Experimental infections of guinea pigs and rabbits with a sublethal dose of L. monocytogenes pathogen caused an allergic reaction in the form of delayed-type hypersensitivity (DTHS), which was detected by an intradermal allergy test with a Listeriose allergen. The status of DTHS in guinea pigs and rabbits was recorded for a longer time as compared to specific antibodies. A positive allergic reaction correlated with listeria, which was confirmed by the isolation of a Listeria culture 6 months after infection of rabbits with a virulent Listeria strain. The research findings showed that an intradermal allergy test with a developed Listeria allergen allows a retrospective diagnosis of Listeriosis and Listeria carrying. A specific feature of Listeria allergen was established through an intradermal provocative test in animals sensitized by heterogeneous microorganisms (Salmonella and E. coli).

DPPD is a Mycobacterium tuberculosis recombinant antigen that elicits specific delayed type hypersensitivity reactions similar in size and morphological aspects to that elicited by purified protein derivative, in both guinea pigs and humans infected with M. tuberculosis. In addition, earlier clinical studies with DPPD suggested that this molecule could improve the specificity of the tuberculin skin test, which is used as an important aid for the diagnosis of tuberculosis. However, these studies could only be performed with DPPD engineered as a fusion molecule with another Mycobacterium spp. protein because no expression of DPPD could be achieved as a single molecule or as a conventional fusion protein in any commercial system. Although recombinant fusion proteins are in general suitable for several biological studies, they are by definition not ideal for studies involving highly purified and defined polypeptide sequences. Here, we report two alternative approaches for the expression of immunologically reactive recombinant genuine DPPD. The first approach used the rapidly growing, nonpathogenic Mycobacterium smegmatis as host cells transformed with the pSMT3 plasmid vector containing the full-length DPPD gene. The second approach used Escherichia coli transformed with the pET-17b plasmid vector containing the DPPD gene engineered in a three-copy fusion manner in tandem with itself. Though at low levels, expression and purification of immunologically reactive DPPD in M. smegmatis could be achieved. More abundant expression and purification of DPPD as a homo-trimer molecule was achieved in E. coli ([Formula: see text]2 mg/L of bacterial broth cultures). Interestingly, expression could only be achieved in host cells transformed with the DPPD gene containing its leader peptide. However, the expressed proteins lacked the leader sequence, which indicates that processing of the M. tuberculosis DPPD gene was accurately achieved and necessary in both M. smegmatis and E. coli. More importantly, the delayed type hypersensitivity reactions elicited by purified molecules in guinea pigs infected with M. tuberculosis were indistinguishable from that elicited by purified protein derivative. Because the DPPD gene is present only in the tuberculosis-complex organisms of the Mycobacterium genus, these highly purified molecules should be helpful in identifying individuals sensitized with tubercle bacilli.Key words: Mycobacterium tuberculosis, Mycobacterium smegmatis skin test, DTH, DPPD.

Aim. Study the role of LPS in induction of anti-tularemia immunity in humans and animals. Materials and methods. Activity of various antigenic preparations of tularemia microbe, including highly purified from protein and S- and R-LPS, was studied using leukocytolysis reaction with blood of vaccinated humans and guinea pigs and skin allergy test (guinea pigs). Results. Only the whole cells of Francisella tularensis, killed in protein non-denaturating conditions and conserving full S-LPS structure (tularin+) were shown to be inductors of delayed-type hypersensitivity reaction. Alterations in LPS structure (tularin) results in a significant decrease, and denaturation of bacterial proteins (during boiling) results in a complete loss of immune stimulating properties of the preparations. Purified LPS preparations and O-polysaccharide fraction of S-LPS are not able to activate cell-mediated immunity. Conclusion. The presence of LPS with the full structure affects the ability of antigenic preparations of F.tularensis to cause allergic reactions, and thus, form cell-mediated anti-tularemia immunity. LPS of F.tularensis can not be excluded as an adjuvant and provides the most effective presentation of epitopes of protein molecules for interaction with receptors of T-lymphocytes.

Tattoos have become increasingly popular worldwide making adverse effects from tattoos a growing concern. In our report, we present a 51-year-old man who developed an unusual allergic reaction to the red ink portions of his tattoos that coincided with the initiation of ledipasvir/sofosbuvir treatment for his hepatitis C. Clinical and histological features were consistent with a delayed-type hypersensitivity reaction to red ink.

Abstract The present research studied the anti-bacterial effect of silver-coated red soil nanoparticles on Gram-negative bacteria Escherichia coli (E. coli) from water. The effects of disinfectant concentration (0.02, 0.05 and 0.1 g/mL), contact time (10, 20 and 30 minutes) and bacteria number (102, 104 and 106 CFU/mL) have been also investigated. To obtain important factors, the interactions between factors and optimal experimental design in surface response method were used based on Box-Behnken design. According to the research findings, the system is efficient in eliminating E. coli. The results showed that E. coli elimination efficiency intensified through increasing the amount of disinfectant from 0.02 to 0.1 g/mL. Expanding contact time from 10 minutes to 30 minutes also heightened the E. coli elimination rate. R2 for E. coli elimination is 0.9956 indicating a good agreement between model experimental data and forecasting data.

In this study the status of milk and dairy production marketing in Samangan province is explained. Locking of ranchers accessing to a respondent dairy production market is the main problem focused herein. In Afghanistan illiteracy, traditional customs dominance and low economy level are the major gaps somewhat nationally remained unresolved among most of ranchers. The main objective of this study is assessing the dairy productions market situation to empower the economy level of farmers. A complex method was applied in this study including observation, interview, and questionnaire. Totally 117 questionnaires were handled. Results of analyzed collected data clarify that 90% of ranchers were illiterate, dairy cattle keeping is more than other milky animals because of its lower needs to silage and land. It has been concluded that farmers of Samangan province have adequate experience of keeping and nourishing the domestic animals in order to get a noticeable income. For getting the highest income and having modernized dairy production activities, from the viewpoint of authors, gradually researches accommodating are recommended and this research findings and conclusions will contribute better designing of the future research.

Though a few provisions for the victim of crimes were indirectly recognized since the nineteenth century, from 2000 onwards, legal entitlements for crime victims are realized in Bangladesh with a specific focus on women and children. So far, few analyses are found to be performed mainly by the legal experts, emphasizing legal rights and remedies with recommendations for legal reform. However, studies on the status of victims’ rights seem to be incomplete without considering administrative as well as social reality—dominated by colonial legacy and traditional practices—beyond the written clauses in the law books. This study is one of the pioneering attempts in Bangladesh to understand the status of crime victims against the backdrop of recent legal changes and to examine the argument whether the legal provisions itself are enough in providing victims with intended benefits without simultaneous social and administrative changes. Within the theoretical framework of balancing victim’s rights and informal social control (victim blaming), this qualitative study (through content analysis) reviewed all criminal laws and research findings related to victim’s rights within a socio-legal approach in terms of victim’s access, participation, protection, services and compensation. Along with the rights legally granted to victims, available research findings were interpreted in connection to those particular rights. It is found that there are unsupportive social milieu, administrative subculture and political practices, where victims of crime are strongly restrained from enjoying their rights. Particularly, the status of crime victims is found to be undermined in the face of corruption, low public confidence on enforcing agencies, gross withdrawal or discharge of criminal cases on political grounds, limited geographical coverage of victim support services and shelter homes, lengthy process for compensation and unavailability of rules or guidelines to enforce the rights.

Oxygen radicals play an important role in the mechanism of acute lung injury. The 21-aminosteroid lazaroid, U-78518F, is a potent antioxidant. We examined the effect of intravenous U-78518F on acute lung injury in septic guinea pigs over 8 h. The experimental groups (n = 6) were 1) saline control, 2) Escherichia coli (2 x 10(9)/kg i.v.), 3) pretreatment (U-78518F 5 mg/kg bolus + 1 mg.kg-1 x h-1, 15 min before E. coli injection), and 4) posttreatment (U-78518F 30 min after E. coli injection). We measured wet-to-dry weight ratio (W/D) as an index of pulmonary edema and concentration ratios of 125I-labeled albumin in lung tissue and bronchoalveolar lavage fluid compared with plasma (L/P and BAL/P, respectively) as indexes of lung protein fluxes. In septic guinea pigs, pretreatment with U-78518F attenuated W/D, L/P, and BAL/P and posttreatment attenuated W/D and BAL/P (P < 0.05 for each). Furthermore, we studied the effect of U-78518F on human neutrophil oxygen radical production (ORP) by using flow cytometry to assess intracellular ORP and lucigenin-dependent chemiluminescence to assess extracellular ORP. Neutrophils (5 x 10(5) were stimulated with 0.5 micrograms/ml of phorbol myristate acetate. With flow cytometry, we measured intracellular ORP, cross-sectional cell area, and degranulation in neutrophils. U-78518F (minimum concn 1.0 microM) decreased intracellular ORP (n = 4; P < 0.05) when the dihydrorhodamine 123 assay was used. U-78518F (minimum concn 1.0 microM) inhibited phorbol myristate acetate-induced neutrophil chemiluminescence (n = 4; P < 0.05).

Visualization is one of the main features of Building Information Modeling (BIM). It has many advantages throughout the lifecycle of a construction project, and it has become a timely research topic in recent years. However, no attempt has been made to give a holistic understanding of the existing BIM-based visualization research status. Thus, this study aims to conduct a scientometric analysis of the existing BIM-based visualization literature and to gain a snapshot of the research status during the period 2010–2019. A total of 255 articles were abstracted from the Scopus database. Then, the VOSviewer program was employed to visualize the research status from the perspectives of scholars, countries/regions, journals, articles, and co-occurring keywords. Results revealed that Wang X. published the greatest number of articles, while Cheng J.C.P. received the greatest average normalized citations. Furthermore, Automation in Construction was identified as the most influential journal and the article “Building information modeling (BIM): trends, benefits, risks, and challenges for the AEC industry” was the most cited paper. Through the keywords co-occurrence analysis, “virtual reality” and “visual programming language” were identified as the emerging themes in this field. The research findings can provide both researchers and practitioners with a better understanding of the status quo and trends of the BIM-based visualization research.
