Implications of Ocular Kinematics for the Internal Updating of Visual Space

Recent studies have suggested that during saccades cortical and subcortical representations of visual targets are represented and remapped in retinal coordinates. If this is correct, then the remapping processes must incorporate the noncommutativity of rotations. For example, our three-dimensional (3-D) simulations of the commutative vector-subtraction model of retinocentric remapping predicted centripetal errors in saccade trajectories between “remembered” eccentric targets, whereas our noncommutative model predicted accurate saccades. We tested between these two models in five head-fixed human subjects. Typically, a central fixation light appeared and two peripheral targets were flashed. With all targets extinguished, subjects were required to saccade to the remembered location of one of the peripheral targets and saccade between their remembered locations. Subjects showed minor misestimations of the spatial locations of targets, but failed to show the cumulative pattern of errors predicted by the commutative model. This experiment indicates that if targets are remapped in a retinal frame, then the remapping process also takes the noncommutativity of 3-D eye rotations into account. Unlike other noncommutative aspects of eye rotations that may have mechanical explanations, the noncommutative aspects of this process must be entirely internal.

Understanding the collective dynamics of crowd movements during stressful emergency situations is central to reducing the risk of deadly crowd disasters. Yet, their systematic experimental study remains a challenging open problem due to ethical and methodological constraints. In this paper, we demonstrate the viability of shared three-dimensional virtual environments as an experimental platform for conducting crowd experiments with real people. In particular, we show that crowds of real human subjects moving and interacting in an immersive three-dimensional virtual environment exhibit typical patterns of real crowds as observed in real-life crowded situations. These include the manifestation of social conventions and the emergence of self-organized patterns during egress scenarios. High-stress evacuation experiments conducted in this virtual environment reveal movements characterized by mass herding and dangerous overcrowding as they occur in crowd disasters. We describe the behavioural mechanisms at play under such extreme conditions and identify critical zones where overcrowding may occur. Furthermore, we show that herding spontaneously emerges from a density effect without the need to assume an increase of the individual tendency to imitate peers. Our experiments reveal the promise of immersive virtual environments as an ethical, cost-efficient, yet accurate platform for exploring crowd behaviour in high-risk situations with real human subjects.

A technique is developed for quantitative measurement of general three-dimensional motion, and this technique is applied to the kinematics of anatomical joints. The spatial locations of three orthogonal points representing coordinate frames on each member of the joint are measured during motion of the joint by photo encoders of a three-dimensional mechanical pointer. Kinematic calculations are used to derive, from the experimentally collected data, the six orthogonal components of the motion of one member relative to the other. The accuracy of this technique is presented. Applications to the knee and ankle are discussed.

Section topographs recorded at different spatial locations and at different rocking angles of a highly oriented pyrolytic graphite (HOPG) crystal allow three-dimensional maps of the local angular-dependent scattering power to be obtained. This is performed with a direct reconstruction from the intensity distribution on such topographs. The maps allow the extraction of information on local structural parameters such as size, form and internal mosaic spread of crystalline domains. This data analysis leads to a new method for the characterization of mosaic crystals. Perspectives and limits of applicability of this method are discussed.

Twelve males were tested under a control and two alcohol treatments in a perimeter apparatus used for testing peripheral vision. They were required to fixate either on a steady-state central fixation light and detect peripheral lights, or to count blinks produced by the cessations of the fixation light and to detect peripheral lights. Alcohol produced an impairment of peripheral vision only under conditions where the central fixation light blinked and thus required information processing. No performance decrement occurred when the central light did not blink. The results suggest that alcohol interferes with central information processing rather than peripheral sensory mechanisms.

The purpose of this study was to quantify the effect of soft tissue artifact during three-dimensional motion capture and assess the effectiveness of an optimization method to reduce this effect. Four subjects were captured performing upper-arm internal-external rotation with retro-reflective marker sets attached to their upper extremities. A mechanical arm, with the same marker set attached, replicated the tasks human subjects performed. Artificial sinusoidal noise was then added to the recorded mechanical arm data to simulate soft tissue artifact. All data were processed by an optimization model. The result from both human and mechanical arm kinematic data demonstrates that soft tissue artifact can be reduced by an optimization model, although this error cannot be successfully eliminated. The soft tissue artifact from human subjects and the simulated soft tissue artifact from artificial sinusoidal noise were demonstrated to be considerably different. It was therefore concluded that the kinematic noise caused by skin movement artifact during upper-arm internal-external rotation does not follow a sinusoidal pattern and cannot be effectively eliminated by an optimization model.

Humans have the inherent ability of performing highly dexterous and skillful tasks with their arms, involving maintenance of posture, movement, and interaction with the environment. The latter requires the human to control the dynamic characteristics of the upper limb musculoskeletal system. These characteristics are quantitatively represented by inertia, damping, and stiffness, which are measures of mechanical impedance. Many previous studies have shown that arm posture is a dominant factor in determining the end point impedance on a horizontal (transverse) plane. This paper presents the characterization of the end point impedance of the human arm in three-dimensional space. Moreover, it models the regulation of the arm impedance with respect to various levels of muscle co-contraction. The characterization is made by route of experimental trials where human subjects maintained arm posture while their arms were perturbed by a robot arm. Furthermore, the subjects were asked to control the level of their arm muscles’ co-contraction, using visual feedback of their muscles’ activation, in order to investigate the effect of this muscle co-contraction on the arm impedance. The results of this study show a very interesting, anisotropic increase of arm stiffness due to muscle co-contraction. These results could lead to very useful conclusions about the human’s arm biomechanics, as well as many implications for human motor control-specifically the control of arm impedance through muscle co-contraction.

AbstractHuman cerebral malaria (HCM), a severe encephalopathy associated with Plasmodium falciparum infection, has a 20–30% mortality rate and predominantly affects African children. The mechanisms mediating HCM-associated brain injury are difficult to study in human subjects, highlighting the urgent need for non-invasive ex vivo human models. HCM elevates the systemic levels of free heme, which damages the blood-brain barrier and neurons in distinct regions of the brain. We determined the effects of heme on induced pluripotent stem cells (iPSCs) and a three-dimensional cortical organoid system and assessed apoptosis and differentiation. We evaluated biomarkers associated with heme-induced brain injury, including a pro-inflammatory chemokine, CXCL-10, and its receptor, CXCR3, brain-derived neurotrophic factor (BDNF) and a receptor tyrosine-protein kinase, ERBB4, in the organoids. We then tested the neuroprotective effect of neuregulin-1 (NRG-1) against heme treatment in organoids. Neural stem and mature cells differentially expressed CXCL-10, CXCR3, BDNF and ERBB4 in the developing organoids and in response to heme-induced neuronal injury. The organoids underwent apoptosis and structural changes that were attenuated by NRG-1. Thus, cortical organoids can be used to model heme-induced cortical brain injury associated with HCM pathogenesis as well as for testing agents that reduce brain injury and neurological sequelae.
