Bread wheat (Triticum aestivum L.) is one of the most important food crops, however it is only moderately tolerant to salinity stress. To improve wheat yield under saline conditions, breeding for improved salinity tolerance of wheat is needed. We have identified nine quantitative trail loci (QTL) for different salt tolerance sub-traits in a recombinant inbred line (RIL) population, derived from the bi-parental cross of Excalibur × Kukri. This population was screened for salinity tolerance subtraits using a combination of both destructive and non-destructive phenotyping. Genotyping by sequencing (GBS) was used to construct a high-density genetic linkage map, consisting of 3236 markers, and utilised for mapping QTL. Of the nine mapped QTL, six were detected under salt stress, including QTL for maintenance of shoot growth under salinity (QG(1-5).asl-5A, QG(1-5).asl-7B) sodium accumulation (QNa.asl-2A), chloride accumulation (QCl.asl-2A, QCl.asl-3A) and potassium:sodium ratio (QK:Na.asl-2DS2). Potential candidate genes within these QTL intervals were shortlisted using bioinformatics tools. These findings are expected to facilitate the breeding of new salt tolerant wheat cultivars.

Drought and heat factors have negative impacts on wheat yield and growth worldwide. Improving wheat tolerance to heat and drought stress is of the utmost importance to maintain crop yield. WRKY transcription factors help improve plant resistance to environmental factors. In this investigation, Arabidopsis WRKY30 (AtWRKY30) transcription factor was cloned and expressed in wheat. Plants growth, biomass, gas-exchange attributes, chlorophyll content, relative water content, prolines content, soluble proteins content, soluble sugars content, and antioxidant enzymes activities (catalase (CAT), superoxide dismutase (SOD), peroxidase (POX), and ascorbate peroxidase (APX)) of the AtWRKY30-overexpressing wheat plants were higher than those of the wild type. However, levels of electrolyte leakage, malondialdehyde, and hydrogen peroxide of the AtWRKY30-overexpressing wheat plants were significantly less than those of the wild-type. Additionally, the expression level of antioxidant enzyme-encoding genes and stress-responsive genes (ERF5a, DREB1, DREB3, WRKY19, TIP2, and AQP7) were significantly induced in the transgenic wheat plants in comparison with the wild type. In conclusion, the results demonstrated that AtWRKY30 overexpression promotes heat and drought tolerance in wheat by inducing gas-exchange attributes, antioxidant machinery, osmolytes biosynthesis, and stress-related gene expression. AtWRKY30 could serve as a potential candidate gene for improving stress tolerance in wheat.

Evaluation of turfgrass salt tolerance is a basic strategy for selecting grasses that can be grown in areas with salt-affected water or soils. Our objectives were to determine the relative salinity tolerances of 32 grasses and to evaluate potential shoot-based criteria for assessing salinity tolerance. Shoot growth responses to salinity of 28 seashore paspalums (Paspalum vaginatum Swartz) and four bermudagrass [Cynodon dactylon (L.) × C. transvalensis Burtt-Davy] cultivars were investigated under solution/sand culture in a greenhouse. Turfgrasses were grown in a sea-salt amended nutrient solution. Salinity ranges were 1.1 to 41.1 dS·m-1 based on electrical conductivity of the solution (ECw). Selection criteria to assess salt tolerance were absolute growth at 1.1 (ECw0), 24.8 (ECw24), 33.1 (ECw32), and 41.1 dS·m-1 (ECw40); threshold ECw; ECw for 25% and 50% growth reduction based on ECw0 growth; and leaf firing (LF) at ECw0 and ECw40 (LF0 and LF40, respectively). Significant variations among 32 entries were observed for all shoot responses except threshold ECw. Ranges of values for shoot parameters were: inherent growth at ECw0 = 0.10 to 0.98 g dry weight (10-fold difference); growth at 24.8 dS·m-1 = 0.11 to 0.64 g; growth at 33.1 dS·m-1 = 0.09 to 0.54 g; growth at 41.4 dS·m-1 = 0.06 to 0.35 g; threshold ECW = 3.9 to 12.3 dS·m-1; ECw25 % = 14 to 38 dS·m-1; ECw50% = 22 to 43 dS·m-1; and LF40 = 7% to 41%. Results in this study indicated substantial genetic-based variation in salt tolerance within seashore paspalums. When evaluation of salt tolerance based on shoot responses is attempted at wide salinity levels up to 40 dS·m-1, all seven criteria exhibiting a significant F test can be used. Five entries (SI 92, SI 93-1, SI 91, SI 93-2, SI 89) were ranked in the top statistical grouping for all seven-growth parameters, followed by SI 90 ranked in six out of seven, and three paspalums (SI 94-1, `Sea Isle 1', and `Taliaferro') were ranked in five out of seven categories.

A cross was made between high yielding salt susceptible BINA variety (Binadhan-5) with salt tolerant rice landrace (Harkuch) to identify salt tolerant rice lines. Thirty six F3 rice lines of Binadhan-5 x Harkuch were tested for salinity tolerance at the seedling stage in hydroponic system using nutrient solution. In F3 population, six lines were found as salt tolerant and 10 lines were moderately tolerant based on phenotypic screening at the seedling stage. Twelve SSR markers were used for parental survey and among them three polymorphic SSR markers viz., OSR34, RM443 and RM169 were selected to evaluate 26 F3 rice lines for salt tolerance. With respect to marker OSR34, 15 lines were identified as salt tolerant, 9 lines were susceptible and 2 lines were heterozygous. While RM443 identified 3 tolerant, 14 susceptible and 9 heterozygous rice lines. Eight tolerant, 11 susceptible and 7 heterozygous lines were identified with the marker RM169. Thus the tested markers could be efficiently used for tagging salt tolerant genes in marker-assisted breeding programme.DOI: http://dx.doi.org/10.3329/pa.v19i2.16929 Progress. Soil salinity is one of the main factors limiting cereal productivity in worldwide agriculture. Exploitation of natural variation in local barley germplasm is an effective approach to overcome yield losses. Three gene pools of North African Hordeum vulgare L. grown in Tunisia, Algeria and Egypt were evaluated at the reproductive stage under control and saline conditions. Assessment of stress tolerance was monitored using morphological, yield-related traits and phenological parameters of reproductive organs showing significant genetic variation. High heritability and positive relationships were found suggesting that some traits associated with salt tolerance could be used as selection criteria. The phenotypic correlations revealed that vegetative traits including shoot biomass, tiller number and leaf number along with yield-related traits such as spike number, one spike dry weight, grain number/plant and grain number/spike were highly positively correlated with grain yield under saline conditions. Hence, these traits can be used as reliable selection criteria to improve barley grain yield. Keeping a higher shoot biomass and longer heading and maturity periods as well as privileged filling ability might contribute to higher grain production in barley and thus could be potential target traits in barley crop breeding toward improvement of salinity tolerance. Multiple selection indices revealed that salt tolerance trait index provided a better discrimination of barley landraces allowing selection of highly salt-tolerant and highly productive genotypes under severe salinity level. Effective evaluation of salt tolerance requires an integration of selection indices to successfully identify and characterize salt tolerant lines required for valuable exploitation in the management of salt-affected areas.

Abstract Polyploidy occurs prevalently and plays an important role during plant speciation and evolution. This phenomenon suggests polyploidy could develop novel features that enable them to adapt wider range of environmental conditions compared with diploid progenitors. Bread wheat (Triticum aestivum L., BBAADD) is a typical allohexaploid species and generally exhibits greater salt tolerance than its tetraploid wheat progenitor (BBAA). However, little is known about the underlying molecular basis and the regulatory pathway of this trait. Here, we show that the histone acetyltransferase TaHAG1 acts as a crucial regulator to strengthen salt tolerance of hexaploid wheat. Salinity-induced TaHAG1 expression was associated with tolerance variation in polyploidy wheat. Overexpression, silencing and CRISPR-mediated knockout of TaHAG1 validated the role of TaHAG1 in salinity tolerance of wheat. TaHAG1 contributed to salt tolerance by modulating ROS production and signal specificity. Moreover, TaHAG1 directly targeted a subset of genes that are responsible for hydrogen peroxide production, and enrichment of TaHAG1 triggered increased H3 acetylation and transcriptional upregulation of these loci under salt stress. In addition, we found the salinity-induced TaHAG1-mediated ROS production pathway is involved in salt tolerance difference of wheat accessions with varying ploidy. Our findings provide insight into the molecular mechanism of how an epigenetic regulatory factor facilitates adaptability of polyploidy wheat and highlights this epigenetic modulator as a strategy for salt tolerance breeding in bread wheat.

Agronomic practices, such as planting date, seeding rate, and genotype, commonly influence hard red spring wheat (HRSW, Triticum aestivum L. emend. Thell.) production. Determining the agronomic optimum seeding rate (AOSR) of newly developed hybrids is needed as they respond to seeding rates differently from inbred cultivars. The objectives of this research were to determine the AOSR of new HRSW hybrids, how seeding rate alters their various yield components, and whether hybrids offer increased end-use quality, compared to conventional cultivars. The performance of two cultivars (inbreds) and five hybrids was evaluated in nine North Dakota environments at five seeding rates in 2019−2020. Responses to seeding rate for yield and protein yield differed among the genotypes. The AOSR ranged from 3.60 to 5.19 million seeds ha−1 and 2.22 to 3.89 million seeds ha−1 for yield and protein yield, respectively. The average AOSR for yield for the hybrids was similar to that of conventional cultivars. However, the maximum protein yield of the hybrids was achieved at 0.50 million seeds ha−1 less than that of the cultivars tested. The yield component that explained the greatest proportion of differences in yield as seeding rates varied was kernels spike−1 (r = 0.17 to 0.43). The end-use quality of the hybrids tested was not superior to that of the conventional cultivars, indicating that yield will likely be the determinant of the economic feasibility of any future released hybrids.

Salinity is one of the most significant environmental stresses for sustainable crop production in major arable lands of the globe. Thus, we conducted experiments with 27 tomato genotypes to screen for salinity tolerance at seedling stage, which were treated with non-salinized (S1) control (18.2 mM NaCl) and salinized (S2) (200 mM NaCl) irrigation water. In all genotypes, the elevated salinity treatment contributed to a major depression in morphological and physiological characteristics; however, a smaller decrease was found in certain tolerant genotypes. Principal component analyses (PCA) and clustering with percentage reduction in growth parameters and different salt tolerance indices classified the tomato accessions into five key clusters. In particular, the tolerant genotypes were assembled into one cluster. The growth and tolerance indices PCA also showed the order of salt-tolerance of the studied genotypes, where Saniora was the most tolerant genotype and P.Guyu was the most susceptible genotype. To investigate the possible biochemical basis for salt stress tolerance, we further characterized six tomato genotypes with varying levels of salinity tolerance. A higher increase in proline content, and antioxidants activities were observed for the salt-tolerant genotypes in comparison to the susceptible genotypes. Salt-tolerant genotypes identified in this work herald a promising source in the tomato improvement program or for grafting as scions with improved salinity tolerance in tomato.

We investigated genetic differences in salinity tolerance among 20 saltgrass (Distichlis spicata (L.) Greene) genotypes, including constitutive, gender-based and phenotypic plasticity traits, to better understand the basis of adaptation and acclimation by saltgrass in diverse environments. On average, the plants survived NaCl treatments up to ~1 M, with reductions in growth and health that varied with genotype. For these 20 genotypes in a greenhouse study, we showed that greater plasticity in one salt tolerance mechanism was physiologically linked to lesser plasticity in another. Under various levels of constant salinity stress, genotypes employing a strategy of greater plasticity in foliar Na and lesser plasticity in both foliar K : Na and Na turnover rate were better able to substitute Na for K in some cellular functions, especially osmotic adjustment, leading to increased salinity tolerance. Although we observed gender segregation with salinity in the Owens (Dry) Lake Playa (Inyo County, CA, USA) population planted for dust control, from which the genotypes were collected, we did not observe gender differences in salinity tolerance in the greenhouse. Significant physiological plasticity tradeoffs among genotypes, however, did affect overall salinity tolerance and may be important for this species survival in diverse managed and natural habitats.

The callus-promoting activity of most commonly known as well as some rarely tested auxins was compared with that of 2,4-dichlorophenoxyacetic acid (2,4-D) for in vitro culture of the excised embryo of spring and winter wheat (Triticum aestivum L.), cv. Chinese Spring and cv. Fredrick. Different auxins in a concentration range from 1 to 50 μM showed widely different activities. Also the two wheat cultivars responded differently to the auxins. When rapid callus formation with limited root growth was used as the basis for comparison, 2-(2-methyl-4-chlorophenoxy)propionic acid (2-MCPP), α-naphthaleneacetic acid, 3,6-dichloro-2-methoxybenzoic acid (dicamba), 4-amino-3,5,6,trichloropicolinic acid (picloram), γ-(2,4-dichlorophenoxy)butyric acid, 2,4,5-trichlorophenoxyacetic acid, and 2,4,5-trichlorophenoxypropionic acid, in the order of effectiveness, were superior to 2,4,-D for callus induction from the embryo of 'Chinese Spring,' although the concentration required was higher than that of 2,4-D. For the winter wheat 'Fredrick,' however, only picloram, dicamba, and 2-MCPP performed as well as 2,4-D. All auxins tested promoted shoot growth; 2-methyl-4-chlorophenoxypropionic acid was most effective for 'Chinese Spring,' whereas picloram was most effective for 'Fredrick.'
