Determining aphid taxonomic affinities and life cycles with molecular data: a case study of the tribe Cerataphidini (Hormaphididae:Aphidoidea: Hemiptera)

The biodiversity of freshwater ecosystems globally still leaves much to be discovered, not least in the trematode parasite fauna they support. Echinostome trematode parasites have complex, multiple-host life-cycles, often involving migratory bird definitive hosts, thus leading to widespread distributions. Here, we examined the echinostome diversity in freshwater ecosystems at high latitude locations in Iceland, Finland, Ireland and Alaska (USA). We report 14 echinostome species identified morphologically and molecularly from analyses of nad1 and 28S rDNA sequence data. We found echinostomes parasitising snails of 11 species from the families Lymnaeidae, Planorbidae, Physidae and Valvatidae. The number of echinostome species in different hosts did not vary greatly and ranged from one to three species. Of these 14 trematode species, we discovered four species (Echinoparyphium sp. 1, Echinoparyphium sp. 2, Neopetasiger sp. 5, and Echinostomatidae gen. sp.) as novel in Europe; we provide descriptions for the newly recorded species and those not previously associated with DNA sequences. Two species from Iceland (Neopetasiger islandicus and Echinoparyphium sp. 2) were recorded in both Iceland and North America. All species found in Ireland are new records for this country. Via an integrative taxonomic approach taken, both morphological and molecular data are provided for comparison with future studies to elucidate many of the unknown parasite life cycles and transmission routes. Our reports of species distributions spanning Europe and North America highlight the need for parasite biodiversity assessments across large geographical areas.

Traditional solutions to address interoperability issues are mainly process-centric so that consistent interactions among collaborating enterprises are ensured. These solutions examine interoperability from a technological perspective with focus on exchanging information messages between distributed and heterogeneous applications. However, interoperability from a business perspective has been overlooked in the past due to the complexity of reconciling diverse business strategies, organizational constraints, and IT infrastructures. Business interoperability denotes the ability of diverse enterprises to collaborate together to coproduce added-value products and services. In this chapter, a new line of thinking is promoted whereby interoperability is data-centric instead of process-centric. Business interoperability is dealt with by adopting business artifacts that are able to cross organizational boundaries, and by introducing a stack of three layers - strategy, service, and resource. Artifacts are self-contained business records that include attributes, states, and life cycles that reflect the changes in these states. The artifact concept not only describes a business entity, but also encompasses knowledge about what to process without explaining how to do it. The shift from processes to artifacts makes business interoperability “quite simple’’ to deploy and renders collaboration easy to manage and analyze. The chapter also introduces several interaction patterns that regulate the exchange of artifacts between enterprises. The ideas and proposals in this chapter are discussed via a realistic case-study to demonstrate how business people can seamlessly manage their day-to-day activities and intuitively construct interoperable and sustainable collaborations at the business and technological levels.

AbstractAs training in helminthology has declined in the medical microbiology curriculum, many rare species of zoonotic cestodes have fallen into obscurity. Even among specialist practitioners, knowledge of human intestinal cestode infections is often limited to three genera, Taenia, Hymenolepis and Dibothriocephalus. However, five genera of uncommonly encountered zoonotic Cyclophyllidea (Bertiella, Dipylidium, Raillietina, Inermicapsifer and Mesocestoides) may also cause patent intestinal infections in humans worldwide. Due to the limited availability of summarized and taxonomically accurate data, such cases may present a diagnostic dilemma to clinicians and laboratories alike. In this review, historical literature on these cestodes is synthesized and knowledge gaps are highlighted. Clinically relevant taxonomy, nomenclature, life cycles, morphology of human-infecting species are discussed and clarified, along with the clinical presentation, diagnostic features and molecular advances, where available. Due to the limited awareness of these agents and identifying features, it is difficult to assess the true incidence of these ‘forgotten’ cestodiases as clinical misidentifications are likely to occur. Also, the taxonomic status of many of the human-infecting species of these tapeworms is unclear, hampering accurate species identification. Further studies combining molecular data and morphological observations are necessary to resolve these long-standing taxonomic issues and to elucidate other unknown aspects of transmission and ecology.

The concept of palynological compass was introduced in 1969 by Erdtman to express the way palynological evidence points to systematic relationships. Recent evidence shows that, in the cases studied, the palynological compass points to the same phylogenetic direction as the molecular data.

Crowther et al. (1999) described an island population of an Australian dasyurid marsupial, Sminthopsis griseoventer, as a new subspecies on molecular (allozyme) and morphological grounds despite a previously published, contradictory genetic study and evidence of sympatry with its mainland conspecific. The legitimacy of this taxonomic arrangement has implications for the allocation of scarce management resources because the new taxon could be considered 'Critically Endangered'. Samples of the original tissues from which the molecular data cited by Crowther et al. were obtained no longer exist. Therefore, salient components of that work were rerun with fresh tissue and the results used to reassess evidence that the island population warrants recognition as a subspecies. We conclude that neither the molecular nor the morphological data supports differentiation at taxonomic or evolutionarily significant levels. Nevertheless, there is a good case for recognising the island population as an important management unit. This case study emphasises the importance of sound taxonomy determined by macro and molecular characters as a prerequisite to allocating resources for conservation.

Abstract Evolution does not always result in dichotomous phylogenetic trees. For instance, in anacladogenetic speciation, where a new species originates by budding, the ancestral taxon is often initially paraphyletic. Here we study Cymbalaria muelleri (Plantaginaceae), a chasmophytic species endemic to Sardinia, a major island in the Mediterranean Basin. Its distribution range is divided into two well-delimited geographical groups with some morphological differences. Using a combination of morphology, molecular data (amplified fragment length polymorphism fingerprinting) and relative genome size, we found that the two geographical groups should be considered two separate taxa, which split through anacladogenesis. Accordingly, we formally describe the new paraphyletic subspecies C. muelleri subsp. villosa as the ancestor, from which C. muelleri subsp. muelleri originated by budding. Morphological analyses support the differentiation of the two subspecies, and there are strong diagnostic characters to differentiate them. In addition to morphology and genetics, slightly divergent habitat preferences and the disjunct distribution of the two subspecies also support the recognition of two taxa. Genome size data obtained for the two subspecies are consistent with the previously established hexaploidy of C. muelleri.

The building sector plays a central role in addressing the problem of global energy consumption. Therefore, effective design measures need to be taken to ensure efficient usage and management of new structures. The challenging task for designers is to reduce energy demands while maintaining a high-quality indoor environment and low costs of construction and operations. This study proposes a methodological framework that enables decision-makers to resolve conflicts between energy demand and life cycle costs. A case study is analyzed to validate the proposed method, adopting different solutions for walls, roofs, floors, windows, window-to-wall ratios and geographical locations. Models are created on the basis of all the possible combinations between these elements, enriched by their thermal properties and construction/management costs. After the alternative models are defined, energy analyses are carried out for an estimation of consumption. By calculating the total cost of each model as the sum of construction, energy and maintenance costs, a joint analysis is carried out for variable life cycles. The obtained results from the proposed method confirm the importance of a preliminary assessment from both energy and cost points of view, and demonstrate the impact of considering different building life cycles on the choice of design alternatives.
