Introduction. Early childhood caries is a kind of caries occurring in deciduous teeth. Bacteria are among the main factors. Antibacterial agents such as fluoride are used in both prevention and treatment, but their application in children faces limitations such as fluorosis. Therefore, novel methods of caries prevention among the children are mainly focused on the use of fluoride-free active ingredients. In this comparative study, antibacterial effects of gels containing propolis and aloe vera, fluoride, xylitol, and CPP-ACP were investigated. Methods. This is an in vitro study. By plate well technique, plates containing gels were created in the culture medium of Streptococcus mutans and Lactobacillus, and their antibacterial impacts were evaluated by measuring the inhibition zone after 24, 48, and 72 hours. Then, different concentrations of each gel were evaluated in the same way for the antibacterial properties. For each sample, this process was iterated 3 times, where the average was declared as the final number. The collected data were entered in SPSS 24. Results. In both bacteria, propolis gel and aloe vera had the highest zone of inhibition, followed by fluoride and xylitol in the second and third places, respectively. Different concentrations of gels are significantly different in terms of antimicrobial effect (P value ≤ 0/05). The antimicrobial effect of propolis and aloe vera gel was kept up to the concentration of 1 / 16 . As the bacterial and gel contact time is prolonged, the antibacterial effect of different gels increases, but the difference is not statistically significant (P value = 0.109). CPP-ACP gel had no antimicrobial effect at any concentration. Conclusion. Propolis and aloe vera gel had a greater antimicrobial effect than other gels, where such effect was observed in low concentrations. CPP-ACP gel had no antimicrobial properties.

Abstract Objectives The aim of the present in vitro study is to compare the remineralization brushing effect of three toothpastes and Aloe vera (AV) gel. Materials and Methods Forty sound extracted teeth were placed in a demineralizing solution for 4 days and randomly assigned to four groups: group A: 1,450-ppm fluoride toothpaste; group B: AV nonfluoridated toothpaste; group C: AV 1,000-ppm fluoridated toothpaste; and group D: AV gel. A 3-minute pH cycling was performed twice a day for each group for 12 days. Specimens were analyzed before and after by scanning electron microscope—energy dispersive X-ray. Statistical analysis The outcomes were analyzed by Kolmogorov–Smirnov’s tests, repeated-measures analyses of variance followed by univariate analyses, and Bonferroni’s multiple comparisons tests to compare the calcium-to-phosphorus (Ca:P) ratio within time among toothpaste groups. Results Following remineralization, the Ca:P ratio increased in all groups. The difference of the Ca:P ratio was not significant between groups C, D, and A. The mean ratio was significantly lower in group B (p-value = 0.026). Conclusions The AV gel demonstrated a remineralization capacity equal to that of the 1,450-ppm fluoride toothpaste. In contrast, fluoride-free AV toothpaste showed a lower remineralization efficiency. Further studies are required to understand its mechanism.

BACKGROUND AND OBJECTIVES: This study evaluated and compared the antimicrobial efficacy and minimal inhibitory concentration (MIC) of chlorhexidine 0.12% and extracts of aloe vera and mushroom against Streptococcus mutans and lactobacillus. MATERIALS & METHODOLOGY: The agar disc diffusion and broth micro dilution method was used to check the antimicrobial activity of 0.12% chlorhexidine and aqueous extracts of two medicinal plants. The test samples were divided as follows- Streptococcus mutans, group I: chlorhexidine, group II: Aloe vera and group III: Mushroom. Lactobacillus, Group I: chlorhexidine, group II: Aloe vera extract and group III: Mushroom extract. The zone of inhibition and MIC values were tabulated and the data was statistically analyzed using ANOVA and Bonferroni post- hoc tests. RESULTS: Chlorhexidine shows maximum antibacterial action against Streptococcus mutans and lactobacillus followed by mushroom extract and aloe veraextracts with the zone of inhibition of (20.3mm/ 24.13), (17.03/ 16.05), (14.09/14.26) respectively. Both the extracts had MIC value of 80µg/ ml and chlorhexidine was 40µg/ml. CONCLUSION: Within the limitations of this in- vitro study, it can be concluded that all the herbal extracts tested in this study demonstrated antibacterial activity against MS and lactobacillus. Chlorhexidine showed maximum antibacterial action against Streptococcus mutans and lactobacillus followed by mushroom extract and then aloe veraextracts with the zone of inhibition of (20.3mm/24.13), (17.03/ 16.05), (14.09/14.26) respectively. Herbal products have potent antimicrobial activity that can be looked at as an alternative to chlorhexidine. However, further in vitro and long-term in vivo studies are recommended to confirm and correlate the findings of this study clinically.

Abstract Background The mechanism of action of silver diammine fluoride (SDF) on plaque micro-ecology is seldom studied. This study investigated micro-ecological changes in dental plaque on extensive caries of deciduous teeth after topical SDF treatment. Methods Deciduous teeth with extensive caries freshly removed from school children were collected in clinic. Unstimulated saliva collection and initial plaque sampling were done before tooth extraction, then each caries was topically treated with 38% SDF in vitro. After intervention, each tooth was stored respectively in artificial saliva at 37°C. Repeated plaque collections were done at 24 hours and 1 week post-intervention. Post-intervention micro-ecological changes including microbial diversity, microbial metabolism function as well as species correlations were analyzed and compared after pyrosequencing of the DNA from the plaque sample using Illumina MiSeq platform. Results After SDF application, microbial diversity decreased (p>0.05), although not statistically significant. Microbial community composition post-intervention was noticeably different from that of supragingival and pre-intervention plaque as well as saliva. At 1 week post-intervention, the relative content of Pseudomonas , Fusobacterium and Pseudoramibacter was noticeably higher than before, while most of the other bacteria was noticeably reduced, although not statistically significant (P>0.05). The inter-microbial associations became more complex, much more positive associations among survived bacteria were observed than negative ones. COG function classification diagram showed carbohydrate transportation and metabolic functions in the plaque were significantly reduced at 24 hours and 1 week post-intervention. Conclusions SDF has extensive antimicrobial effect on dental plaque, which may reduce carbohydrate metabolism in dental plaque and help promote new balance of the plaque flora.

Background: Aloe vera contains active components such as antrhaquinones,saponin, glucomannans, and acemannans that have been known for its antimicrobial properties. In vitro study showed that Aloe vera extract can inhibit the growth of Streptococcus mutans. Studies reported that children with caries have high level of salivary S. mutans which is the main pathogen in dental caries. The gold standard mouthrinse for dental caries is chlorhexidine gluconate, but it has several side effects. The objective of this study was to compare the effect between Aloe vera juice and 0.06% chlorhexidine mouth rinse on salivary S. mutans in children with caries.Methods: This experimental research with pretest and post-test control group design conducted on 30 children (6-12 years old) with number of carious teeth >4. Subjects divided into 2 groups: Aloe vera juice group and 0.06% chlorhexidine group, which instructed to rinse their mouth with 10 ml stated mouthwash, twice a day, during 7-day period. Unstimulated saliva was collected at baseline and after 7 days then inoculated on mitis salivarius bacitracin agar. S. mutans were counted and expressed in CFU/ml. Data were analyzed using paired t-test and independent t-test.Result: The result showed that a significant reduction in salivary S.mutanscounts was observed with Aloe vera juice mouth rinse and 0.06% chlorhexidine (p<0.05). The mean reduction of S. mutans counts was not statistically significantly between grup (p>0.05).Conclusion: These findings suggest that Aloe vera juice can reduce salivary S. mutans in children with caries so it can be considered as alternative mouthwash to prevent caries.
