Pooling is an attractive strategy in screening infected specimens, especially for rare diseases. An essential step of performing the pooled test is to determine the group size. Sometimes, equal group size is not appropriate due to population heterogeneity. In this case, varying group sizes are preferred and could be determined while individual information is available. In this study, we propose a sequential procedure to determine varying group sizes through fully utilizing available information. This procedure is data driven. Simulations show that it has good performance in estimating parameters.

The subject of the work is the research on relevant factors influencing participation in the success of brownfield revitalization, especially in the territory of small municipalities. Research has so far dealt with the issue of determining disparities in the municipalities of the Czech Republic, not excluding small municipalities, but their subsequent application has usually been presented in larger cities. The focus on smaller municipalities or cities was usually addressed only in general. The introduction provides an overview of theoretical knowledge in the field of brownfield revitalization. Defining the level of knowledge of the monitored issues is an essential step for the purposes of more effective determination of disparities. Disparities will be determined on the basis of information on localities that have been successfully revitalized. The identified disparities are then monitored in the territory of small municipalities. For the purposes of processing, it was determined that a small municipality or city is an area with a maximum of 5000 inhabitants. Using appropriately selected statistical methods, an overview of disparities and their weights is determined, which significantly affect the success of revitalization. In small municipalities, the issue of brownfields is not emphasized but, in terms of maintaining community strength and reducing population turnover, the reuse of brownfields is a crucial theme.

Vernal keratoconjunctivitis (VKC) and atopic keratoconjunctivitis (AKC) are potentially severe and complex disease in its management among the various allergic eye diseases. In this regard, studies clarified the etiopathogenetic mechanisms. The workup should be multidisciplinary. The treatment includes topical and systemic medications with anti-inflammatory and immunosuppressant activity. However, a definition of nationally- and internationally-shared diagnostic protocols would also be needed and validated access to therapeutic options of proven safety and efficacy to avoid the use of galenic preparations, up to now still essential in the management of moderate-severe VKC. Finally, recognizing VKC and AKC, among rare diseases, at a national and international level would be an essential step to allow the management of VKC with adequate timings and settings within the National Health System.

The sample preparation is considered an essential step in the analysis process. The complexity of the matrix and how the compounds are linked is one of the main difficulties in the extraction. Thus a solvent system combined with an extraction method, both need to be effective against such mishaps. In this work we used the 23 factorial design to determine the optimum conditions for sample preparation. The analysis of the samples were performed in a molecular fluorescence spectrophotometer. The factors selected to investigate the influence on the extraction system were: time of use of ultrasound, the type of solvent (hexane or dichloromethane) and ratio solvent / sample (1:1 and 1:3). The best result was obtained with using the dichloromethane in time of 10 min of ultrasound and the ratio solvent / sample 1:3.

International agreements and policies on climate change urge for a drastic reduction in greenhouse gas (GHG) emissions to prevent a temperature rise above 2 °C at the end of the century. Determination of the product carbon footprint (CF), identifying carbon hotspots and examining ways for CF reduction is an essential step towards mitigation actions. Viticulture and winemaking are very important for the economy of Mediterranean regions, especially for the sustainability of rural areas. To determine the CF for wine, the Life Cycle Assessment (LCA) approach was adopted with system boundaries from vineyard to market. Input data were collected from 20 vineyards on the island of Cyprus, where the indigenous Xynisteri variety is cultivated and from an SME winery that uses the grapes to produce wine. The winery CF was 99,586 kg CO2-eq for 76,000 bottles produced (1.31 kg/0.75 L bottle). The uncertainty factor was determined to be ±50%, which was considered adequate for the methodology followed. The share of electrical energy was 46%, of packaging 18% and of the vineyard 16%. Fuel (transportation and heating), as well as waste management (solid and wastewater) contributed 10% each to the CF. There is potential for mitigation of the CF by replacing the bottles currently used by lighter ones, lowering the energy use and reusing a part of the solid waste as fertilizer. Research towards eco-innovation of viticulture/winemaking is essential for reducing the footprint of the sector to promote sustainable wine production.

Resilience refers to the ability to recover from illness or adversity. At the cell, tissue, organ and whole-organism levels, the response to perturbations such as infections and injury involves the acute inflammatory response, which in turn is connected to and controlled by changes in physiology across all organ systems. When coordinated properly, inflammation can lead to the clearance of infection and healing of damaged tissues. However, when either overly or insufficiently robust, inflammation can drive further cell stress, tissue damage, organ dysfunction and death through a feed-forward process of inflammation → damage → inflammation. To address this complexity, we have obtained extensive datasets regarding the dynamics of inflammation in cells, animals and patients, and created data-driven and mechanistic computational simulations of inflammation and its recursive effects on tissue, organ and whole-organism (patho)physiology. Through this approach, we have discerned key regulatory mechanisms, recapitulated in silico key features of clinical trials for acute inflammation and captured diverse, patient-specific outcomes. These insights may allow for the determination of individual-specific tolerances to illness and adversity, thereby defining the role of inflammation in resilience.
