ABSTRACTBackgroundRecurring damage to aerial organs of plants necessitates their prompt repair, particularly their vasculature. While vascular regeneration assay in aerial plant parts such as stem and inflorescence stalk are well established, those on leaf vasculature remained unexplored. Recently we established a new vascular regeneration assay in growing leaf and discovered the underlying molecular mechanism.ResultsHere we describe the detailed stepwise method of incision and the regeneration assay used for studying the leaf vascular regeneration. By using a combination of micro-surgical perturbations, brightfield microscopy and other experimental approaches, our new findings show that the regeneration efficiency decreases with aging of the leaf, and increases with the nearness of the wound towards the proximal end of the leaf.ConclusionThis easy-to-master vascular regeneration assay is an efficient and rapid method to study the mechanism of vascular regeneration in growing leaves. It can be readily adapted for other plant species and can be combined with cellular and molecular biology techniques.

This paper aims to identify the botanical origin of pollen loads collected by Apis mellifera L. in Canavieiras municipality, Bahia state. It provides a list of polliniferous plant species from the Atlantic Forest biome that are important for the development of regional apiculture. Using the acetolysis method, 35 bee-pollen samples were analyzed qualitatively and quantitatively. Results showed that pollen types Elaeis (23.99%), Mimosa pudica (22.78%) and Cecropia (13.68%) were the most abundant among the samples. These also showed the highest relative frequencies of the material studied and were important pollen sources for bees in the study area.

The purpose of this study is to characterize native bee plants regarding their capacity to extract and accumulate trace elements from the soil and its consequences to the sanity of the produced pollen. The trace elements Cu, Mn, Pb and Zn were analyzed in soil, plants and bee pollen from Teresina region (PI), Brazil, by flame atomic absorption spectrophotometer. Considering the studied plant species, Cu and Pb metals presented in the highest levels in the roots of B. platypetala with 47.35 and 32.71 μg.mL-1 and H. suaveolens with 39.69 and 17.06 μg.mL-1, respectively, while in the aerial parts Mn and Zn metals presented the highest levels in S. verticillata with 199.18 and 85.73 μg.mL-1. In the pollen, the levels of Cu, Mn, Pb and Zn vary from 5.44 to 11.75 μg.mL-1; 34.31 to 85.75 μg.mL-1; 13.98 to 18.19 μg.mL-1 and 50.19 to 90.35 μg.mL-1, respectively. These results indicate that in the apicultural pasture the translocation (from soil to pollen) of Mn and Zn was more effective than in case of Cu and Pb, therefore, the bee pollen can be used as food supplement without causing risks to human health.

The flora of mountain formations in the Caatinga biome is composed predominantly by semi-deciduous species with representatives of both Atlantic and Amazon forest. Information on the potential for bee pollen production of these species is limited. In this study we evaluated the potential of production, the temporal variation, the botanical origin and the nutritional value of bee pollen produced in a seasonal semideciduous forest in northeastern Brazil. We identified a total of 252 flowering plant species throughout the year. The diet of Apis mellifera consisted of 74 pollen types distributed in 58 genera and 27 families. We identified two production peaks of bee pollen, the highest occurring in the rainy season. Nutritional value considering crude protein, carbohydrates, lipids and mineral matter changed over the study period, with influence of rainfall on the dry matter level. Some taxonomic groups of plants showed a strong relationship with nutrients, suggesting that although the diet of A. mellifera is broadly diversified, this species devoted most of its pollen foraging effort on the genus Mimosa and the palm tree species of Attalea speciosa. The results show that the seasonal semideciduous forest of the mountain range in the Northeast Brazil presents plant species: Mimosa caesalpiniifolia, Baccharis trinervis, Mimosa tenuiflora Myracrodruom urundeuva, Cecropia pachystachya, Attalea speciosa, with high nutritional level and potential for the pollen production.

The study of preferred crystallographic orientation (texture) in ceramics is assuming greater importance as their anisotropic crystal properties are being used to advantage in an increasing number of applications. The quantification of texture by a reliable and rapid method is required. Analysis of backscattered electron Kikuchi patterns (BEKPs) can be used to provide the crystallographic orientation of as many grains as time and resources allow. The technique is relatively slow, particularly for noncubic materials, but the data are more accurate than any comparable technique when a sufficient number of grains are analyzed. Thus, BEKP is well-suited as a verification method for data obtained in faster ways, such as x-ray or neutron diffraction. We have compared texture data obtained using BEKP, x-ray diffraction and neutron diffraction. Alumina specimens displaying differing levels of axisymmetric (0001) texture normal to the specimen surface were investigated.BEKP patterns were obtained from about a hundred grains selected at random in each specimen.
