The uptake- and depuration-rate constants (k1 and k2 respectively), according to first-order kinetics, were measured for a series of chlorobenzenes with juvenile crabs, Portunus pelagicus. These constants were found to be related to the lipophilicity of the chlorobenzenes as expressed by the octanol-water coefficient (log Kow), giving relationships similar to those observed with fish. However, the actual magnitude of k1 and k2 on a lipid basis ranged from 720 to 5880 h-1 and from 0.492 to 0.0102 h-1 respectively, which is about ten times faster than those for fish. The bioconcentration factor (KB in wet weight units) obtained from these rate constants was related to Kow, by the following expression. log KB=-2.88+1.09 log Kow. The empirical constants in this equation are similar to those observed with other aquatic organisms except that the value of -2.88 is lower than that reported with the other organisms. This is probably due to the relatively low lipid content of the crabs. It is suggested that the bioconcentration of essentially nonbiodegradable lipophilic compounds occurs as a result of partitioning between biota lipid and water. Other physicochemical properties (molar volume and aqueous solubility) and two molecular descriptors (zero- and first-order Randik indices) exhibited good correlations with the bioconcentration characteristics described above.

Resting cells of Pseudomonas strain LB400, grown on biphenyl, transformed 80, 50, and 17% of Aroclor 1242, 1254, and 1260, respectively. Resting cells grown on glucose or glycerol also transformed these polychlorinated biphenyl (PCB) mixtures to the extent of 60, 35, and 9% for Aroclors 1242, 1254, and 1260, respectively. Time courses of the transformation of the separated individual congeners in the Aroclors were plotted and used to determine the transformation rate constants (k). By analysis of the rate constants, it was concluded that the order of degradation of the different congeners in an Aroclor were similar regardless of the growth substrate. In general, k values for the conversion of a particular congener were lower for cells grown on glucose or glycerol compared with cells grown on biphenyl. Generally, k values for the transformation of the same congener in different Aroclors were not the same: rate constants had highest values for the congener in Aroclor 1242 and lowest values in Aroclor 1260. The data allowed congeners to be grouped according to their relative rates of degradation. The ratio of k values for transformation of individual congeners in Aroclors by cells grown on biphenyl and glucose were not constant.Key words: Pseudomonas strain LB400, polychlorinated biphenyls, Aroclors, transformation, resting cells.

The widespread contamination of the environment with PCB, a compound used extensively by industry in hydraulic and heat transfer fluids as well as plasticizers and solvents in adhesives and sealants, has resulted in detectable tissue levels in a large portion of the human population, domestic animals, and wildlife. Intoxication with PCB produces severe hepatic necrosis, degeneration of lymphoid tissues and kidney, skin lesions, decreased reproductive performance, reduced feed efficiency, and decreased weight gain. PCB also has been reported to reduce the binding of thyroid hormone to serum proteins and enhance the peripheral metabolism of thyroxine with increased excretion of thyroxine-glucuronide in the bile (Bastomsky, Endocrinology 95: 1150-1155, 1974).The objectives of this investigation were (1) to investigate the histopathologic, histochemical, and ultrastructural changes in thyroid FC produced by the acute (4 week) and chronic (12 week) administration of low (50 ppm) and high (500 ppm) doses of PCB to rats, (2) to correlate these alterations to changes in serum immunoreactive thyroxine concentration, and (3) to investigate the persistence of the effects of PCB on the thyroid gland.

Polychlorinated biphenyls (PCBs) are a ubiquitous class of environmental pollutants with toxic and hepatocellular effects, including accumulation of fat, proliferated smooth endoplasmic recticulum (SER), and concentric membrane arrays (CMAs) (1-3). The CMAs appear to be a membrane storage and degeneration organelle composed of a large number of concentric membrane layers usually surrounding one or more lipid droplets often with internalized membrane fragments (3). The present study documents liver alteration after a short term single dose exposure to PCBs with high chlorine content, and correlates them with reported animal weights and central nervous system (CNS) measures. In the brain PCB congeners were concentrated in particular regions (4) while catecholamine concentrations were decreased (4-6). Urinary levels of homovanillic acid a dopamine metabolite were evaluated (7).Wistar rats were gavaged with corn oil (6 controls), or with a 1:1 mixture of Aroclor 1254 and 1260 in corn oil at 500 or 1000 mg total PCB/kg (6 at each level).

SummaryRecombinant chimaeric molecules between tissue-type plasminogen activator (t-PA) and single chain urokinase-type plasminogen activator (scu-PA) or two chain urokinase-type plasminogen activator (tcu-PA) have intact enzymatic properties of scu-PA or tcu-PA towards natural and synthetic substrates (Nelles et al., J Biol Chem 1987; 262: 10855-10862). In the present study, we have compared the reactivity with inhibitors of both the single chain and two chain variants of recombinant u-PA and two recombinant chimaeric molecules between t-PA and scu-PA (t-PA/u-PA-s: amino acids 1-263 of t-PA and 144-411 of u-PA; t-PA/u-PA-e: amino acids 1-274 of t-PA and 138-411 of u-PA). Incubation with human plasma in the absence of a fibrin clot for 3 h at 37° C at equipotent concentrations (50% clot lysis in 2 h), resulted in significant fibrinogen breakdown (to about 40% of the normal value) for all two chain molecules, but not for their single chain counterparts. Preincubation of the plasminogen activators with plasma for 3 h at 37° C, resulted in complete inhibition of the fibrinolytic potency of the two chain molecules but did not alter the potency of the single chain molecules. Inhibition of the two chain molecules occurred with a t½ of approximately 45 min. The two chain variants were inhibited by the synthetic urokinase inhibitor Glu-Gly-Arg-CH2CCl with apparent second-order rate constants of 8,000-10,000 M−1s−1, by purified α2-antiplasmin with second-order rate constants of about 300 M−1s−1, and by plasminogen activator inhibitor-1 (PAI-1) with second-order rate constants of approximately 2 × 107 M−1s−1.It is concluded that the reactivity of single chain and two chain forms of t-PA/u-PA chimaers with inhibitors is very similar to that of the single and two chain forms of intact u-PA.
