Description of Aphelenchoides huntensis sp. n. (Nematoda: Aphelenchoididae) isolated from Pinus sylvestris in western Iran

Aphelenchoides huntensissp. n. is described and illustrated from wood and bark samples of a dead scots pine in Kermanshah Province, western Iran. The new species has a body length of 507-673 μm (females) and 636-640 μm (males). The cuticle is weakly annulated with four lateral lines. Lip region offset. The stylet is 9.5 (9.0-10.0) μm long with small basal swellings. The excretory pore is locatedcaone body diam. posterior to the metacorpus valve. The spicules are large with the dorsal limb 37 or 38 μm long (n = 2), and have a prominent rostrum and rounded apex. The female tail is conical, the terminus having a complicated step-like projection, usually with many tiny nodular protuberances. The male tail bears six (2 + 2 + 2) caudal papillae and a well-developed mucron. The new species belongs to the Group 2 category ofAphelenchoidesspecies. Phylogenetic analysis based on the 18S and 28S D2-D3 regions of rDNA confirmed its status as a new species.

Aphelenchoides eldaricusn. sp. is described and illustrated from wood and bark samples of a dead Mondell pine in Kermanshah Province, western Iran. The new species has a body length of 597 (507-700) μm (females) and 662 (636-695) μm (males). The cuticle is weakly annulated with three lateral lines and the lip region is offset. The stylet is 9.6 (9-10) μm long with small basal swellings and the excretory pore is locatedcaone body diam. posterior to the metacorpus valve. The spicules are large with the dorsal limb 40 (38-42) μm long with a prominent rostrum and rounded apex. The female tail is conical, tapering gradually to an acute terminus with a simple ventral mucron. The male tail bears six (2 + 2 + 2) caudal papillae and a well-developed mucron. The new species belongs to the Group 2 category ofAphelenchoidesspecies and is close to four known species, namelyA. cibolensis,A. indicus,A. rutgersiandA. sacchari. Molecular analyses of the partial 18S and the D2-D3 expansion segments of the 28S rRNA gene confirmed its morphological status as a new species.

Ruehmaphelenchus quercophilus n. sp., isolated from a dying oak, Quercus robur, in the State Forests, Łopuchówko District, Poland, is described and figured. It is characterised by three lateral lines, relatively stout body, excretory pore posterior to nerve ring, five caudal papillae, spicules relatively small (12-14 μm) with weakly developed condylus and rostrum, bursa absent, vulva positioned at ca 78% of body length, vulval flap absent, post-uterine branch extending for slightly over half of vulva to anus distance, and conoid tail with a bend at the distal end forming a terminal projection 10-14 μm long. The new species is very similar to four other described species, namely R. formosanus, R. juliae, R. thailandae, and R. sirisus. Detailed phylogenetic analysis based on 18S, 28S D2-D3 and ITS sequences has confirmed the status of this nematode as a new species.

Aphelenchoides salixaen. sp. was isolated from wood and bark samples of a dead willow in Kermanshah Province, western Iran. The nematodes were collected directly from wood samples and were successfully reared onBotrytis cinerea. The new species has a body length of 567 (507-620) μm (females) and 382 or 400 μm (males). The cuticle is weakly annulated with four lateral lines. Lip region offset. The stylet is 9.5 (9-10) μm long with small basal swellings. The excretory pore is locatedcaone body diam. posterior to the metacorpus valve. The spicules are short with the dorsal limb 14 or 15 μm long (n = 2), apex and rostrum rounded and only slight offset. The female tail is cylindrical with a broadly rounded terminus. The male tail is conical, bearing six (2 + 2 + 2) caudal papillae with a hamate mucron at terminus. The new species belongs to the Group 1 category ofAphelenchoidesspecies and is close to seven known species, namely:A. capsuloplanus,A. confusus,A. limberi,A. obtusicaudatus,A. obtusus,A. orientalisandA. rotundicaudatus. Molecular analyses of the partial 18S, D2-D3 expansion segments of the 28S rRNA genes and internal transcribed spacer (ITS) supported it as a new species and that the new species is closest toA.besseyi,A. fujianensisandA. ritzemabosiin dendrograms inferred using 18S and 28S D2-D3 genes.

Summary Ruehmaphelenchus taedae n. sp., isolated from Loblolly pine logs (Pinus taedae L.) from the USA, is described and figured. It is characterised by a relatively slim body (a = 42 and 43 for males and females, respectively), three lines in the lateral field, male spicules relatively small (12-18 μm) with high and dorsally bent condylus and weakly developed rostrum, bursal flap absent, short tail possessing a long terminal spike ending in a bluntly rounded tip and 8.7-13.3 μm long, vulva positioned at ca 83% of body length, vulval flap absent, vulval lips slightly protruding, post-vulval uterine branch extending for less than half of vulva to anus distance, and female tail conoid, ca 3-4 anal body diam. long, with 13.7-18.5 μm terminal projection. The new species can be separated from all other species of the genus by the male tail possessing a long terminal spike and the more anterior excretory pore. Detailed phylogenetic analysis based on 28S D2-D3 region sequences confirmed the status of this nematode as a new species.

Aphelenchoides xui sp. n. is described and illustrated from imported packaging wood from South Africa intercepted at Ningbo Port, China. The new species has a body length of 564-820 μm (males) and 549-882 μm (females). The cuticle is weakly annulated with four lines in the lateral field. The stylet is 11-13 μm long and has small basal swellings. The excretory pore is located ca one body diam. posterior to the median bulb, or 75-100 μm from the head. Spicules are relatively large (18-23 μm) with apex and rostrum rounded and well developed and the end of the dorsal limb clearly curved ventrad like a hook. The male tail bears six (2 + 2 + 2) caudal papillae. The spermatheca is axial and oblong and contains disc-like sperm. The female tail is conical, terminating in a complicated step-like projection, usually with many tiny nodular protuberances. The new species is morphologically similar to A. arcticus, A. haguei and A. parasaprophilus in Group 2 sensu Shahina, but is distinguished by spicule shape and form of the female tail terminus. Phylogenetic analysis based on SSU and partial LSU sequences revealed that A. xui sp. n. was closest to A. varicaudatus, although some species of Laimaphelenchus and Schistonchus were also included in the same branch.

Abstract Bursaphelenchus chengi sp. n. is described and illustrated. Dauer juveniles were isolated from imported wood packaging materials from Taiwan to Nanjing Port, China. Bursaphelenchus chengi sp. n. was reared and maintained on Petri dish cultures of the fungus Botrytis cinerea. The new species is characterised by the medium body size in both sexes, the presence of only two incisures in the lateral field and the robust and strongly curved spicules. The spicule lamina is angular distally, the rostrum digitate and the condylus rounded. The tail is arcuate with a pointed terminus. The bursa is usually truncate with the posterior margin indented in some specimens or rounded with a fine axial point. Females have a small vulval flap formed by a short extension of the cuticle of the anterior lip, and a conical tail that gradually tapers to an almost straight or slightly recurved, pointed or rounded terminus. Because of the presence of two lateral lines, similar spicule shape, tapering female tail and the presence of a small vulval flap, B. chengi sp. n. should be grouped in the abietinus-group sensu Braasch. together with B. abietinus, B. antoniae, B. hellenicus, B. hylobianum and B. rainulfi. ITS-RFLP profiles support the proposal of the new species, and phylogenetic analysis of the 28S rDNA D2/D3 domain sequence places it close to B. antoniae and other species of the abietinus-group.

Summary Aphelenchoides kheirii n. sp. was isolated during a survey of nematodes associated with bark samples of pine trees (Pinus nigra) in West Azerbaijan province, north-western Iran. The new species has a body length of 500 (448-520) μm in the female and 442 (402-480) μm in the male. The cuticle is weakly annulated with four lateral lines and the lip region is offset. The stylet is 10.4 (10-11) μm long with small basal swellings and the excretory pore located immediately posterior to base of metacorpus; hemizonid invisible. The post-vulval uterine sac length 37.7 (28-45) μm. Spicules are relatively short (18-20 μm in dorsal limb) and the end of the dorsal limb clearly curved ventrally like a hook. The male tail has usual three pairs of caudal papillae (2 + 2 + 2) and a well-developed mucron. The female tail is conical, terminating in a complicated step-like projection, usually with many tiny nodular protuberances. The new species belongs to the Group 2 category of Aphelenchoides species sensu Shahina in which ten known species among Groups 2 and 4 sensu Shahina, namely: A. arcticus, A. blastophthorus, A. fuchsi, A. parasaprophilus, A. paraxui, A. xui, A. dactylocercus, A. gynotylurus, A. iranicus and A. saprophilus are the most similar species. Phylogenetic analysis based on small subunit (SSU) and partial large subunit (LSU) sequences of rRNA supported the morphological results.

Summary Bursaphelenchus parayongensis n. sp. was isolated from packaging wood of a consignment from China exported to Germany. The new species is characterised by the lateral field with three lines, 9-13 μm long stylet with small basal swellings, and the excretory pore located near the posterior part of metacorpus. The male spicules are mitten-shaped, the lamina smoothly tapers towards the distal tip. The condylus is broadly rounded, with a small dorsally bent hook, rostrum triangular with sharply pointed tip, and without cucullus. One precloacal papilla, one adcloacal pair and two postcloacal pairs of male papillae present, with the posteriormost pair forming ‘gland papillae’. Bursal flap long, spade-like with posterior margin truncate or irregular. Females have a small vulval flap of 2-3 μm long, and a post-vulval uterine sac extending for 55-77% of the vulva to anus distance. Tail cylindrical, mostly (about 70%) with a robust mucron, very short to less than 3 μm, but occasionally tail terminus bluntly rounded, without mucron. It is similar to species in subgroup 3 of the eggersi group including B. carpini, B. clavicauda, B. cryphali and B. yongensis, but morphologically different. Detailed phylogenetic analysis based on partial 18S, ITS and D2-D3 region of 28S sequences has confirmed the status of this nematode as a new species.

Aphelenchoides stellatusn. sp. is described and figured. The new species was isolated from packaging wood from Japan imported to Ningbo harbour, China. The new species has a body length of 485-533 μm (males) and 547-699 μm (females). The cuticle is weakly annulated and there are four lines in the lateral field. The stylet is 9-11 μm long and has small basal swellings. The excretory pore is located posterior to the nerve ring. Spicules smoothly curved, rose-thorn shaped. Apex and rostrum round, only slightly offset, dorsal limb 19-21 μm long. Male tail bearing six (2 + 2 + 2) caudal papillae. The female spermatheca is axial and oblong with round sperm present in multiple rows. Both male and female tail pegs have 3-4 processes, appearing star-like under SEM. The new species belongs to the Group 3 category ofAphelenchoidesspecies. Phylogenetic analyses based on full length ITS and 28S D2/D3 region of rDNA confirmed its morphological status as a new species.

Summary Aphelenchoides smolae n. sp. isolated in Ningbo Port, China, from medium soil and tissues of Lilium orientalis bulbs imported from The Netherlands, is described. It is characterised by the female body length of 826 (672-1002) μm and male body length of 785 (683-844) μm, lateral field with four lines (three bands), stylet 13.0-14.9 μm long with small basal swellings, excretory pore situated slightly posterior to nerve ring, vulval flap absent, post-vulval uterine sac well developed and extending for ca 35.7-62.4% of vulva to anus distance, conical tail with terminus bearing a single mucron with many tiny nodular protuberances under SEM, males common, with spicules 25.7-29.8 μm long (median curved line), the condylus and rostrum rounded, well developed, the dorsal limb with a hook-like tip, and tail bearing three pairs (2 + 2 + 2) of caudal papillae and a sharply pointed mucron. The new species belongs to the Group 2 category of Aphelenchoides species sensu Shahina. Phylogenetic analysis based on 18S, 28S D2-D3 of rDNA and COI mtDNA further characterised the new species.
