Immunocytochemical localization of β1-4 galactosyltransferase in endometrial carcinoma cells

We have paid attention to the alteration of glycosyltransferase in carcinoma cells, because it might be related to the malignancy of the cells. In this connection, localization of β1-4 galactosyl transferase (β1-4 Gal T) in human endometrial carcinoma cells was examined immunocytochemically using two kinds of cell lines, each of which showed different degree of differentiation.An antibody was purified from the rabbit antiserum against the synthetic peptide, IFNRLVFRGMSC (W89) of human β1-4 Gal T coupled with KLH (keyhole limpet hemocyanine) by protein A column and peptide-affinity column chromatography. The anti-W89 serum reacts to the C-terminus of human β 1-4 Gal T and to both membrane-bound and soluble forms of the enzyme. Cell line of well differentiated endometrial adenocarcinoma (I) and that of poorly differentiated endometrial adenocarcinoma (50B) were cultivated respectively in MEM medium containing 15% FCS and 2 mM glutamine for 4 d at 37°C under 5% CO2. The cells were fixed in a mixture of 4% paraformaldehyde and 0.1% glutaraldehyde in 0.1 M Soerensen’s phosphate buffer (pH 7.4) at 4°C for 30 min, washed with PBS, then freezed and thawed. The indirect method of the peroxidase- labeled antibody technique was used for immunocytochemistry of both LM and TEM on the cell lines. The cells were dehydrated in ethanol and embedded in TAAB 812. Ultrathin sections were observed under a TEM, JEM-100S.

Recent published data have demonstrated elevated levels of human GH (hGH) in endometriosis and endometrial adenocarcinoma. Herein, we demonstrate that autocrine production of hGH can enhance the in vitro and in vivo oncogenic potential of endometrial carcinoma cells. Forced expression of hGH in endometrial carcinoma cell lines RL95-2 and AN3 resulted in an increased total cell number through enhanced cell cycle progression and decreased apoptotic cell death. In addition, autocrine hGH expression in endometrial carcinoma cells promoted anchorage-independent growth and increased cell migration/invasion in vitro. In a xenograft model of human endometrial carcinoma, autocrine hGH enhanced tumor size and progression. Changes in endometrial carcinoma cell gene expression stimulated by autocrine hGH was consistent with the altered in vitro and in vivo behavior. Functional antagonism of hGH in wild-type RL95-2 cells significantly reduced cell proliferation, cell survival, and anchorage-independent cell growth. These studies demonstrate a functional role for autocrine hGH in the development and progression of endometrial carcinoma and indicate potential therapeutic relevance of hGH antagonism in the treatment of endometrial carcinoma.

ABSTRACT Various doses of progesterone in oil and of two progestational compounds (17α-hydroxy-19-nor-progesterone caproate and 17α-hydroxyprogesterone p-butoxyphenyl propionate) have been given to 15 patients with recurrent and/or metastatic endometrial adenocarcinoma and to one patient with metastatic cervical adenocarcinoma, for periods of up to 27 weeks. Regression of lung metastases was noted in 4 of 13 patients, softening of pelvic tumour in 2 of 4, and histological alterations of tumour tissue in 4 of 5 patients. In the patient with metastases from a cervical adenocarcinoma, the disease progressed during the treatment. The significance of the observations is discussed.

Abstract Background This study aimed to investigate the expression level of the GATA6 gene in different oral cancer cells. Methods In this study, we sub-cultured normal oral epithelial cell lines HOK, human tongue squamous cell carcinoma cell lines CAL-27 and SCC-4, and human salivary gland adenoid cystic carcinoma cell lines SACC-LM and SACC-83. Subsequently, we used reverse transcription-polymerase chain reaction RT-PCR and Western blot methods to detect the mRNA and the protein expressions of GATA6 in normal oral epithelial cells, human tongue squamous cell carcinoma cells, and human salivary gland adenoid cystic carcinoma cells. Results The results of this study showed that the mRNA expression levels of GATA6 in CAL-27, SCC-4, and SACC-LM cells were significantly increased when compared with the HOK cells. However, the mRNA expression level of GATA6 in the SACC-83 cells had no significant difference compared with the HOK cells. The protein expression levels of GATA6 in the SCC-4 and SACC-LM cells were, however, significantly increased whereas the protein expression levels of GATA6 in the CAL-27 and SACC-83 cells had no significant difference when compared with the HOK cells. Conclusion The GATA6 gene may be related to the occurrence and progression of certain oral cancers.

The localization of two synthetic fragments of the C-terminal portion of atrial natriuretic factor: Arg 101-Tyr 126 which displays full biological activity and Leu 94-Arg 109 which is completely devoid of biological activity, has been investigated by immunohisto- and immunocytochemical methods in the heart of mammals (rat, mouse, guinea pig, hamster, rabbit, cat, dog, man) and nonmammalian vertebrates toad (Bufo marinus), frog (Rana catesbeiana), fish (Cyprinus carpio, Puntius schwanenfeldi, Cichlosoma biocellatum, Carrasius auratus), snake (Python reticulatus) and hen. Antibodies against the synthetic fragments of ANF were raised in rabbits and used either for immunofluorescence (Coons' technique), immunohistochemistry (unlabeled antibody technique) or immunocytochemistry (protein A-gold technique). Results obtained by immunofluorescence and by the unlabeled antibody technique were similar: antibodies against Arg 101-Tyr 126 ANF allowed visualization of granulated cardiocytes in the atria of all mammals. While the reaction was very strong in rat and mouse, it was less so in the rabbit and very weak in all other species studied including man. Antibodies against Leu 94-Arg 109 ANF produced a reaction only in the rat and mouse. In nonmammalian vertebrates, the reaction was always much stronger in atria than ventricles of all species with both antibodies.
