Milk progesterone profiles during anoestrus through to pregnancy in Greek dairy goats (Capra prisca): the effect of melatonin treatment and male introduction

AbstractTwo groups of 10 lactating does were housed separately, in isolation from bucks. Does in one group (T) were implanted with melatonin; the second group (U) were not implanted. After 30 days, five implanted and five non-implanted bucks were introduced, singly, into both groups of females each day. Subsamples of milk were taken from the daily output of each doe on 3 days each week for progesterone analysis.Mean milk progesterone concentration during anoestrus varied significantly amongst individuals (P < 0·001), but the difference between T and U groups was not significant (1·51 and 1·93 (s.e.d. 0·52) μg/l, respectively). Elevated progesterone levels, indicative of a short oestrous cycle and commencing days 2 to 5 after male exposure, were observed in most does. Mean progesterone concentrations were 9·9 and 9·6 (s.e.d. 1·32) μg/l in T and U does respectively. Most does showed a second short cycle starting at 12 to 18 days, (mean concentration 8·4 and 10·6 (s.e.d. 1·64) ng/l, respectively). Untreated does then returned to anoestrus until mated at 63·1 (s.e. 0·94) days after male introduction. In contrast, T does either became pregnant or had a normal oestrous cycle, followed by a repeat cycle or pregnancy (mean onset 21·7 (s.e. 2·22) days after male introduction). The mean time of effective mating for group T does was 37·3 (s.e. 5·91) days, 26 days earlier than for U does (P < 0·001). Mean progesterone concentration in early pregnancy was 65·7 (s.e. 5·78) and 67·6 (s.e. 5·41) μg/l in T and U does, respectively. Males did not become sexually active until after the onset of luteal activity in the females, although treated males were earlier than untreated. There were no significant differences in lactation pattern associated with the progesterone profiles in the two groups.The introduction of males during late anoestrus initiates short luteal cycles. Only those does closest to onset of the breeding season, in this case induced by melatonin, continue to cycle normally and mating success may require early stimulation of sexual activity in males.

SUMMARY Progesterone levels in the blood of ewes were assayed chemically. No progesterone was detected in blood from the jugular vein of any ewe. It was found in blood from the vein draining the active ovary during the oestrous cycle. Detectable amounts appeared on the 3rd day, and the mean concentration increased to about 1·8 μg/ml. on the 7th day. This level was maintained until the 16th day and fell to <0·15 μg/ml. on the 17th or last day of the cycle. Blood from the vein draining the active ovary during pregnancy showed a mean level similar to that reached during the oestrous cycle until about the 17th week when it gradually fell to, and remained at, <0·15 μg/ml. a few days before parturition. In blood from the vein draining the pregnant horn of the uterus, progesterone was detected in low concentrations between the 9th and 18th weeks of pregnancy in only six out of 143 cases. Considerable variation between ewes in progesterone concentration was found.

ABSTRACT Progesterone was determined and identified in human peripheral blood during the preovulatory period of the menstrual cycle, by combined isotope derivative and recrystallization analysis. The mean concentration of progesterone in 1.095 ml of plasma obtained 9 days before ovulation was 0.084 μg/100 ml. However, the mean concentration of progesterone in 1.122 ml of plasma obtained 4 days before ovulation was 0.279 μg/100 ml. These data demonstrate a source of progesterone secretion other than the corpus luteum. The higher plasma-progesterone concentration 4 days before ovulation may indicate progesterone secretion of the ripening Graafian follicle of the ovary.

Objective: The main objective of this study is to evaluate the availability of lateral antebrachial cutaneous nerve (LACN) autograft for acute or delayed repair of segmented digital nerve injuries. Patients and Methods: 13 digital nerve defects of 11 patients; treated with interposition of LACN graft that harvested from ipsilateral extremity were included in the study. Mean follow up period was 35, 7 months. The mean time from injury to grafting is 53, 3 days. The results of the mean 2PDT and SWMT values of injured /uninjured finger at the end of follow up period were evaluated with Paired T test. The correlation between the defect length and the difference of 2PDT, SWMT values between the uninjured and injured finger at the end of follow up period; were evaluated with Pearson - correlation analysis. Results: The mean value of our 2PDT and SWMT results are ~5,923, ~3, 52, respectively in which can be interpreted between the normal and diminished light touch. The defect length and difference percentage of SWMT values is positively and significantly correlated statistically. Mean length of interposed nerve grafts was 18.5 mm. The age of the patient and the mean values of 2PDT and SWMT with the difference % of 2PDT and % of SWMT are not statistically correlated. Conclusion: Based on results regarding sensory regaining at recipient side and negligible sensory deficit at harvesting side, we suggest that lateral antebrachial cutaneous nerve might be a valuable graft option for digital nerve defects.

Objectives: To analyze the reproducibility of the new iC100 rebound tonometer, to compare its results with the applanation tonometry and iCare PRO and to evaluate the preference between them. Materials and methods: For the study of reproducibility, 15 eyes of 15 healthy Caucasian subjects were included. Three measurements were taken each day in three separate sessions. For the comparative study, 150 eyes of 150 Caucasian subjects were included (75 normal subjects and 75 patients with glaucoma). Three consecutive measurements were collected with each tonometer, randomizing the order of use. The discomfort caused by each tonometer was evaluated using the visual analogue scale. Results: No statistically significant differences were detected between sessions. In the comparison between tonometers, the measurements with iC100 were statistically lower than those of Perkins (−1.35 ± 0.417, p = 0.004) and that iCare PRO (−1.41 ± 0.417, p = 0.002). The difference between PRO and Perkins was not statistically significant ( p = 0.990). The mean time of measurement (in seconds) with iC100 was significantly lower than with Perkins (6.74 ± 1.46 vs 15.53 ± 2.01, p < 0.001) and that PRO (6.74 ± 1.46 vs 11.53 ± 1.85, p < 0.001). Visual analogue scale score with iC100 was lower than Perkins (1.33 ± 0.99 vs 1.73 ± 1.10, p < 0.05). In total, 61.7% preferred iC100 against Perkins. Conclusion: The reproducibility of this instrument has been proven good. iC100 underestimates intraocular pressure compared to applanation tonometry at normal values and tends to overestimate it in high intraocular pressure values. Most of the subjects preferred iC100 tonometer.

Category: Trauma Introduction/Purpose: The aim of this study was to evaluate and compare the clinical and radiological results of internal fixation with headless cannulated screw and locking compression distal ulna hook plate for the fracture at the base of fifth metatarsal bone, Zone 1. Methods: From April 2012 to April 2015, thirty cases (29 patients) were evaluated retrospectively. The mean follow up periods was 13 months. There were divided two groups based on use of the screw (group A, n=15) or the plate (group B, n=15).We measured the displacement to diastasis of the fracture on the foot oblique radiographs taken pre- and post-operatively in each group, checked the time to bone union and the difference of the reduction distance in each group. Clinical results were evaluated using American Orthopedic Foot and Ankle Society (AOFAS) midfoot score at 12 months postoperative. Results: In group A, the mean time to union was 54.2±9.3 days, the mean displacement to diastasis improved to 0.3±0.4 mm postoperatively (p<0.001), and the mean reduction distance was 2.9±1.0 mm. In group B, the mean time to union was 41.5±7.0 days, the mean displacement to diastasis improved to 0.06±0.2 mm postoperatively (p<0.001), and the mean reduction distance was 4.1±1.6 mm. AOFAS score was verified 97.7±3.4 in group A and 98.2±3.2 in group B. The time to union was significantly different between groups A and B (p=0.01).There were no complications. Conclusion: We suggest that the plate is more effective method for the shorter union time in surgical treatment of fifth metatarsal base fractures.

SUMMARY Seven trials were conducted with sixty-nine ovariectomized Suffolk cross-bred ewes to determine the requirements of oestradiol benzoate (ODB), given alone or preceded by 75 mg progesterone (6 × 12·5 mg in oil over 3 days, followed 2 days later by oestrogen), for oestrous behaviour and characteristic vaginal changes. Progesterone pretreatment resulted in a marked increase of sensitivity to oestrogen. For oestrus, the respective values of the median effective dose (ED 50) for ODB preceded by progesterone and for ODB alone were 22 and 64 μg, the difference being significant (P<0·001). The 99% fiducial limits associated with these estimates were respectively 19 and 26 μg and 52 and 81 μg. For vaginal changes the corresponding values were 10-14-17 and 20-24-28 (P<0·001). Progesterone pretreatment resulted in an apparently steeper dose-response line for oestrus, and advanced the mean time of onset by about 12 hr. The behaviour pattern following progesterone—ODB appeared to differ from that following ODB alone. Oestrus in the ewe appears to be under dual hormonal control. Endogenous oestrogen production is insufficient to induce the full psychic and physiological changes associated with normal oestrus, unless the animal has been conditioned previously by progesterone.

ABSTRACT The internal variations (i. e. in timing, duration and amplitude) of the LH surge mechanism within individual rats were examined by monitoring from 3 to 7 successive pro-oestrous LH surges in each of 5 regular 4-day cycling rats fitted with chronic intravenous cannulas. On each successive pro-oestrus blood was collected (0.5–0.6 ml hourly from 14.00–21.00) for radioimmunoassay of LH. The surgery of cannulation had no long-term effect on the regularity of the oestrous cycle. Two rats did, however, show briefly irregular cycles, including one with a 9-day period of anoestrus (pseudopregnancy). In three of the five rats successive pro-oestrous plasma LH curves (4 in one and 3 each in the other two) were internally very consistent in timing, shape and amplitude, However, between each 2 of these 3 animals there were distinct differences in the LH secretory patterns, by as much as 2 h in timing of the onset of the surge and its peak amplitude. The first two surges of the other two rats were atypical of their subsequent surges, which were mostly consistent in timing and amplitude. The pro-oestrous LH surge following the 9-day period of anoestrus was advanced by 2 h and elevated to twice the mean peak amplitude of the cyclic LH surges in that rat. Subsequently, post-PSP surges were studied in rats made pseudopregnant by mechanical stimulation of the cervix. In all cases the immediate post-PSP surge occurred earlier in the afternoon and with a greater peak amplitude than the subsequent cyclic LH surge in the same rat.

ABSTRACT The concentration of oestrone sulphate in whey obtained from 66 pregnant dairy goats was measured by direct radioimmunoassay. The mean time at which pregnancy was first detected was day 41 of gestation. Levels remained low (37 pmol/l–0·96 nmol/l, mean = 167 pmol/l) until week 5 of gestation when they rose rapidly. The test had an accuracy of 95·6%, was able to distinguish true from pseudopregnancy, and suggested that pregnancy in females carrying multiple fetuses could be detected earlier, possibly as a result of the fetal-placental origin of oestrone sulphate. SUMMARY Progesterone concentrations in the peripheral plasma of goats were measured by a protein-binding assay. The mean concentration was extremely low on the day of oestrus (0·2 ng/ml) and was not significantly different from that found in anoestrous or ovariectomized animals. The concentration increased to a maximum of 4 ng/ml on about day 10 of the 21-day cycle, and decreased rapidly during the last 3 days of the cycle. Plasma progesterone concentration during early pregnancy (2·5–3·5 ng/ml) was similar to the luteal phase value and remained steady from day 8 to day 60. Between days 60 and 70 there was a secondary increase in progesterone concentration which was maintained at this increased level (4·5–5·5 ng/ml) until just before parturition. In twin-bearing animals, the secondary increase was greater. Progesterone concentration decreased rapidly during the 1–2 days preceding parturition, but the concentration was still quite high on the day of parturition (1·25 ng/ml). The progesterone concentration in peripheral plasma was markedly increased during anaesthesia and the operation. After bilateral ovariectomy of the pregnant goat, peripheral progesterone concentration fell rapidly from 9 to 2·5 ng/ml during the first ½ h and then more slowly during the next 5–6 h. The animals aborted 36–48 h later. A consistent positive arterio—venous difference for progesterone was observed across the pregnant uterus in two unanaesthetized goats. These results indicate that the ovary is the main site of progesterone production in the pregnant goat and that production by the placenta is small and unlikely to influence the level of this hormone in the maternal circulation.

SummaryThe progestagen concentration of milk was measured in cows, at various times of the reproductive cycle by a competitive protein binding procedure and by radioimmunoassay. The apparent progesterone concentration was found to be significantly greater in milk (P <0–05) than in plasma, probably due to the presence of an unidentified metabolite(s). A simple, rapid radioimmunoassay was developed to measure directly the progestagen concentration in milk. A mean concentration of about 1 ng/ml at oestrus (or mating) increased, 20–24 days later, to 3–4 ng/ml in cows subsequently found to be non-pregnant and to about 7 ng/ml in pregnant animals. The possible application of this analytical procedure as the basis of a method of pregnancy diagnosis is discussed.
