AbstractIn this research, the effects of melatonin on germination and amino acid content in wheat (Triticum aestivum L.) seeds under polyethylene glycol (PEG-6000) stress were investigated. The results revealed that 10 μM melatonin could aggravate the adverse effects of drought stress on germination percentage, germination index, and germination potential of two wheat varieties (JM22 and HG35). However, 300 μM melatonin could obviously alleviate the negative effect of water stress on germination and increase radicle length, radicle number and plumule length of germinated seeds. Principal component analysis showed that amino acid content apparently changed in germination and the variation amplitude was different due to melatonin concentrations and wheat varieties. Lys content in wheat seed under 300 μM melatonin plus PEG treatment increased compared with that under PEG. Meanwhile, Lys content was significantly and positively correlated with germination percentage, germination index, germination potential, radicle length, and plumule length, respectively. Our results suggested Lys play an important role in the regulation of melatonin on drought resistance of wheat seed during germination. This may be of value for saving water resources in North China Plain.

We analysed the crude protein content, amino acid content, amino acid composition of four forage and milling III. quality winter wheat varieties (Magor, Hunor, Róna and Kondor) from their samples from five following years (2003, 2004, 2005, 2006, 2007). We found that quantity of essential and non-essential amino acids rose with increase in crude protein content. On examination of protein amino acid composition in relation to crude protein content we found that the crude protein content increased the quantities of the non-essential amino acids also rose, while those of the essential amino acids decreased as the lysine, the limiting amino acid of wheat. We also established that, as crude protein content increased, the biological value of the protein decreased.

As regards wheat varieties constituting a natural ploid series the issue of analysing diploid, tetraploid and hexaploid species is topical since ancient varieties can play significant roles in contemporary agriculture as well. Seventeen winter wheat varieties, out of which 2 diploid varieties carried genome A, 9 diploidic types had genomes AB, two varieties had genomes AG and four varieties were hexaploid ones with genomes ABD, were analysed from the point of view of their amino acid compositions. The amino acid contents of Asp, Thr, Ser, Glu, Gly, Ala, Cys, Val, Met, Ile, Leu, Tyr, Phe, His, Lys, Arg, Pro (a total of 17) were determined in the varieties listed above. It has been found that the amino acid contents of the grains genotype AA Triticum boeticum and T. monococcum exceeded the amino acid content of T. aestivum in respect of all the amino acids analysed in this experiment, with Glu being the only exception. In comparison with the aestivum wheat, essential amino acid contents showed a similarly favourable picture in the diploidic varieties mentioned. As regards type AB tetraploid varieties excesses of 13-16%, in comparison to the aestivum wheat, were found in essential amino acid contents. The amounts of non-essential amino acids in all the winter wheat varieties showed decreases irrespective of the ploid level. What concerns the total amino acid content, all the winter wheat varieties with the exception of T. monococcum (A), T. dicoccoides (AB) and T. dicoccum (AB) contained less amino acid than the aestivum wheat. All the monocarbonic acid and aromatic as well as heterocyclic amino acid contents of the wildly growing Triticum boeticum (A) and the grown Triticum monococcum (A) (with polaric, apolaric R groups, diamino radicles) exceeded the same contents of T. aestivum. The value of the monoamino-dicarbonic acid, however, was lower in our experiment.

AbstractWild soybean (Glycine soja) is a valuable genetic resource for soybean improvement. Seed composition profiles provide beneficial information for the effective conservation and utilization of wild soybeans. Therefore, this study aimed to assess the variation in free amino acid abundance in the seeds of wild soybean germplasm collected in Japan. The free amino acid content in the seeds from 316 accessions of wild soybean ranged from 0.965 to 5.987 mg/g seed dry weight (DW), representing a 6.2-fold difference. Three amino acids had the highest coefficient of variation (CV): asparagine (1.15), histidine (0.95) and glutamine (0.94). Arginine (0.775 mg/g DW) was the predominant amino acid in wild soybean seeds, whereas the least abundant seed amino acid was glutamine (0.008 mg/g DW). A correlation network revealed significant positive relationships among most amino acids. Wild soybean seeds from different regions of origin had significantly different levels of several amino acids. In addition, a significant correlation between latitude and longitude of the collection sites and the total free amino acid content of seeds was observed. Our study reports diverse phenotypic data on the free amino acid content in seeds of wild soybean resources collected from throughout Japan. This information will be useful in conservation programmes for Japanese wild soybean and for the selection of accessions with favourable characteristics in future legume crop improvement efforts.
