Background: The antioxidant defense in the pancreas is low because they are exposed to toxic substances daily. This study aimed to evaluate the ameliorating effect of Viola odorata flowers Extract (VOE) on the pancreas histology and function in Streptozotocin (STZ)-induced diabetic rats. Methods: Forty male rats were divided into five groups, consisting of controls; STZ; and STZ plus various doses of VOE (100, 200 or 400 mg/kg). The amylase, lipase, insulin and total antioxidant capacity levels were measured in the sera. The homeostatic model assessment of insulin resistance was also measured. The histopathological alterations of the rats’ pancreases were examined microscopically. Results: The serum amylase and total antioxidant activities were reduced in diabetic rats (P=0.001). Varying doses of VOE reduced the serum amylase and glucose levels, and increased the total antioxidant activities compared to that of the diabetic rats (P<0.05). There were no significant differences in the serum lipase and insulin levels among the groups. Treatment with VOE at all doses significantly lowered the insulin resistance compared to that of the diabetic group (P=0.001). Significant reductions were observed in the areas of the pancreatic Langerhans islets and the number of beta cells in the STZ group (P=0.001). Conclusion: This study demonstrated that VOE ameliorated the adverse effects induced by STZ in the rat’s pancreas in the short-term. These effects are likely to be due to the reduced insulin resistance and amylase activity, and increased total antioxidant activity along with the histopathological alterations in the pancreas.

The increase in consumption of herbal liquors in Nigeria is a cause for alarm. These are consumed with the mis-conception that they are without toxic effects. The aim of this study is to investigate the genotoxic and histopathological alterations in rats exposed to herbal liquors. Female rats were exposed to herbal liquors for 6 weeks. The histopathological and genotoxic evaluation were done to assess extent of damage. Pathological examination revealed incidences of aggregates chronic inflammatory cell infiltrates in the heart of Jedi treated group while the heart of the other groups had no abnormalities. The histologic sections of the kidney tissue revealed congested vessels while the lung showed reduction in air filled alveolar spaces with infiltration of alveoli and interstitium by aggregates of inflammatory cells indnadicating moderate to severe pulmonary inflammation. Histologic sections of lung tissue in rats treated with herbal liquors reveals congestion of pulmonary vessels and interstitial hemorrhages. Genotoxic evaluation of rat lymphocytes exposed to herbal liquors via comet assay shows that rats administered with the different herbal liquors developed significant (p < 0.05) as revealed in the % DNA in tail, % DNA in head, olive moment, tail length and tail moment which indicates the presence of DNA strand breaks and a marker for oxidative DNA damage. This result reveals that herbal liquors contain substances that produce reactive oxygen species that have pathological effect on certain organs as well as inducing DNA strand breaks that could compromise the integrity of the DNA which can lead to mutation.

Pesticides such as endosulfan, heptachlor and dieldrin persist in aquatic environments as a result of their resistance to biodegradation. However, there is no adequate information about the toxicity of endosulfan, heptachlor and dieldrin to the aquatic organism, African catfish (Clarias gariepinus)—a high valued widely distributed commercially interesting species. The current experiment was performed with the aim to determine the median lethal concentration (LC50) of endosulfan, heptachlor and dieldrin to African catfish (Clarias gariepinus); their behavioral abnormalities and histopathological alterations in several vital organs. A total of 324 juvenile fish were exposed for 96 h to six concentrations of endosulfan and dieldrin at 0, 0.001, 0.002, 0.004, 0.008 and 0.016 ppm, and to heptachlor at concentrations of 0, 0.02, 0.04, 0.08, 0.16 and 0.32 ppm for dose-response tests. The study demonstrated that the species is highly susceptible to those contaminants showing a number of behavioral abnormalities and histopathological changes in gill, liver and muscle. The 96-h LC50 value of endosulfan, dieldrin and heptachlor for the African catfish was found as 0.004 (0.001−0.01) mg/L, 0.006 mg/L and 0.056 (0.006−0.144) mg/L, respectively. Abnormal behaviors such as erratic jerky swimming, frequent surfacing movement with gulping of air, secretion of mucus on the body and gills were observed in response to the increasing exposure concentrations. Histopathological alterations of liver, gill and muscle tissues were demonstrated as vacuolization in hepatocytes, congestion of red blood cells (RBCs) in hepatic portal vein; deformed secondary lamellae and disintegrated myotomes with disintegrated epidermis, respectively. These findings are important to monitor and responsibly manage pesticide use in and around C. gariepinus aquacultural areas.

Among other scorpion species, Colombia has two genera of the Buthidae family Centruroides and Tityus, considered to be dangerous to humans. This research shares scientific knowledge aiming to a better understanding about the pathophysiological effects of such venoms. The venom of the three species: Centruroides margaritarus, Tityus pachyurus, and T. n. sp. aff. metuendus with biomedical interest were studied. An initial pre-glycemic sample was taken from ICR mice. They were later intraperitoneally inoculated with doses of 35% and 70% of LD50 of total venom. Poisoning signs were observed during a 6-h period to determine the level of scorpionism. After observation, a second glycemic sample was taken, and a histopathological evaluation of different organs was performed. This work revealed that all three venoms showed considerably notorious histopathological alterations in main organs such as heart and lungs; and inducing multiple organ failure, in relation to the glycemia values, only C. margaritatus and T. n. sp. aff. metuendus showed significant changes through manifestation of hyperglycemia. According to the Colombian scorpionism level; signs were mild to severe affecting the autonomous nervous system.

Histopathological alterations in various fish organs have a pronounced value in aquatic toxicology and are widely used in environmental monitoring. The aim of this study was to evaluate whether histopathological alterations in fish gills can discriminate seasonal variations in environmental conditions within the same aquatic ecosystem, and if so, which alterations contributed the most to seasonal differentiation. Microscopic examination of common bream Abramis brama gills displayed various alterations in gill structure, including epithelial hypertrophy, hyperplasia, mucous and chloride cell alterations, epithelial lifting, necrosis, hyperemia and aneurism. These alterations were subsequently quantified by a semi-quantitative analysis in order to detect differences in the intensity of the mentioned alterations. Epithelial hypertrophy, hyperplasia, epithelial lifting and necrosis varied significantly between seasons with only necrosis being significantly higher in the first season. Discriminant canonical analysis displayed that epithelial hyperplasia, mucous cell alterations, epithelial lifting and necrosis contributed the most to discrimination between seasons. Overall, this study demonstrates that histopathological biomarkers in fish gills can be used in discriminating seasonal variations in water quality within the same aquatic ecosystem.
