The Hedgehog-signaling pathway is an important target in cancer research and regenerative medicine; yet, on the cellular level, many steps are still poorly understood. Extensive studies of the bulk behavior of the key proteins in the pathway established that during signal transduction they dynamically localize in primary cilia, antenna-like solitary organelles present on most cells. The secreted Hedgehog ligand Sonic Hedgehog (SHH) binds to its receptor Patched1 (PTCH1) in primary cilia, causing its inactivation and delocalization from cilia. At the same time, the transmembrane protein Smoothened (SMO) is released of its inhibition by PTCH1 and accumulates in cilia. We used advanced, single molecule-based microscopy to investigate these processes in live cells. As previously observed for SMO, PTCH1 molecules in cilia predominantly move by diffusion and less frequently by directional transport, and spend a fraction of time confined. After treatment with SHH we observed two major changes in the motional dynamics of PTCH1 in cilia. First, PTCH1 molecules spend more time as confined, and less time freely diffusing. This result could be mimicked by a depletion of cholesterol from cells. Second, after treatment with SHH, but not after cholesterol depletion, the molecules that remain in the diffusive state showed a significant increase in the diffusion coefficient. Therefore, PTCH1 inactivation by SHH changes the diffusive motion of PTCH1, possibly by modifying the membrane microenvironment in which PTCH1 resides.

Heavy truck cab manual oil pump consumes more time and strength. Using tilting motor can produce larger tilting force for reversing the heavy truck cab and improve the tilting efficiency. The power of tilting motor, permanent magnet, armature plate and winding parameters are optimized on the basis of analyzing the cab tilting system works and the characteristics of the permanent magnet DC motor. The designed Nb-Fe-B PM DC motor is small volume, light weight high efficiency, simple structure and reliable operation characteristics, which can improve the security of tilting device of the heavy truck cab.

A feasibility study was conducted to determine the use of polyphthalamide/glass-fiber and polyphthalamide/glass-fiber/polytetrafluoroethylene-based composites as substitutes for aluminum and steel, respectively, in the production of motorcycle oil pump parts (housing, shaft/inner gerotor and outer gerotor). New and used (80,000 km) oil pumps were subjected to performance tests, whose results indicated that the pressure and temperature of the used pump reached a maximum of 1.8 bar and 93℃, respectively. Thermogravimetric analysis indicated that the materials are stable at the maximum operating temperature, which is 20℃ lower than the minimum glass transition temperature obtained by dynamic mechanical analysis for both materials at the analyzed frequencies (defined after calculations based on rotations in neutral, medium and high gear). The pressure value was multiplied by a safety factor of at least 1.6 (i.e., 3 bar), which was used as input for a finite element analysis of the parts, as well as the elasticity modulus at glass transition temperatures obtained by dynamic mechanical analysis. The finite element analysis indicated that the von Mises stresses to which the composite parts were subjected are 7 to 50 times lower than those the materials can withstand. The results suggest that it is feasible to manufacture motorcycle oil pump parts with these composites.

An electric oil pump (EOP) was integrated into the hydraulic system and an automatic transmission (AT) mechanical oil pump (MOP) was downsized. These processes were performed to combine a start-stop function with the AT and further improve the transmission efficiency. Furthermore, this study established a dynamics model of power loss and leakage of an 8-speed AT; a flow-based control algorithm of the EOP was then developed to realize the start-stop function and support the MOP to meet the flow requirement of the system. Based on a driving simulation method, sizes of the MOP and EOP that ensured optimal fuel economy were selected. A control strategy for the starting clutch was also developed to minimize the starting delay of the test vehicle. A test environment on a rig and prototype vehicle was established to verify the feasibility of the proposed control strategies. The test results indicated that the transmission functioned favorably with the novel two-pump system presented, and a quick and smooth starting performance was achieved when the engine was restarted. The findings in this study are extremely valuable for forward designs of an AT for realizing start-stop function and improving efficiency.
