DISSOCIATION AND SUBUNIT SIZE OF β-LIPOVITELLIN

Dissociation of β-lipovitellin was detectable at pH 5.8 and increased linearly with pH. Light-scattering measurements at different pH values were consistent with a molecular weight of 2.27 × 105 for the subunit and twice this value for the associated form, confirming that it is a monomer–dimer system. This value for the molecular weight of the dimer is somewhat higher than previously reported (4.0 × 105) partly because a solvent displacement correction was used in estimating concentration by a dry weight method. Dissociation constants evaluated from light-scattering measurements and by ultracentrifugal separation were 22 × 10−6 and 29 × 10−6 respectively, in good agreement with provisional values already reported. Preliminary studies on the kinetics of this reaction indicate that, when the pH is altered, dissociation reaches the new equilibrium in about 0.5 minute but that reassociation requires about 50 minutes.

A water-soluble pigment excreted from Serratia marcescens has been purified by precipitation with ammonium sulphate, dialysis, and ultracentrifugation at different pH values. The purified pigment showed a single band in the ultracentrifuge and by electrophoretic analysis at several pH values. An average molecular weight of 5 × 106 was calculated from light-scattering measurements. This pigment is composed of carbohydrate and protein combined with prodigiosin, and several properties of the complex are described.

Light scattering measurements performed on DNA after irradiation in the presence of psoralen clearly show that inter strand cross linkings are present in the macromolecule. In fact after heat denaturation and successive cooling irradiated macromolecule shows a molecular weight practically unchanged while a DNA sample after the same treatment shows a molecular weight half of the intact native DNA. Also the general conformation of irradiated DNA undergoes practically to no modifications after the same heat treatment while native DNA shows itself to have been strongly modified. Moreover, on the basis of flow dichroism determinations, DNA cross-linked by psoralen after heat denaturation showed to be able to restore its ordered double helix structure, during the successive cooling.

The response of quasielastic light scattering to the polydispersity of scattering objects has been investigated. A new method of the polydispersity index determination has been suggested, suitable for the range 1.02 ⪬ Mw/Mn ⪬ 2.0 and consisting in the measurement of the dependence of the apparent decay time on the correlator sampling time. The polydispersity index can be determined by comparing these dependences with the theoretical ones obtained using correlation curves simulated for various values of the polydispersity index, assuming lognormal and Schulz-Zimm distributions of molecular weights. The test measurements on polystyrene standards having molecular weights in the range 9 103 – 20.6 106 give polydispersity index values Mw/Mn that are in a good agreement with those given by the manufacturer. The polydispersity index for polystyrene having the molecular weight Mw = 20.6 106 thus determined was Mw/Mn = 1.35.

The acid dissociation constants for alternariol monomethyl ether (AME), altertoxin-I (ATX-I), and alternariol (AOH), three of the Alternaria alternata genus mycotoxins, are estimated through conventional UV-VIS spectroscopic studies at different pH values. Experimental variations of absorbance as a function of pH at a given wavelength were fitted by using the exact equations that describe the systems studied. Good agreement between the experimental absorbance vs. pH plots and the curves generated by the fitting process was found. A comparison between the acidic constants for the first step of dissociation of these mycotoxins and the value for the acidic constant of phenol is discussed by assuming that phenolic structures are responsible for the acidity of these mycotoxins.Key words: mycotoxins, alternariol monomethyl ether, altertoxin-I, alternariol, acidic constants.

ABSTRACTThe modelling of the aggregation kinetics of iron oxides has been succesful in predicting the increase in aggregate size as determined by dynamic light scattering measurements. The aggregates were found to exhibit fractal behaviour with fractal dimensions obtained from the scattering exponent in static light scattering studies dependent on the aggregation mechanism and ranging from 2.3 for rapid (diffusion limited) to 2.8 for slow (reaction limited) aggregation. Polydispersity and restructuring of aggregates were found not to affect the relationship between scattering exponent and aggregate fractal dimension. Excellent correspondence over a range of temperatures and ionic strengths has been obtained between results of sizing experiments using dynamic light scattering and sizes predicted using a modified Smoluchowski model incorporating fractal dimensions.
