Study of the spike (S) glycoprotein from the SARS-Cov-2 as a possible source of translocating peptides of biomedical interest

Seaweed lectins, especially high-mannose-specific lectins from red algae, have been identified as potential antiviral agents that are capable of blocking the replication of various enveloped viruses like influenza virus, herpes virus, and HIV-1 in vitro. Their antiviral activity depends on the recognition of glycoprotein receptors on the surface of sensitive host cells—in particular, hemagglutinin for influenza virus or gp120 for HIV-1, which in turn triggers fusion events, allowing the entry of the viral genome into the cells and its subsequent replication. The diversity of glycans present on the S-glycoproteins forming the spikes covering the SARS-CoV-2 envelope, essentially complex type N-glycans and high-mannose type N-glycans, suggests that high-mannose-specific seaweed lectins are particularly well adapted as glycan probes for coronaviruses. This review presents a detailed study of the carbohydrate-binding specificity of high-mannose-specific seaweed lectins, demonstrating their potential to be used as specific glycan probes for coronaviruses, as well as the biomedical interest for both the detection and immobilization of SARS-CoV-2 to avoid shedding of the virus into the environment. The use of these seaweed lectins as replication blockers for SARS-CoV-2 is also discussed.

A fluorine nuclear magnetic resonance (19F-NMR)-based method is employed to assess the binding preferences and interaction details of a library of synthetic fluorinated monosaccharides towards dendritic cell-specific intercellular adhesion molecule 3-grabbing non-integrin (DC-SIGN), a lectin of biomedical interest, which is involved in different viral infections, including HIV and Ebola, and is able to recognize a variety of self- and non-self-glycans. The strategy employed allows not only screening of a mixture of compounds, but also obtaining valuable information on the specific sugar–protein interactions. The analysis of the data demonstrates that monosaccharides Fuc, Man, Glc, and Gal are able to bind DC-SIGN, although with decreasing affinity. Moreover, a new binding mode between Man moieties and DC-SIGN, which might have biological implications, is also detected for the first time. The combination of the 19F with standard proton saturation transfer difference (1H-STD-NMR) data, assisted by molecular dynamics (MD) simulations, permits us to successfully define this new binding epitope, where Man coordinates a Ca2+ ion of the lectin carbohydrate recognition domain (CRD) through the axial OH-2 and equatorial OH-3 groups, thus mimicking the Fuc/DC-SIGN binding architecture.

This article offers an analysis of the social sources of biomedical interest in women’s sports injuries through a case study of anterior cruciate ligament (ACL) injuries. Although both men and women incur them, there is extensive research interest in women’s ACL injuries. Drawing on interviews with researchers who have contributed to this research, the investigation examines the social sources of this interest. Explanations lie largely in the evolution of the agenda in sport medicine to a concern with injury prevention, which coincides with a movement toward the inclusion of women in health research. The article concludes with a consideration of the political and ideological implications of the interaction of the prevention and inclusion agendas in research on women’s sport injuries.Cet article propose une analyse des sources sociales de l’intérêt biomédical pour les blessures dans les sports féminins à travers l’étude du cas des blessures au ligament croisé antérieur (LCA). Bien que les hommes et les femmes en soient tous deux victimes, il y a énormément d’intérêt en recherche pour les blessures au LCA chez les femmes. S’appuyant sur des entrevues avec des chercheurs qui ont contribué à ce projet, l’étude examine les sources sociales de cet intérêt. Les explications reposent grandement sur l’évolution de l’agenda en médecine du sport vers un souci de prévention des blessures, ce qui coïncide avec un mouvement vers l’inclusion des femmes dans la recherche sur la santé. L’article conclut par une considération des implications politiques et idéologiques de l’interaction des agendas de prévention et d’inclusion en recherche sur les blessures sportives chez les femmes.

Among other scorpion species, Colombia has two genera of the Buthidae family Centruroides and Tityus, considered to be dangerous to humans. This research shares scientific knowledge aiming to a better understanding about the pathophysiological effects of such venoms. The venom of the three species: Centruroides margaritarus, Tityus pachyurus, and T. n. sp. aff. metuendus with biomedical interest were studied. An initial pre-glycemic sample was taken from ICR mice. They were later intraperitoneally inoculated with doses of 35% and 70% of LD50 of total venom. Poisoning signs were observed during a 6-h period to determine the level of scorpionism. After observation, a second glycemic sample was taken, and a histopathological evaluation of different organs was performed. This work revealed that all three venoms showed considerably notorious histopathological alterations in main organs such as heart and lungs; and inducing multiple organ failure, in relation to the glycemia values, only C. margaritatus and T. n. sp. aff. metuendus showed significant changes through manifestation of hyperglycemia. According to the Colombian scorpionism level; signs were mild to severe affecting the autonomous nervous system.
