Varietal reactions of Trifolium subterraneum L. to Phaseolus virus 2 Pierce.

A number of varieties of subterranean clover (Trifolium subterraneum L.) were tested with a severe strain of Phaseolus virus 2 Pierce. Northam First Early, Dwalganup, and Pink Flowered gave lethal necrotic reactions while the rest developed mottles and chlorosis of different degrees of intensity. Reductions in the mean fresh weights of plants of five subterranean clover varieties ranged from 26.1 to 76.7 per cent. Epidermal cells of mottled leaves contained irregular aggregates of viroplasts stained by treatment with phloxine-trypan blue. Viroplasts in young tip leaves did not stain if the plants were held at temperatures below 44.6°F while those in mature basal leaves were not so affected. The lethal-reactors Northam First Early, Dwalganup, and Pink Flowered were resistant in the field to Phaseolus virus 2. With hand inoculation in the glass-house an average of 10 per cent. of plants of these varieties remained free from systemic infection, whereas all the plants of mottle-reacting varieties became systemically infected. The virus had a discontinous distribution in plants of lethal-reactors, compared with a continuous distribution in those of mottle-reactors such as Mount Barker. The lethal reaction is heritable in a dominant fashion in most crosses, so that the development of desirable new virus- resistant varieties is possible.

In 1989 and 1990, infection with subterranean clover mottle sobemovirus (SCMV) was widespread in subterranean clover ( Trifolium subterraneum L.) pastures in the south-west of Western Australia. The virus was detected in 61% of the pastures sampled and incidences of infection ranged from 1 to 50%. The virus was more common in old pastures than in pastures resown with newer cultivars during the preceeding 5 year period. When 12 isolates of SCMV were inoculated to subterranean clover plants grown in the glasshouse, symptoms varied from mild to severe. SCMV isolates P23 and F4 decreased the herbage dry weight of cw. Daliak and Woogenellup grown in plots as spaced plants by 81-88% while the Type isolate caused losses of 92%. By contrast, losses were 37-49% with cv. Karridale, a cultivar in which systemic infection was either delayed or prevented during winter. Infection decreased seed yield by c. 90% in cvv. Karridale and Woogenellup with all three isolates; seed weight was decreased 21-55%. A small proportion of cv. Woogenellup transplants outgrew the infection in new shoots during late spring to produce abundant healthy foliage. SCMV seed transmission rates in seed collected from infected transplants of cv. Woogenellup were 0.06, 0.07 and 0.43% for the Type, P23 and F4 isolates respectively. It was concluded that SCMV was present in most pastures, but at low incidences, and that it persists in them from year to year. Extended growing seasons and hard grazing are likely to increase its incidence.

Over the period 1952-1954 there was no significant difference in the yield of herbage produced annually by the addition to a Phalaris tuberosa L.–Trifolium subterraneum L. pasture mixture, of any one of the following grasses: Agropyron obtusiusculum Lange., Bromus coloratus Steud., Bromus inermis Leyss., Dactylis glomerata L., or Festuca arundinacea Schreb. There were indications of a small change in seasonal production where certain grasses, notably D. glomerata, were included in the mixture. Over the 3-year period the proportion of the sown grass component, in the mixtures where either D. glomerata, B. inermis, or B. coloratus were included, increased by a greater amount than where the simple mixture of phalaris and subterranean clover was used. The addition of each grass also lessened the amount of P. tuberosa in the sown grass component of the yield. In the third year, despite the varying proportions of the phalaris and associated sown grass species, the mean population of the sown perennial grasses in each treatment did not differ significantly from the mean figure of 1.34 plants/sq. lk. The increased production of the sown grass yield component following the association of certain of the above species with P. tuberosa suggests that the latter does not fully exploit the environment. The principle of including another perennial grass when sowing phalaris and subterranean clover might have wide application as a means of combatting "phalaris staggers".

Responses of redlegged earth mite (Halotydeus destructor) to seedlings of three resistant and four susceptible varieties of subterranean clover (Trifolium subterraneum) were measured after 7 or 14 days in pot experiments in the glasshouse. With a single variety/pot, mites on resistant varieties (DGI007, EP145SubD and Rutherglen B) produced 45% of the progeny that were produced by mites on the susceptible varieties (89838G, Dalkeith, Junee and 70088B). Number of stages completed and survival were little affected by varieties. Feeding damage (silvering of cotyledons) on resistant varieties averaged 45% of that on susceptible varieties with a single varietylpot. H. destructor fed less on resistant varieties in choice than in single variety experiments. On Junee and 89838G seedlings, feeding damage was similar to that on other susceptible varieties, but there were about half as many H. destructor progeny as on Dalkeith and 70088B. Mites laid more eggs on soil away from Junee plants, compared to the other three susceptible varieties. Different factors adversely affected the number of progeny produced on resistant varieties and on Junee.

Studies have been made of the digestion and metabolism in Merino wethers of the isoflavones in subterranean clover (Trtfolium subterraneurn cv. Clare) and red clover (T. pratense). The dietary intake of isoflavones with both clovers was about 9 g per day. With the subterranean clover, the isoflavones were predominantly genistein and biochanin A, and slight teat length increases in the wethers ingesting this clover indicated a low level of oestrogenicity. With the red clover formononetin represented 60% of the isoflavone present and the wethers on this diet exhibited maximal teat length increases indicating a high level of oestrogenicity. Less than 1 % of the daily intake of the isoflavones was excreted as such in the faeces and urine; hence most of these compounds were metabolized or retained in the sheep. The dietary isoflavones were found to disappear rapidly from the rumen, and it was estimated by using marker techniques that the removal of these compounds from the stomach (rumen, reticulum, omasum, and abomasum) was virtually complete. Equol (7,4'-dihydroxyisoflavan), a metabolite of formononetin, was the predominant phyto-oestrogen in the digesta and excreta when red clover was given. The excretion of 3.9 g/day of this compound, mainly in urine, was equivalent to 70% of the intake of formononetin. It was calculated that about 86% of the equol produced in the rumen was absorbed from that organ; the mean residence time for equol in the rumen was estimated to be 1.7 hr. The isoflavones were present in blood plasma mainly in conjugated forms. Equol predominated with both clovers. The levels of equol were much lower with the subterranean clover than with the red clover diet; the concentrations of the conjugated form were respectively 13 and 300-440 �g/100 ml. Equol in the free form, although not detectable with the subterranean clover, was present at 4-10 �g/100 ml with red clover. The data were considered to be consistent with the conclusion that equol accounts for most of the phyto-oestrogenic activity in sheep fed on clovers containing high levels of formononetin.

Subterranean clover mottle sobemovirus (SCMoV) is an important pathogen of subterranean clover (Trifolium subterraneum) pastures in Australia. Cultivars of subterranean clover are susceptible (e.g. Dalkeith, Woogenellup), moderately susceptible (e.g. Green Range), partially resistant (e.g. Goulburn) or highly resistant (e.g. Meteora, Trikkala) to SCMoV. The resistant class does not become systemically infected on sap inoculation, while only a proportion of plants develop systemic infection with the moderately susceptible and partially resistant classes (Wroth and Jones 1992b; Ferris and Jones 1994). These different classes were used to examine the basis of SCMoV resistance using cell biological approaches. Virus levels were quantified at the tissue level by ELISA and at the cell level by counting protoplasts which fluoresced after antibody staining. Three aspects were investigated: (1) initial cell infection, (2) frequencies of cell infection in sap-inoculated leaves and in systemically infected leaves from graft-inoculated plants, and (3, ability of the virus to replicate in isolated protoplasts. Fewer and smaller starch lesions formed in inoculated leaves of a highly resistant cultivar than in those of moderately susceptible and susceptible cultivars. In protoplasts isolated from epidermal cell strips from inoculated leaves, initially the numbers of fluorescing protoplasts were the same for all four classes of cultivars (5-7%). Subsequently, however, the numbers remained unchanged for the two highly resistant cultivars, but increased considerably in the susceptible one used, and to a lesser extent in the moderately susceptible and partially resistant ones. For protoplasts isolated from whole inoculated leaves, the percentage of protoplasts that fluoresced was greatest in the susceptible cultivars used, lower in the moderately susceptible and partially resistant cultivars, and least in the highly resistant ones. All classes became systemically infected when graft-inoculated, but with the systemically infected leaves the proportions of protoplasts that fluoresced were least for the highly resistant and greatest for the susceptible classes. When protoplasts were inoculated with SCMoV in vitro, the proportions which fluoresced were the same (53-56%) for highly resistant, moderately susceptible and susceptible cultivars, showing that resistance was not expressed at the single cell level as an inhibition of virus replication. Because movement of SCMoV was restricted in inoculated leaves of the highly resistant class (starch lesion, epidermal strip and whole leaf data) and in graft-induced systemically infected leaves, it is concluded that the basis of resistance to SCMoV in highly resistant cultivars of subterranean clover is restriction of cell-to-cell movement of the virus.

Plants were grown in limed Muchea sand and the zinc response was measured as the difference in dry weight between plants with and without added zinc. The light intensities are given as the mean daily maximum intensities.

When fed to spayed guinea pigs under the conditions outlined in Part I (Alexander and Watson 1951), whole clover from 83 per cent. of all samples and lamina from all of the few samples of this fraction yielded values of the mean log uterine weight in decigrams (mean log u.dg.) above 0.75, and provided data from which relative oestrogenic activities could be calculated by application of the previously derived statistics. The activity of fresh samples of four varieties of whole clover rose to a maximum in the mid-winter months, then decreased as the season progressed. However, both the absolute level and the magnitude and time relations of the changes varied with the different materials. Samples of lamina possessed high activity, while samples of petiole and stem exhibited little or no activity. Both the lamina and whole clover fractions of samples from grazed areas exhibited higher activity than corresponding material from adjacent ungrazed areas. Dehydration of clover reduced its activity. Little further change in activity occurred during storage of dried material over a period of 17 months. No demonstrable activity was present in the few species of plants other than subterranean clover which were tested.

Seedlings of 3 perennial grasses, Danthonia linkii Kunthcv. Bunderra, D. richardsonii Cashmore cv. Taranna(wallaby grasses), and Phalaris aquatica L. cv. Sirosa,were each grown in replacement series mixtures with seedlings ofTrifolium repens L. (white clover),Trifolium subterraneum L. var. brachycalycinum (Katzn.et Morley) Zorahy & Heller cv. Clare (subterraneanclover), and Lolium rigidum L. (annual ryegrass). Plantswere sown 5 cm apart in boxes (45 by 29 by 20 cm) at a density of 307plants/m2. Maximum likelihood estimates were usedto derive parameters of a non-linear competition model using the dry matterweights of perennial grasses and competitors at 3 harvests, approximately 168,216, and 271 days after sowing. Intra-plant competition was examined inmonocultures of each species, grown at plant spacings of 2, 5, and 8 cm apartwith plants harvested at the above times.Competition occurred in all perennial grass–competitor mixtures, exceptin those of each perennial grass with white clover and thephalaris–subterranean clover mixture (Harvest 1) and those withD. richardsonii and phalaris grown with white clover(Harvest 2). For D. richardsonii (Harvests 1 and 2) andD. linkii (Harvest 1 only) grown with white clover andthe phalaris–subterranean clover (Harvest 1), the two species in themixture were not competing. In the phalaris–white clover mixture, eachspecies was equally competitive (Harvests 1 and 2). These differences incompetition and aggressiveness reflected differences in individual plantweights in monocultures where there was an effect (P < 0.05) of species ondry matter weight per box, but no significant effect of plant spacing.These data indicated that for successful establishment,D. richardsonii and D. linkiishould not be sown in swards with either subterranean clover or white clover,or where populations of annual ryegrass seedlings are likely to be high.Phalaris was more compatible with both white clover and subterranean clover,but aggressively competed with by annual ryegrass.

AbstractThis study investigated the ability of neonatal larvae of the root-feeding weevil, Sitona lepidus Gyllenhal, to locate white clover Trifolium repens L. (Fabaceae) roots growing in soil and to distinguish them from the roots of other species of clover and a co-occurring grass species. Choice experiments used a combination of invasive techniques and the novel technique of high resolution X-ray microtomography to non-invasively track larval movement in the soil towards plant roots. Burrowing distances towards roots of different plant species were also examined. Newly hatched S. lepidus recognized T. repens roots and moved preferentially towards them when given a choice of roots of subterranean clover, Trifolium subterraneum L. (Fabaceae), strawberry clover Trifolium fragiferum L. (Fabaceae), or perennial ryegrass Lolium perenneL. (Poaceae). Larvae recognized T. repens roots, whether released in groups of five or singly, when released 25 mm (meso-scale recognition) or 60 mm (macro-scale recognition) away from plant roots. There was no statistically significant difference in movement rates of larvae.

Changes in the pre-emergence distribution of dry matter in subterranean clover (Trifolium subterraneum L.) variety Bacchus Marsh were followed at 21°C, using three sizes of seed and three depths of sowing, ½, 1¼, and 2 in. Decreasing seed size and increasing depth of sowing both reduce the weight of the cotyledons a t emergence. Seed of the three sizes were sown a t three depths in pot culture a t staggered intervals so that emergence was simultaneous. Dry weight in the early vegetative stage was proportional to seed size, and total leaf area and leaf numbers showed similar trends. Plants of each seed size grew at the same relative rate. No effect of depth of sowing could be detected, and this was shown to be due to the cotyledon area a t emergence being constant for any given seed size, regardless of varying depth of sowing and hence of cotyledon weight. It was concluded that seed size in a plant having epigeal germination and without endosperm is of importance: firstly, in limiting the maximum hypocotyl elongation and hence depth of sowing, and secondly, in determining cotyledon area. Cotyledon area in turn influences seedling growth, which is not affected by cotyledon weight. Once emergence has taken place, cotyledonary reserves are of no further significance in the growth of the plants.
