An investigation was conducted to study the beneficial effects of Ephedra alata monjauzeana crude extract (EamCE). The chemical profile was determined using RP-HPLC–ESI-QTOF-MS analysis, revealing the presence of twenty-one flavonoids and phenolic acids. A series of antioxidant assays was carried out using ten different methods. The EamCE has demonstrated a significant antioxidant potential, with interesting IC50 values not exceeding 40 µg/mL in almost activities. Likewise, a significant inhibition of key enzymes, involved in some health issues, such as Alzheimer’s disease, diabetes, hyperpigmentation, dermatological disorders, gastric/urinary bacterial infections, and obesity, was observed for the first time. The IC50 values ranged from 22.46 to 54.93. The anti-inflammatory and non-cytotoxic activities were assessed by heat-induced hemolysis and cell culture methods, respectively; the EamCE has shown a prominent effect in both tests, notably for the anti-inflammatory effect that was superior to the reference compound “diclofenac” (IC50: 71.03 ± 1.38 > 70.23 ± 0.99 (µg/mL)). According to these results, this plant could be used in a large spectrum as a food supplement, as a natural remedy for various physiological disorders and pathologies; and it might serve as a preventive and health care agent.

Obesity is a worldwide epidemic with severe metabolic consequences. Polyphenols are secondary metabolites in plants and the most abundant dietary antioxidants, which possess a wide range of health effects. The most relevant food sources are fruit and vegetables, red wine, black and green tea, coffee, virgin olive oil, and chocolate, as well as nuts, seeds, herbs, and spices. The aim of this work was to evaluate the ability of a pure, isolated polyphenol supplementation to counteract the pernicious metabolic effects of a high-fat diet (HFD). Our results indicated that the administration of pure, isolated polyphenols under HFD conditions for 26 weeks worsened the glucose metabolism in diet-induced obese mice. The data showed that the main target organ for these undesirable effects were the kidneys, where we observed fibrotic, oxidative, and kidney-disease markers. This work led us to conclude that the administration of pure polyphenols as a food supplement would not be advisable. Instead, the ingestion of complete “whole” foods would be the best way to get the health effects of bioactive compounds such as polyphenols.

Tight junctions (TJ) are formed by transmembrane and intracellular proteins that seal the intercellular space and control selective permeability of epithelia. Integrity of the epithelial barrier is central to tissue homeostasis and barrier dysfunction has been linked to many pathological conditions. TJ support the maintenance of cell polarity through interactions with the Par complex (Cdc42-Par-6-Par-3-aPKC) in which Par-6 is an adaptor and links the proteins of the complex together. Studies have shown that Par-6 overexpression delays the assembly of TJ proteins suggesting that Par-6 negatively regulates TJ assembly. Because restoring barrier integrity is of key therapeutic and prophylactic value, we focus on finding compounds that have epithelial barrier reinforcement properties; we developed a screening platform (theLiTE™) to identify compounds that modulate Par-6 expression in follicular epithelial cells from Par-6-GFP Drosophila melanogaster egg chambers. Hits identified were then tested whether they improve epithelial barrier function, using measurements of transepithelial electrical resistance (TEER) or dye efflux to evaluate paracellular permeability. We tested 2,400 compounds, found in total 10 hits. Here we present data on six of them: the first four hits allowed us to sequentially build confidence in theLiTE™ and two compounds that were shortlisted for further development (myricetin and quercetin). We selected quercetin due to its clinical and scientific validation as a compound that regulates TJ; food supplement formulated on the basis of this discovery is currently undergoing clinical evaluation in gastroesophageal reflux disease (GERD) sufferers.

The group of plants, apart from the forest trees, agricultural, horticultural crop and medicinal plants, certain weed plants also play important role to formulate the qualitative and quantitative food and health supplements. These plants may be very specific in producing certain compound to heal the specific ailment chronically or serve as a good nutritive food supplement. There are numbers of crop species have been fortified with multi-nutrients or with the particular nutrient supplements e.g. Rice, Wheat, Maize, Sweet Potato. Instead, weeds remained underutilized and un-exploited and also treated as unwanted species. The nutritive compounds available in these plants such as polyphenols and vitamins can be the extraction from different plant portion to formulate the fortified food or curing medicine. The weed species prevailing in the nature are one of the major sources of dietary and health supplements for humans and animals since past i.e. Amaranthus lividus, Chenopodium album and many more weed species. There is still much to explore the Indian ecologic components present in wild form of flora. Hence, this chapter will majorly contribute to highlight nutritive nutaceurtical and edible weeds prevailing in Indian ecosystems.

Merang Mushroom (Volvariella volvaceae) is one of the edible mushrooms. The nutritional content of merang mushrooms makes this mushroom has potential as a medicine and food supplement. Merang mushrooms are known to function as antioxidants, antidiabetic, antiviral, and can lower cholesterol. The purpose of the research is to find out the nutritional content of merang mushrooms that are cultivated in corncob media with the addition of rice bran. The analysis used was proximate analysis to determine moisture, ash, and fiber content using the AOAC method, fat content analysis using the Soxhlet method, protein content analysis using the Kjeldahl method, and carbohydrate Analysis (By difference). The results of the analysis were 2.17% ash content, 85.14% water content, 3.65% crude fiber content, 1.99% fat content, 2.7% crude protein content, and 8.0% carbohydrate content. Based on research, it can be said that mushroom has a good nutritional content that can provide health benefits.

Fucoidans from sea cucumber (SC-FUC) have been proven to alleviate insulin resistance in several species. However, there are few studies that clarify the relationship between their structure and bioactivity. The present study evaluated the influence of molecular weight (Mw), sulfation concentrations (Cs), and sulfation position on improving insulin resistance using SC-FUC. Results showed that fucoidans with lower Mw exerted stronger effects. Having a similar Mw, Acaudina molpadioides fucoidans (Am-FUC) with lower Cs and Holothuria tubulosa fucoidans with higher Cs showed similar activities. However, Isostichopus badionotus fucoidans (higher Cs) activity was superior to that of low-Mw Thelenota ananas fucoidans (Ta-LFUC, lower Cs). Eliminating the effects of Mw and Cs, the bioactivity of Am-FUC with sulfation at meta-fucose exceeded that of Ta-FUC with sulfation at ortho-position. Moreover, the effects of Pearsonothuria graeffei fucoidans with 4-O-sulfation were superior to those of Am-LFUC with 2-O-sulfation. These data indicate that low Mw, 4-O-sulfation, and sulfation at meta-fucose contributed considerably to insulin resistance alleviation by SC-FUC, which could accelerate the development of SC-FUC as a potential food supplement to alleviate insulin resistance.

Moringa oleifera (MO) is a multipurpose plant with a high polyphenol content, which is being increasingly consumed to lessen the risk of chronic metabolic diseases such as Type 2 diabetes; however, scientific evidence from clinical trials is scarce. A double-blind, randomized, placebo-controlled, parallel group intervention study with MO leaves as a food supplement was conducted in subjects with prediabetes. They consumed six daily capsules of MO dry leaf powder (2400 mg/day) (MO, n = 31) or placebo (PLC, n = 34) over 12 weeks. Glycemia, appetite-controlling hormones and gut microbiota composition were studied. ANCOVA with the fixed factor “treatment” and the basal value as covariate was used to compare the change score between the groups. The results showed significant differences between groups in the rate of change of fasting blood glucose (FBG) and glycated hemoglobin (HbA1c), which showed opposite directions during the intervention, decreasing in MO and increasing in PLC. No different change scores were found between the groups in microbiota, hepatic and renal function markers or the appetite-controlling hormones measured. In conclusion, MO supplementation resulted in favorable changes in glycaemia markers compared to placebo in the subjects with prediabetes studied, suggesting that MO might act as a natural antihyperglycemic agent.

Aim: Spirulina is a microalga that is widely used as a food supplement and is regarded as having performance enhancing and health promoting properties. We conducted a preliminary evaluation of the possible antidepressant, anti-anxiety, pro-socialization and cognition-enhancing effects of Spirulina in mouse models Methods: Sixty male BalbC mice aged 3 weeks were administered phycocyanin-rich Spirulina extract [PRSE, 545 mg/kg], fluoxetine [20 mg/kg] or water orally for 5 weeks. During the last 2 weeks of the experiment a series of behavioral-cognitive tests was performed to evaluate motor activity, antidepressant and anti-anxiety effects, socialization and cognitive effects. Effects of PRSE and fluoxetine were compared to those of water. Results: There was a significant effect of PRSE in the activity domain, manifesting as an increase in velocity in the open field [p=0.0007 vs. water]. Fluoxetine significantly enhanced immobility in the tail suspension test and the forced swim test reflecting the known antidepressant effect of this compound, but not PRSE. There were no significant effects of PRSE in tests of anxiety, socialization or cognition. Conclusions: The most striking observation in this study was that PRSE significantly enhanced activity in the open field test. Further studies are indicated to confirm and extend this finding and investigate possible mechanisms of action. The results of the current study do not support sporadic reports of possible antidepressant or cognition-enhancing effects of PRSE. Nevertheless, additional studies are indicated using depression models rather than naïve mice, alternative mouse strains, using additional cognitive tests, and administering higher PRSE doses.

Abstract: Satureja genus belongs to the Lamiaceae family, they are used in food products and by pharmaceutical and cosmetic industries. The chemical composition of Satureja are responsible for its pharmacological and phytochemical properties. Among the various biological activities, notably antioxidant, antibacterial and antifungal, Satureja also has a potential antiviral activity. The existence of a microbiota modulation potential by Satureja in farming animals has also been reported. Viral pathologies are one of the main causes of disease in the world. It is commonly known that gut microbiota plays a crucial role in the fight and progression of viral infection. Previous studies conducted on coronavirus disease 2019 (COVID-19) pandemic have prouved an imbalance in the intestinal and pulmonary microbiota via gut-lung axis. Knowing the properties of Satureja and its traditional use, one can suggest the possibility of using it as dietary supplement to modulate immune system in order to prevent and fight viral infections. The objective of this review is to reveal the potential impact of medicinal plants such as Satureja genus as food supplement, on immune enhancing during SARS-CoV-2 infection and their relationships with the intestinal microbiota.
