Control of steady-state pH in rabbit proximal straight tubules

Steady-state pH (defined as the limiting pH reached at slow flow rates) was measured in isolated perfused rabbit proximal straight tubules (S2). With normal bath conditions (pH 7.4, bicarbonate 25 mM) the luminal steady-state pH was 6.85. Steady-state pH was directly related to bath pH and bicarbonate, but not to bath PCO2. Lowering of bath pH or bicarbonate consistently decreased luminal steady-state pH, and raising either caused steady-state pH to increase. When bath PCO2 was increased, on the other hand, steady-state pH either increased or decreased, depending on the concomitant changes in bicarbonate and pH. The changes in steady-state pH observed in the present studies following alterations in bath pH and bicarbonate concentration should, when extrapolated to the in vivo kidney, result in decreased delivery of bicarbonate from the proximal tubule in acidosis and increased delivery in alkalosis. The effects of potassium and chloride were also determined. Removal of potassium from the bath increased the steady-state pH, but removal of chloride from both the perfusate and bath had no significant effect.

The effect of parathyroid hormone on bicarbonate absorption was tested in rabbit proximal renal tubules perfused in vitro. In proximal straight tubules 0.05 U/ml of parathyroid hormone caused a large and reversible increase in the steady-state bicarbonate concentration in tubule fluid. Further, the rates of bicarbonate and fluid absorption (measured at faster flow rates) were inhibited approximately 50% by the hormone. We conclude that parathyroid hormone directly inhibits fluid and bicarbonate absorption by proximal straight tubules, causing an increase in the bicarbonate concentration in the tubule fluid, and we suggest that this action of the hormone contributes to the increase in renal bicarbonate excretion that is generally caused by the hormone. In proximal convoluted tubules, parathyroid hormone was previously demonstrated by other investigators to inhibit fluid and bicarbonate absorption approximately proportionally, so that there was little or no change in the bicarbonate concentration in tubule fluid. In agreement we found in the present studies that 0.05 U/ml of the hormone did not affect the steady-state bicarbonate concentration in proximal convoluted tubule fluid and that 5 U/ml caused only an equivocal increase in tubule fluid bicarbonate concentration.

[8-Neopentylglycine]oxytocin (II) and [8-cycloleucine]oxytocin (III) were prepared by a combination of solid-phase synthesis and fragment condensation. Both analogues exhibited decreased uterotonic potency in vitro, each being about 15-30% that of oxytocin. Analogue II also displayed similarly decreased uterotonic potency in vivo and galactogogic potency. On the other hand, analogue III exhibited almost the same potency as oxytocin in the uterotonic assay in vivo and in the galactogogic assay.

We prepared [meso-tetrakis(4-carboxylatophenyl)porphyrinato]oxovanadium(IV) tetrasodium, ([VO(tcpp)]Na4), and investigated its in vitro insulin-mimetic activity and in vivo metallokinetic feature in healthy rats. The results were compared with those of previously proposed insulin-mimetic oxovanadium(IV)porphyrin complexes and oxovanadium(IV) sulphate. The in vitro insulin-mimetic activity and bioavailability of [VO(tcpp)]Na4 were considerably better than those of [meso-tetrakis (1-methylpyridinium-4-yl)porphyrinato]oxovanadium(IV)(4+) tetraperchlorate ([VO(tmpyp)](ClO4)4) and oxovanadium(IV) sulphate. On the other hand, [VO(tcpp)]Na4 and [meso-tetrakis(4-sulfonatophenyl) porphyrinato]oxidovanadate(IV)(4-)([VO(tpps)]) showed very similar in vitro insulin-mimetic activity and in vivo metallokinetic feature in healthy rats. In particular, the order of in vitro insulin-mimetic activity of the complexes was determined to be: [VO(tcpp)]Na4 ≈ [VO(tpps)] > ([VO(tmpyp)](ClO4)4 > oxovanadium(IV) sulphate.

The flow path close to the suction side of fan rotor blades mostly affects the overall drag of the blading. The blade lift is affected as well because of the separation of the low energy boundary layer that drives the blade into stall at low fan flow rates. Forward sweep allows to position the airfoil sections of blades featuring a positive circulation gradient along the span so that they “accompany” the near-wall flow trajectories at the blade suction side. So, rotor efficiency and stall margin of the fan can be improved. On the other hand, blade end effects play a relevant role in high hub-to-tip and low aspect ratio rotors and may compromise the effectiveness of forward sweep. Nevertheless, some authors in the literature stated the beneficial contribution of changing the sweep angle at the ends of the blade both at design and off-design conditions. The paper studies the end effects on constant-swirl design rotors by means of CFD simulations focusing on the distribution of blade sweep in the near-tip region. In particular, the performance and efficiency calculated for a forward swept tube-axial fan featuring a hub-to-tip ratio equal to 0.4 are compared with those estimated for the corresponding unswept fan at equal duty point. Several modifications of the sweep distribution in the blade tip region are considered in the swept fan to quantify their effect on performance, efficiency and stall margin. Results show that the addition of up to 6 degrees of local forward sweep at the blade tip to the unswept blading does not affect fan pressure at design operation. On the other hand, this local increase of the sweep angle allows for a very notable increase of the peak pressure and efficiency at flow rates close to stall inception.

Mammalian spermatozoa acquire their fertilizing ability in the female reproductive tract (sperm capacitation). On the other hand, seminal vesicle secretion, which is a major component of seminal plasma, inhibits the initiation of sperm capacitation (capacitation inhibition) and reduces the fertility of the capacitated spermatozoa (decapacitation). There are seven major proteins involved in murine seminal vesicle secretion (SVS1-7), and we have previously shown that SVS2 acts as both a capacitation inhibitor and a decapacitation factor, and is indispensable forin vivofertilization. However, the effects of SVSs other than SVS2 on the sperm have not been elucidated. Since mouseSvs2–Svs6genes evolved by gene duplication belong to the same gene family, it is possible that SVSs other than SVS2 also have some effects on sperm capacitation. In this study, we examined the effects of SVS3 and SVS4 on sperm capacitation. Our results showed that both SVS3 and SVS4 are able to bind to spermatozoa, but SVS3 alone showed no effects on sperm capacitation. On the other hand, SVS4 acted as a capacitation inhibitor, although it did not show decapacitation abilities. Interestingly, SVS3 showed an affinity for SVS2 and it facilitated the effects of SVS2. Interaction of SVS2 and spermatozoa is mediated by the ganglioside GM1 in the sperm membrane; however, both SVS3 and SVS4 had weaker affinities for GM1 than SVS2. Therefore, we suggest that separate processes may cause capacitation inhibition and decapacitation, and SVS3 and SVS4 act on sperm capacitation cooperatively with SVS2.

We investigate the transfer of bipartite (measured by cocurrence) and multipartite (measured by global discord) quantum correlations though spin chains under phase decoherence. The influence of phase decoherence and anisotropy parameter upon quantum correlations transfer is investigated. On the one hand, in the case of no phase decoherence, there is no steady state quantum correlations between spins. On the other hand, if the phase decoherence is larger than zero, the bipartite quantum correlations can be transferred through a Heisenberg XXX chain for a long time and there is steady state bipartite entanglement. For a Heisenberg XX chain, bipartite entanglement between two spins is destroyed completely after a long time. Multipartite quantum correlations of all spins are more robust than bipartite quantum correlations. Thus, one can store multipartite quantum correlations in spin chains for a long time under phase decoherence.

Since many of newly synthesised D-secoestratriene derivatives showed antiestrogenic effect, with almost a total loss of estrogenic activity, we studied the effects of some of these compounds on estrogen receptors (ER), the translocation of the estrogen-ER complexes formed in presence of competing substances into the nucleus, as well as the binding of these complexes to DNA. The results of uterotrophic effects of analysed derivatives are in agreement with the influence of these compounds on activity and binding parameters of estrogen receptors. Namely, compounds that show relatively high antiestrogenic activity predominantly increase Kd and inhibit translocation to nuclei of radioactive complexes formed in their presence. On the other hand, compounds that do not significantly change binding parameters of estrogen receptors do not show antiestrogenic effect in in vivo experiments.

Permeability of the tubular epithelium to urate presented on the peritubular side was studied in anesthetized rats during mannitol diuresis by capillary microinjections of [14C]urate and [3H]inulin, [14C]PAH and [3H]inulin, or [14C]urate and [3H]PAH. Recovery of isotopes was determined in urine collected serially from the injected and contralateral kidney. Appearance and peak excretion of urate in the experimental but not in the contralateral kidney preceded inulin and coincided with PAH, indicating proximal permeability to urate. Recovery of urate was higher from the injected than from the contralateral kidney. Urate precession and recovery did not change after addition of PAH (1.5-6.4 mm) to [14C]urate-[3H]inulin solutions, whereas they decreased significantly in the experimental kidney after pyrazinoate (1.6-3.2 mM) addition. On the other hand, no effect of urate on [14C]PAH-[3H]inulin injections was detectable. These findings are suggestive of a carrier-mediated transtubular influx of urate in rat proximal tubule. Absence of competition with PAH may suggest differences in the secretory mechanisms for organic acids.

ABSTRACT The autotransplanted ovary of the ewe was perfused in vivo via the ovarian artery with either 14C or 3H labelled C19 steroids. 17β-Oestradiol was the major phenolic steroid isolated in ovarian venous blood from either testosterone or androstenedione. Smaller amounts of oestrone were obtained but there was no 17α-oestradiol, oestriol nor conjugated oestrogens isolated. The yield of oestrogen was approximately ten fold greater from androstenedione than from testosterone suggesting that the main route of oestrogen biosynthesis in the ovine ovary is via the former steroid. The effect of infusing luteinizing hormone (LH) at the rate of 10 μg per hour on the conversion of androstenedione to 17β-oestradiol was measured in 5 experiments. In 2 experiments, when the steady state was not achieved, the increasing rate of conversion was halted. On the other hand LH resulted in a temporary increase followed by a decrease in the rate of conversion in the remaining 3 experiments in which there was a constant rate of conversion in the control samples. These results are compatible with the concept that LH stimulates the aromatisation of androstenedione to oestrogens by the ovary in vivo.

In constitutional democracies, laws take time to be deliberated upon, to be passed, and to be implemented. Motivated by this observation, we study the properties of optimal tax reform when it has to be announced in advance of its implementation. We find that a delay between announcement and implementation has large effects on the optimal fiscal policy during the transition to the new steady state. On the other hand, we find that the welfare gains from optimal tax reform are fairly robust to the introduction of an implementation lag. Increasing the lag from zero to four years reduces the welfare gains by less than a quarter. Moreover, it turns out that this reduction of the welfare gain is mainly due to the delay itself rather than the effect of preannouncement on the character of the optimal tax reform.
