AbstractIn this Research Communication we present a study of the effect of Ca-binding salts on the recovery of milk fat globule membrane (MFGM) from buttermilk. Sodium phosphate buffer was used for the purpose of MFGM recovery from buttermilk for the first time and we showed that 0.1 M buffer at pH 7.2 was the most effective for the recovery of MFGM. The fact of high efficacy of sodium phosphate buffer in recovery of MFGM from buttermilk allowed us to suggest that MFGM in buttermilk is present in association with casein through Ca- bridges formed between phospholipids of MFGM and phosphate groups of casein, primarily with k-casein as the peripheral protein of casein micelles.

Polyvinyl alcohol (PVA) and pectin blends were used to entrapα-amylase (Termamyl) using glutaraldehyde as a cross-linker. The effect of glutaraldehyde concentration (0.25, 0.5, 0.75, 1.0, and 1.25%) on the activity of the immobilized enzyme and rate of enzyme released was tested during a 24 h period. Characteristics of the material, such as scanning electron microscopy (SEM), tensile strength (TS), elongation, and rate of dissolution in water (pH 5.7), ruminal buffering solution (pH 7.0), and reactor containing 0.1 mol L−1sodium phosphate buffer (pH 6.5), were also analyzed. SEM results showed that the surfaces of the pectin/PVA/amylase films were highly irregular and rough. TS values increased as a function of glutaraldehyde concentration, whereas percentage of elongation (%E) decreased. Pectin/PVA/amylase films presented similar values of solubility in the tested solvents. The material obtained with 0.25% glutaraldehyde performed best with repeated use (active for 24 h), in a phosphate buffer reactor. By contrast, the material obtained with 1.25% glutaraldehyde presented higher performance duringin vitrotesting using an artificial rumen. The results suggest that pectin/PVA/amylase is a highly promising material for biotechnological applications.

A study is reported of the effect of different methods of treatment on the toxemia of cardiac obstruction. The average duration of life of untreated dogs is 3 days. Three dogs treated with 1 per cent salt solution subcutaneously lived 32, 36, and 45 days respectively without developing a toxemia. 2 per cent glucose similarly given, does not alter the course of the toxemia. Concentrated salt solution in small quantities given directly into the jejunum prevents the marked rise in non-protein nitrogen but does not materially prolong life. Release of the obstruction does not change the course of the toxemia in untreated animals. The subcutaneous injection of 1 per cent sodium chloride solution after release of the obstruction causes a rapid return of the blood to normal and allows the animal to recover. A similar amount of fluid given as 2 per cent glucose does not alter the course of the toxemia after release of the obstruction.

Hepatitis B surface antigen was concentrated and purified from plasma by two simple steps of purification. In the first step the antigen was purified 24-fold by polyethylene glycol precipitation. An additional 10-fold purification was achieved by batchwise adsorption to hydroxylapatite and subsequent elution with 0.02 M sodium phosphate buffer.
