Molecular phylogenetics of species of Bulbophyllum sect. Trias (Orchidaceae; Epidendroideae; Malaxidae) based on nrITS and plastid rbcL and matK

Phylogenetic relationships among species of Bulbophyllum sect. Trias and related taxa in Bulbophyllum has not been previously studied due to scarcity of appropriate samples. In this study, we aimed to assess the relationship of these taxa and investigate interspecific relationships of these orchids using DNA sequences from two plastid genes, rbcL and matK, and one nuclear region, nuclear ribosomal internal transcribed spacer, nrITS. Our results showed that all Bulbophyllum sect. Trias species were embedded among other groups of Bulbophyllum, supporting the already published transfer of Trias to synonymy of Bulbophyllum. All trees revealed that the Indochinese sect. Trias form a monophyletic group that could be divided into three groups coincident with their vegetative characters. This study further shows that nrITS sequences can be sufficient for inferring phylogenetic relationship among Trias species, although this marker and the combination of this plus the plastid genes are not able to distinguish the differences between some closely related species.

Species of the Cochlospermaceae, a small mostly pantropical plant family, were evaluated at a continental scale for medicinal uses in traditional medicine. This ethnobotanical information was placed in a phylogenetic framework to make informed predictions in the search for new medicines and bioactive compounds. Medicinal plant-use data were mapped onto a molecular phylogeny based on DNA sequences of nuclear and chloroplast markers. Associations of medicinal uses among closely related species occurring in different geographic regions and among diverse cultures were evaluated. The most common medicinal uses for these species are those used to treat skin ailments, gastro-intestinal problems, malaria, and liver issues. The plant species with the most numerous uses is Cochlospermum tinctorium, which occurs primarily in West Africa. Closely related species being used by cultural groups in different geographic regions to treat the same illnesses suggests the presence of bioactive compounds with potential biomedical value, since they may represent independent discoveries of similar medicinally-active compounds. This leads to the speculation that those closely related species not currently being used to treat these ailments may also contain identical or similar medicinally-active compounds and are worthy of laboratory investigations.

Russula subrutilans sp. nov., a new species of Russula is described from southern China. It is unique for having buff pink to light congo-pink pileus, distant ventricose to subventricose lamellae with rare lamellulae, globose to broadly ellipsoid spores with bluntly conical warts forming a partial reticulum, and narrowly clavate to clavate cheilocystidia and pleurocysitidia with variable tips. Phylogenetic relationships among the new species and other closely related species in the genus are inferred based on the internal transcribed spacer (ITS) region.

Summary A new species of Aporcelaimellus, collected in a watermelon field in Nigeria, is described, including its morphological and molecular (D2-D3 28S-rDNA, 18r-DNA) characterisation. Aporcelaimellus nigeriensis sp. n. is distinguishable by its 2.76-3.55 mm length, very coarse ventral body pores, lip region offset by deep constriction and 24-27 μm broad odontostyle 30-36 μm long at its dorsal and 28-31 μm at its ventral side, neck 648-779 μm long, pharyngeal expansion occupying 54-60% of total neck length, uterus 300-473 μm or 2.1-3.2 body diam. long and tripartite, V = 49-54, tail short and convex conoid (27-41 μm, c = 72-115, c′ = 0.5-0.7), spicules 108-137 μm long, and 9-10 spaced ventromedian supplements with hiatus. LSU analysis revealed a close relationship of A. nigeriensis sp. n. with other Aporcelaimellus species and questioned, once more, the monophyly of Aporcelaimidae. SSU phylogenetic tree was not able to resolve the relationship between the new species and other closely related species.

The medicinal plantFerulahas been widely used in Asian medicine, especially in Uyghur medicine in Xinjiang, China. Given that various substitutes and closely related species have similar morphological characteristics,Ferulais difficult to distinguish based on morphology alone, thereby causing confusion and threatening the safe use ofFerula. In this study, internal transcribed spacer 2 (ITS2) sequences were analyzed and assessed for the accurate identification of two salableFerulaspecies (Ferula sinkiangensisandFerula fukangensis) and eight substitutes or closely related species. Results showed that the sequence length of ITS2 ranged from 451 bp to 45 bp, whereas guanine and cytosine contents (GC) were from 53.6% to 56.2%. A total of 77 variation sites were detected, including 63 base mutations and 14 insertion/deletion mutations. The ITS2 sequence correctly identified 100% of the samples at the species level using the basic local alignment search tool 1 and nearest-distance method. Furthermore, neighbor-joining tree successfully identified the genuine plantsF. sinkiangensisandF. fukangensisfrom their succedaneum and closely related species. These results indicated that ITS2 sequence could be used as a valuable barcode to distinguish Uyghur medicineFerulafrom counterfeits and closely related species. This study may broaden DNA barcoding application in the Uyghur medicinal plant field.

Recent studies have successfully extended the repertoire of microsatellite analysis beyond population genetics and demonstrate that phylogenetic relationships of closely related species can be inferred accurately with fewer loci than previously assumed.

A new species of Trogia (Marasmiaceae, Basidiomycota) is described from West Bengal, India. Analysis of the molecular sequence (nuclear ribosomal internal transcribed spacer region) data suggests that T. benghalesis is phylogenetically distinct from its closely related species. Comprehensive description based on macro- and microscopic characters, photographs and comparisons with morphologically similar and phylogenetically related species are provided.

The genus Minuartia is represented in Turkey by 34 taxa. Some interesting specimens were collected from Antalya province, and examined. These specimens resemble Minuartia meyeri, and M. multinervis from which differ by characters (macro-, and micromorphological) of inflorescence, alar pedicels, petals, sepals, capsules and seeds. Moreover, by using the DNA sequences of the ITS genes, phylogenetic relationships between this collected species, and the related species were investigated. As a result of the evaluation of molecular, and morphological data, we proposed to described the population from Antalya as a new species for the science. A description, pictures, distribution, habitat, and IUCN category are given.

The beta factor, which interacts with the rpL32 promoter, binds to the sequence 5′-GAGCCGGAAGTG and trans-activates this gene. Comparison of the DNA sequences bound by the beta factor with those bound by other known DNA-binding proteins revealed that the ETS proteins interact with similar DNA sequences. Consequently we have examined the relationship of the beta factor to the several ETS proteins so far reported. Antibody and oligonucleotide competition experiments, performed by using electrophoretic shift analysis, revealed that the beta factor contains ETS epitopes and that it is immunologically related to both of the GA-binding proteins (GABPs), implying that the beta factor may consist of two separate protein subunits.
