Elucidation of the Nucleophilic Potential of Diazocyclopentadiene

Diazocyclopentadiene reacts with benzhydrylium ions (Ar2CH+) to give 2,5-dibenzhydryl-substituted diazocyclopentadienes. The kinetics have been determined photometrically in dichloromethane under pseudo-first-order conditions using diazocyclopentadiene in excess. Plots of the second-order rate constants (log k 2) versus the electrophilicity parameters E of the benzhydrylium ions gave the nucleo­philicity parameter N = 4.84 and susceptibility s N = 1.06 for diazo­cyclopentadiene according to the correlation log k(20 °C) = s N(E + N). Diazocyclopentadiene thus has a similar nucleophilic reactivity as pyrrole. Previously reported electrophilic substitutions of diazocyclopentadiene are rationalized by these parameters and new reaction possibilities are predicted.

Visual Interpretation of the Meaning of kcat/KM in Enzyme Kinetics

kcat and kcat/KM are the two fundamental kinetic parameters in enzyme kinetics. kcat is the first-order rate constant that determines the reaction rate when the enzyme is fully occupied at a saturating concentration of the substrate. kcat/KM is the second-order rate constant that determines the reaction rate when the enzyme is mostly free at a very low concentration of the substrate. Both parameters provide critical information on how the enzyme lowers the energy barriers along the reaction pathway for catalysis. However, it is surprising how often kcat/KM is used inappropriately as a composite parameter derived by dividing kcat with KM to assess both catalytic power and affinity to the substrate of the enzyme. The main challenge in explaining the true meaning of kcat/KM is the difficulty to demonstrate how the reaction energetics of enzyme catalysis determines kcat/KM in a simple way. Here, I report a step-by-step demonstration on how to visualize the meaning of kcat/KM on the reaction energy diagram. By using the reciprocal form of the expression of kcat/KM with the elementary rate constants in kinetic models, I show that kcat/KM is a harmonic sum of several kinetic terms that correspond to the heights of the transition states relative to the free enzyme. Then, I demonstrate that the height of the highest transition state has the dominant influence on kcat/KM, i. e. the step with the highest transition state is the limiting step for kcat/KM. The visualization of the meaning of kcat/KM on the reaction energy diagram offers an intuitive way to understand all the known properties of kcat/KM, including the Haldane relationship.

Application of polymer-coated Macadamia integrifolia nutshell biomass impregnated with palladium for chromium(VI) remediation

Freely suspended and porous basket restrained granules of palladium nanoparticles supported on polymer-grafted Macadamia nutshell biomass (Pd@Polym-MNS) composite were used for the treatment chromium(VI)-containing water. In the presence of formic acid, the Pd@Polym-MNS demonstrated its activity in the adsorption-reduction-based conversion of noxious chromium(VI) to less toxic chromium(III) with a low activation energy of 13.4 kJ mol–1, ΔH0 (+ 10.8 kJ mol–1), ΔS0 (−270.0 J mol–1 K–1), and ΔG0 (+ 91.3 to + 98.0 kJ mol–1) indicated the exothermic, endergonic and non-spontaneous nature of the catalytic redox reaction. In addition to facilitating easy recovery, rinsing, and reuse, restraining the Pd@Polym-MNS in the basket reactor helped maintain the integrity of the catalysts by preventing violent collisions of suspended granules with the mixing apparatus and the walls of the reaction vessel. Whereas the pseudo-first-order rate constant was recorded as 0.157 min–1 upon initial use, values of the mean and relative standard deviation for the second, third and fourth consecutive uses were found to be 0.219 min–1 and 1.3%, respectively. According to a response surface methodological approach to batch experimentation, the initial concentration of chromium(VI) and catalyst dosage had the greatest impact on the redox reaction rate, accounting for 85.7% and 11.6% of the variability in the value of the pseudo-first-order rate constant, respectively. Mutually beneficial effects of the combinations of high formic acid and low chromium(VI) concentration, high temperature and catalyst dosage as well as high formic acid and catalyst dosage were recorded.

<p>The reaction of bis-(diaminoethane)nickel(II) chloride, ([Ni(en)2]Cl2 in methanol with formaldehyde and nitroethane in the presence of triethylamine proceeds readily to produce (6, 13-dimethyl-6, 13-dinitro-1, 4, 8, 11-tetraazacyclotetradecane)nickel(II) chloride, [Ni(dini)] - Cl2. Reduction of the nitro groups of this compound by catalytic hydrogenation yields three isomers of the pendant arm macrocyclic complex (6, 13-diamino-6, 13-dimethyl-1, 4, 8, 11-tetraazachyclotetradecane)nickel(II) chloride, designated a-, b- and c-[Ni(diam)]Cl2. These were separated by fractional crystallization. The aisomer was observed to isomerizes slowly in solution to the b- form. A parallel dissociation reaction of the a- isomer was also observed. The demetallation of a- and b- isomers of the diam complex of nickel by reaction with cyanide or concentrated acid at 140 degrees C produces the macrocycle meso-(6, 13-diamino-6, 13-dimethyl-1, 4, 8, 11-tetraazacyclotetra-decane), diam. A variety of hexamine, pentamine and tetramine complexes of diam with nickel(II), copper(II), cobalt(II) and (III), chromium(III), palladium(II), rhodium(III), zinc(II) and cadmium(II) were prepared. Hexamine and tetramine forms of labile metal complexes could be rapidly and reversibly interconverted by altering the pH. The hexamine cobalt(III) cation, [Co(diam)]3+ was by far the most inert of the prepared cobalt(III) complexes, remaining unaffected in hot acidic solutions. In contrast, a single pendant arm of the hexamine [Cr(diam)]3+ cation could be dissociated in acid. (Two possibly triamine complexes of lead were also prepared). These compounds were characterized by elemental analysis, magnetic measurements, electronic, infrared, 1H and 13C nuclear magnetic resonance spectra. The pendant arm protonation constants (log K) of diam and selected complexes of nickel, copper and palladium were calculated from potentiometric titration measurements at 25 degrees C. The log K values for diam at 25 degrees C (I = 0.1 M NaclO4) were 11.15, 9.7, 6.2 and 5.3. 753. Impact of COVID-19 on Healthcare Facility-Onset Clostridiodes difficile Infection

Background It is estimated that the majority of hospitalized COVID-19 patients around the world received antibiotics despite the fact that bacterial co-infections are rare. This can lead to increased antimicrobial resistance and Clostridioides difficile infections (CDI). Gastrointestinal symptoms of COVID-19 may also contribute to increased testing. The objective of this study was to assess the impact of the COVID-19 pandemic on our healthcare facility-onset (HO) CDI rates. Methods This was a retrospective cross-sectional study comparing CDI rate per 1,000 patient days, C. diff order rate per 1,000 patient days, Standardized Antimicrobial Administration Ratio (SAAR), and Standardized Infection Ratio (SIR) in the pre-COVID-19 period from January 1, 2019 to December 31, 2019 to the COVID-19 period from April 1, 2020 to March 31, 2021 at a 877-bed tertiary care hospital in Detroit, Michigan. CDI and order rates were extracted from the electronic medical record (Epic™ Bugsy). SAAR and SIR data were extracted from National Healthcare Safety Network (NHSN). Results The average CDI rate per 1,000 patient days was 4.29 pre-COVID-19 compared to 1.98 during COVID-19 with a 54% reduction, and the C. diff order rate per 1,000 patient days also decreased from 130.89 to 93.03, resulting in a 29% reduction (Figure 1). The SIR was 0.383 compared to 0.308 during COVID-19 (P-value 0.404). SAAR decreased from 1.095 to 0.945 (P-value &lt; 0.001). However, our institution experienced three COVID-19 waves, with peaks in April 2020, November 2020 and March 2021, that correlated with high risk CDI antibiotic utilization in intensive care unit (ICU) (Figure 2). Conclusion Despite the COVID-19 pandemic, the HO-CDI and C. diff order rates and overall SAAR decreased; however, antibiotic utilization increased in the ICU during the COVID-19 waves. The overall decrease may be multifactorial and related to increased hand hygiene compliance, isolation and personal protective equipment use and overall decreased antibiotic use and C. diff orders. Development and Evaluation of a Threshold-Based Motion Cueing Algorithm

In this paper, a motion cueing algorithm (MCA) without a frequency divider is proposed, which aims to reproduce the longitudinal reference acceleration as far as possible via tilt coordination. Using a second-order rate limit, the human perception thresholds can directly be taken into account when parameterizing the MCA. The washout is compensated by tilt coordination and means of feedback from the translational acceleration. The proposed MCA is compared with the classical washout algorithm and the compensation MCA based on selected qualitative metrics and their workspace demand. In addition, a subjective study on the evaluation of the MCA was conducted. The results show that even high washout rates are not noticeable by the test subjects. Overall, the MCA was rated as very good.

Evaluation of Cr(VI) Reduction Using Indigenous Bacterial Consortium Isolated from a Municipal Wastewater Sludge: Batch and Kinetic Studies

Hexavalent Chromium (Cr(VI)) has long been known to be highly mobile and toxic when compared with the other stable oxidation state, Cr(III). Cr(VI)-soluble environmental pollutants have been detected in soils and water bodies receiving industrial and agricultural waste. The reduction of Cr(VI) by microbial organisms is considered to be an environmentally compatible, less expensive and sustainable remediation alternative when compared to conventional treatment methods, such as chemical neutralization and chemical precipitation of Cr. This study aims to isolate and identify the composition of the microbial consortium culture isolated from waste activated sludge and digested sludge from a local wastewater treatment plant receiving high loads of Cr(VI) from an abandoned chrome foundry in Brits (North Waste Province, South Africa). Furthermore, the Cr(VI) reduction capability and efficiency by the isolated bacteria were investigated under a range of operational conditions, i.e., pH, temperature and Cr(VI) loading. The culture showed great efficiency in reduction capability, with 100% removal in less than 4 h at a nominal loading concentration of 50 mg Cr(VI)/L. The culture showed resilience by achieving total removal at concentrations as high as 400 mg Cr(VI)/L. The consortia exhibited considerable Cr(VI) removal efficiency in the pH range from 2 to 11, with 100% removal being achieved at a pH value of 7 at a 37 ± 1 °C incubation temperature. The time course reduction data fitted well on both first and second-order exponential rate equation yielding first-order rate constants in the range 0.615 to 0.011 h−1 and second order rate constants 0.0532 to 5 × 10−5 L·mg−1·h−1 for Cr(VI) concentration of 50–400 mg/L. This study demonstrated the bacterial consortium from municipal wastewater sludge has a high tolerance and reduction ability over a wide range of experimental conditions. Thus, show promise that bacteria could be used for hexavalent chromium remediate in contaminated sites.
