanism, of either a phosphorylated enzyme intermediate or direct displacement of nucleoside diphosphate by water. Phosphate-water oxygen exchange probes were used to investigate nucleoside triphosphate and inorganic phosphate release steps. A new method of probing the kinetics of inorganic phosphate release directly has been developed. In muscle, this process seems likely to be related directly to force generation. In the GAP-ras system, measurement of phosphate release is allowing the mechanism of the GAP-p21ras interaction to be probed.

A micromethod for the determination of Na-K-ATPase in discrete segments of nephrons from rabbit, rat, and mouse kidneys is described. To facilitate tubule microdissection, the kidneys were perfused with collagenase after it had been verified that collagenase had no effect on ATPase activity. Individual tubule segments were dissected under stereomicroscopic observation, exposed to a hypotonic environment followed by rapid freezing, and incubated in 1 microliter assay medium. Enzyme activity was determined by direct measurement of labeled inorganic phosphate release by the hydrolysis of [gamma-32P]ATP and was expressed as a function of tubule length. This method is technically simple enough to permit simultaneous measurement of the enzyme in large numbers of tubules and sufficiently sensitive to determine its activity in each region of the nephron. Correlation of Na-K-ATPase activity in single tubules with functional measurements obtained in the corresponding segment of the nephron with the perfused tubule or micropuncture techniques should help define the role of this enzyme in tubular ion transport.

The kinetics of alkaline hydrolysis of succinic acid monomethyl and monopropyl esters were studied in mixed aqueous-nonaqueous media at various temperatures and ionic strengths. The results of measurements are discussed in terms of electrostatic and specific interactions between the reactants and other components of the reaction mixture. The kinetic parameters in the media under study are related to the influence of the cosolvent on the solvation sphere of the reactants.

Glycerol causes chemical oscillations in Belousov-Zhabotinskii reaction in a closed system as well as in a reaction solution bubbled with nitrogen. Since the oxidation of glycerol with bromate ions does not proceed autocatalytically and bromine in the oxidation state 0 or +1 in the absence of light does not react with glycerol, hydrolysis of bromine is the probable source of bromide ions in the studied oscillation system.

The rate of hydrolysis of S2O82- ions in acidic medium to peroxomonosulphuric acid was measured at 20 and 30 °C. The composition of the starting solution corresponded to the anolyte flowing out from an electrolyser for production of this acid or its ammonium salt at various degrees of conversion and starting molar ratios of sulphuric acid to ammonium sulphate. The measured data served to calculate the rate constants at both temperatures on the basis of the earlier proposed mechanism of the hydrolysis, and their dependence on the ionic strength was studied.
