ABSTRACTBurkholderia pseudomalleiis a saprophytic bacterium that causes melioidosis and is often isolated from rice fields in Southeast Asia, where the infection incidence is high among rice field workers. The aim of this study was to investigate the relationship between this bacterium and rice through growth experiments where the effect of colonization of domestic rice (Oryza sativaL. cv Amaroo) roots byB. pseudomalleicould be observed. WhenB. pseudomalleiwas exposed to surface-sterilized seeds, the growth of both the root and the aerosphere was retarded compared to that in controls. The organism was found to localize in the root hairs and endodermis of the plant. A biofilm formed around the root and root structures that were colonized. Growth experiments with a wild rice species (Oryza meridionalis) produced similar retardation of growth, while another domestic cultivar (O. sativaL. cv Koshihikari) did not show retarded growth. Here we reportB. pseudomalleiinfection and inhibition ofO. sativaL. cv Amaroo, which might provide insights into plant interactions with this important human pathogen.

An endophytic actinomycete, strain R10-82T, isolated from surface-sterilized roots of rice (Oryza sativa L.) was studied using a polyphasic approach. Strain R10-82T produced branching substrate mycelia and developed spherical spore vesicles on aerial hyphae containing non-motile spores. The major cellular fatty acids were iso-C16 : 0, iso-C14 : 0 and 10-methyl C17 : 0. The predominant menaquinones were MK-9, MK-9(H2), MK-9(H4) and MK-9(H6). Rhamnose, ribose, madurose, mannose and glucose were detected in whole-cell hydrolysates. The diagnostic phospholipids were phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylinositol mannosides, hydroxylphosphatidylethanolamine and ninhydrin-positive phosphoglycolipids. These morphological and chemotaxonomic data were similar to those of the genus Sphaerisporangium . Analysis of the 16S rRNA gene sequence revealed that strain R10-82T was related most closely to Sphaerisporangium cinnabarinum JCM 3291T (98.3 % similarity). The DNA G+C content of strain R10-82T was 74 mol%. DNA–DNA relatedness data in combination with differences in the biochemical and physiological properties suggested that strain R10-82T should be classified as representing a novel species of the genus Sphaerisporangium , for which the name Sphaerisporangium rufum is proposed. The type strain is R10-82T ( = BCC 51287T = NBRC 109079T). An emended description of the genus Sphaerisporangium is also provided.

Morphophysiological response of the rice root (Oryza sativa L.) variety SFL 11 in the seedbed phase to the application of the native strain of Trichoderma sp. and leached from bovine vermicompost Ecuador ocupa el lugar 26 a nivel mundial en la producción de arroz con un consumo percápita de 48 kilogramos anuales. En el país, Manabí es la tercera provincia, con 8 710 hectáreas sembradas y rendimiento promedio de 5,58 tha-1. El sistema radicular constituye la primera línea de defensa de las plantas, indispensable para la productividad de los cultivos. En la rizosfera se encuentran microorganismos benéficos, uno de ellos es el hongo Trichoderma, cosmopolita en suelos gracias a su gran plasticidad ecológica y caracterizado, entre otras funciones, por su efecto estimulador en el crecimiento vegetal al colonizar raíces aumentando los pelos radiculares y la extensión en profundidad de enraizamiento. Por otra parte, los bioestimulantes a base de sustancias húmicas promocionan el crecimiento radicular. El experimento se realizó para evaluar si hay inducción a repuestas morfofisiológicas en las raíces por una cepa nativa de Trichoderma sp. y el lixiviado de vermicompost bovino en el arroz (Oryza sativa L.) variedad SFL 11 en fase de semillero. La investigación se condujo en diseño de bloques completamente al azar con 8 tratamientos, seis con Trichoderma sp., uno con lixiviado de vermicompost bovino y el testigo con agua y se realizó en la parroquia Charapotó del cantón Sucre en la provincia de Manabí. A los 25 días después de la siembra fue determinado a las raíces la longitud (cm), volumen (mL) y masa seca (g) a un total de 20 plantas de las cuatro réplicas del experimento y los resultados sometidos a análisis de varianza simple y las medias comparadas mediante prueba de Duncan al 5%. El análisis estadístico demostró efecto benéfico significativo de los bioestimulantes Trichoderma sp., y el lixiviado de vermicompost bovino en la longitud y el volumen radicular, mientras que la biomasa seca demostró diferencias numéricas en crecimiento de las plantas de arroz (Oryza sativa L.) variedad SFL 11 en fase de semillero en comparación al tratamiento testigo. Abstract Ecuador occupies the 26th place worldwide in the production of rice with a per capita consumption of 48 kilograms per year. In the country, Manabí is the third province, with 8 710 hectares planted and an average of 5.58 tha-1. The root system constitutes the first line of defense of the plants, necessary for the productivity of the crops. In the rhizosphere are specifically functional microorganisms, one of them is the fungus Trichoderma, cosmopolitan in soils thanks to its great ecological plasticity and characterized, among other functions, by its stimulating effect on plant growth by colonizing roots, increasing the root hairs and extension in depth of rooting. On the other hand, biostimulants based on vermicompost leachate promotes root growth. The experiment was carried out to evaluate if there is induction to morphophysiological responses in the roots by a native strain of Trichoderma sp. and the leaching of bovine vermicompost in rice (Oryza sativa L) variety SFL 11 in the seedling stage. The investigation was conducted in completely randomized block design with 8 treatments, six with Trichoderma sp., one with leachate of bovine vermicompost and the control with water and was carried out in the Charapoto parish of the Sucre canton in the province of Manabí. At 25 days after sowing the length (cm), volume (mL) and dry mass (g) were determined to the roots at a total of 20 plants of the four replicates of the experiment and the results subjected to simple variance analysis and the means compared by 5% Duncan test. Statistical analysis showed a significant beneficial effect of Trichoderma sp. Biostimulants, and leaching of bovine vermicompost in root length and volume, while dry biomass showed numerical differences in the growth of rice plants (Oryza sativa L) variety SFL 11 in the seedbed compared to the control treatment. The taxonomic position of three Lactobacillus -like micro-organisms (strains SG293T, SG296 and SG310) isolated from fermented rice grain (Oryza sativa L. subsp. japonica) in Japan was investigated. These heterofermentative lactic acid bacteria were Gram-stain-positive, rod-shaped, facultatively anaerobic, non-motile, non-spore-forming and did not show catalase activity. 16S rRNA gene sequence analysis of strain SG293T revealed that the type strains of Lactobacillus malefermentans (98.3 %), Lactobacillus odoratitofui (96.2 %), Lactobacillus similis (96.1 %), Lactobacillus kimchicus (96.1 %), Lactobacillus paracollinoides (95.9 %) and Lactobacillus collinoides (95.7 %) were the closest neighbours. Additional phylogenetic analysis on the basis of pheS and rpoA gene sequences, as well as biochemical and physiological characteristics, indicated that these three strains were members of the genus Lactobacillus and that the novel isolates had a unique taxonomic position. The predominant cellular fatty acids were C18 : 1ω9c and C19 : 1 cyclo 9,10. Because low DNA–DNA hybridization values among the isolates and Lactobacillus malefermentans JCM 12497T were observed, it is proposed that these unidentified isolates be classified as a novel species of the genus Lactobacillus , Lactobacillus oryzae sp. nov. The type strain is SG293T ( = JCM 18671T = DSM 26518T).

Keragaan morfologi dan kandungan antosianin padi beras merah (Oryza sativa L.) pada Kecamatan Munte dan Kecamatan Payung di Kabupaten Karo. Penelitian ini bertujuan untuk mendapatkan informasi morfologi dan kandungan antosianin padi beras merah (Oryza sativa L.). Penelitian ini dimulai dari Juni 2017dan selesai pada Januari 2018 di kecamatan Munte dan Kecamatan Payung Kabupaten Karo.Metode survei deskriptif menggunakan panduan International Rice Research Institute (IRRI). Teknik penentuan lokasi secara sengaja dan pengamblan sampel secara kebetulan.Uji kandungan antosianin menggunakan metode analitik dengan menghomogenkan sampel. Hasil eksplorasi didapatkan 72 genotipe yang dibagi menjadi tiga lokasi lahan. Berdasaran uji kandungan antosianin didapatkan lahan A memiliki kandungan tertinggi yaitu 0,5 mg/100 g dan terendah yaitu lahan B 0,08 mg/100 g.

Experiments were carried out to assess some insecticide modules against major insect pests of rice. Each module consists of a basal application of carbofuran 3G @ 1 kg a.i ha-1 at 20 DAT and Rynaxypyr 20 SC @ 30 g a.i ha-1 at 45 DAT except untreated control. All modules differ with each other only in third treatment which was applied in 65 DAT. The third treatment includes: Imidacloprid 17.8 SL @ 27 g a.i ha-1, Pymetrozine 50 WG @ 150 g a.i ha-1, Triflumezopyrim 106 SC @ 27 g a.i ha-1, Buprofezin 25 SC @ 250 g a.i ha-1; Glamore (Imidacloprid 40+Ethiprole 40% w/w) 80 WG @ 100 g a.i. ha-1, Thiacloprid 24 SC @ 60 g a.i ha-1, Azadirachtin 0.03 EC @ 8 g a.i ha-1, Dinotefuran 20 SG@ 40 g a.i ha-1 and untreated control. All the treated plots recorded significantly lower percent of dead heart, white ear- head caused by stem borer and silver shoot caused by gall midge. Module with Pymetrozine 50 WG @ 150 g a.i ha-1 treated plot recorded significantly higher per cent reduction of plant hoppers (>80% over untreated control) and produced higher grain yield (50.75 qha-1) than the other modules. Among the different treated modules the maximum number of spiders was found in Azadirachtin 0.03 EC @ 8 g a.i ha-1 treated module plot followed by other treatments.

Tujuan dari penelitian ini adalah untuk mengetahui pengaruh aplikasi abu sekam terhadap kandungan prolin dan silikat daun serta kualitas hasil yaitu protein dan amilosa biji padi gogo dan hubungan korelasi antar keempat komponen tersebut pada kondisi pertanaman 80 persen kapasitas lapang pada skala pot. Penelitian dilakukan di polibag dalam screen house Fakultas Pertanian Unsoed dengan menggunakan rancangan acak kelompok pola faktorial dengan faktor varietas (Situ patenggang, Limboto, Towuti, Batutegi dan Aek sibundong) dan faktor abu sekam (0, 2, 4, 6 t/ha), diulang tiga kali. Hasil penelitian menunjukan bahwa pemberian abu sekam dosis 2−6 t/ha mampu meningkatkan kandungan silikat daun antara 18,49−29,43% dan menurunkan kandungan amilosa biji pada lima varietas sekitar 4,19−6,92%. Pemberian abu sekam dosis 2−6 t/ha mampu meningkatkan kandungan prolin daun antara 27,56−70,63% dan protein biji antara 2,35−16,71%. Antarvarietas menunjukan bahwa kandungan prolin tertinggi dihasilkan oleh varietas Batu tegi 18,58 persen dan protein biji pada varietas Situ patenggang 9,55%. Terdapat korelasi antar karakter fisiologis yaitu antara silikat-prolin (0,62) dan kandungan protein-amilosa biji (-0,78).

Salah satu upaya untuk meningkatkan produksi dan mutu benih di lahan salin yaitu melalui penggunaan metode ratun dengan penambahan bakteri sintetik Synechoccocus sp. Penelitian ini bertujuan untuk mengetahui produksi dan mutu benih padi ratun yang tercekam salinitas dengan penambahan bakteri sintetik Synechoccocus sp. Penelitian dilaksanakan pada bulan Juli - Desember 2015 di Desa Suco, Kecamatan Mumbul Sari Jember dan Laboratorium Teknologi Benih Politeknik Negeri Jember. Rancangan percobaan yang digunakan adalah Rancangan Acak Kelompok Faktorial (RAK) dengan 2 faktor dan 3 ulangan. Faktor pertama adalah cekaman Salinitas (S) yang terdiri dari 5 taraf, S1 = tanpa cekaman (kontrol), S2 = cekaman salinitas 1000 ppm, S3 = cekaman salinitas 2000 ppm, S4 = cekaman salinitas 3000 ppm, S5 = cekaman salinitas 4000 ppm. Faktor kedua inokulasi bakteri sintetik Synechoccocus sp. (B), yang terdiri dari 2 taraf, B1 = tanpa inokulasi bakteri (kontrol), B2 = Inokulasi sintetik Synechoccocus sp. Data dianalisis menggunakan uji F (ANOVA) dan dilanjutkan dengan perhitungan Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukan perlakuan salinitas 1000 ppm (S2) menghasilkan jumlah anakan ratun produktif tertinggi yaitu 19,33 anakan. Cl- mempunyai fungsi utama dalam reaksi fotosintesis sehingga cekaman salintas pada perlakuan 1000 ppm (S2) dapat ditoleran oleh tanaman padi varietas Ciherang pada fase vegetatif. Pemberian Bakteri (B) Synechococcus sp. mampu menghasilkan tunas ratun tertinggi pada fase vegetatif yaitu 40,10 cm. Interaksi dari dua perlakuan menunjukan hasil yang nyata pada parameter jumlah gabah bernas yaitu cekaman salinitas 4000 ppm dengan inokulasi bakteri (B2S5) menghasilkan gabah bernas yang paling tinggi sebesar 99,06 butir. Inokulasi tanaman dengan Synechococcus sp. mampu meningkatkan kandungan nitrogen dan kandungan klorofil dalam jaringan tanaman. Interaksi antara cekaman salinitas 1000 ppm dengan inokulasi bakteri (B2S2) menghasilkan produksi per Ha yang paling tinggi yaitu 1,389 ton dan potensi produksi per Ha tertinggi yaitu 1.66 ton/ha.
