The antioxidant level of commercial tinctures from three Lamiaceae plants, Salvia officinalis, Mentha piperita, and Melissa officinalis, have been determined by the Folin-Ciocalteu method, the 2.2-diphenyl-2-picrylhydracyl hydrate (DPPH) radical scavenging technique and ABTS assay. Total phenolic content was expressed as GAE (gallic acid equivalent) and ranged from 0.24 to 3.99 mg/mL. Antioxidant activity in the ABTS assay, calculated as TEAC (Trolox equivalent antioxidant capacity), ranged from 23.5 to 35.6 μmol Trolox/mL, while in the DPPH method, the EC50 value ranged from 0.04 to 0.07 mL/assay. Radical scavenging activity was correlated with total phenolic content. Correlations between ABTS and F-C methods, DPPH and F-C methods and ABTS and DPPH methods were calculated. The obtained results can be useful as additional information about the antioxidant activity of galenical preparations.

AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.

Dried Pleurotus ostreatus mushrooms stored in polythene and polypropylene packs were exposed to gamma radiation from a cobalt-60 source at doses of 0, 0.5, 1, 1.5, and 2 kGy at a dose rate of 1.7 kGy/hr and stored for a period of 1 month. Total phenolic contents, flavonoids, and free radical scavenging activity DPPH (2,2′-diphenyl-1-picrylhydrazyl) were determined using aqueous, ethanol, and methanol extracts by Folin-Ciocaultaeu method as a source of potential natural antioxidants. Total phenol content ranged 0.56 ± 0.01–10.96 ± 1.7 mg/GAE, flavonoids ranged 1.64 ± 0.05–8.92 ± 0.6 mg/QE, DPPH radical scavenging activity also ranged 7.02 ± 0.10–13.03 ± 0.04%, and IC50 values also ranged 0.08–0.16 mg/mL. Statistical differences (P < 0.05) were recorded for the extracts and the treatment doses of mushrooms stored in polythene and polypropylene packs. A significant linear correlation was confirmed between values for the total phenolic content and antioxidant activity of mushroom extracts. The high contents of phenolic compounds indicated that these compounds contribute to high antioxidant activity. Pleurotus ostreatus can be regarded as a promising candidate for natural mushroom sources of antioxidants with high value. The use of low dose gamma radiation by the local food industry could improve the hygienic quality, extend shelf-life, and preserve nutrients and antinutrients.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

Three different style cuts of minimally processed sweetpotatoes (shredded, French-fry, and sliced) were stored at 0 °C and 5 °C for 4 and 8 days. Total phenolic content, individual phenolic acids, and free radical scavenging activity were determined using Folin-Denis reagent, reversed-phase HPLC, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) methods, respectively. Total phenolic content in sliced cut sweetpotatoes held at 5 °C was higher than in the shredded cut. Both sliced and French-fry cut sweetpotatoes held at 5 °C had significantly higher antioxidant activity than shredded cut sweetpotatoes. All treatments, except shredded sweetpotatoes held at 0 °C, had significantly higher total phenolic content and antioxidant activity after 4 and 8 days of storage. Minimally processed sweetpotatoes held at 5 °C accumulated more phenolic compounds and had a higher antioxidant activity than sweetpotatoes held at 0 °C. Chlorogenic acid followed by 3,5-dicaffeoylquinic acid were the predominant phenolic acids present in sweetpotatoes. The rate of increase in individual phenolic acid content with storage time was higher at 5 °C than at 0 °C. No tissue browning was observed in any of the cuts after 8 days of storage and the products were considered to be marketable.

Antioxidant potential of Rosa L. leaves methanolic extracts was evaluated in vitro using a spectrophotometric method based on measuring the radical scavenging effect on 2,2-diphenyl- 1-picrylhydrazyl (DPPH) radicals. The contents of ellagic acid, quercetin and kaempferol in the extracts from leaves of seventeen rose species were determined using SPE-RPHPLC methods. Additionally, total phenolic content was determined spectrophotometrically according to the Folin-Ciocalteu procedure and calculated as gallic acid equivalents (GAE). Remarkable high antioxidant activity and high total phenolic content (5.7% < GAE < 15.2%), large ellagic acid (EA) content from 9.37 to 19.42 mg/g of dry weight, a quercetin content ranging from 3.68 to 15.81 mg/g of dry weight and kaempferol content from 1.25 to 9.41mg/g of dry weight were found in rose leaves. Significant correlation between EA (r2 = 0.6131), quercetin (r2= 0.5158), total phenolic content (r2= 0.8485) and antioxidant activity was observed. Basing on the studies conducted one may assume that the extracts of rose leaves are a rich source of natural antioxidants and could be used to prevent free-radical-induced deleterious effects.

Abstract Polyphenolic compounds reportedly produce physiological effects that are beneficial to health. Bee products are particularly rich in polyphenolic compounds. We determined the antioxidant capacity and the phenolic and flavonoid compounds content of 81 samples of honey. We used the Folin-Ciocalteu reagent method to evaluate the total phenolic content. The antioxidant activities were evaluated using in vitro scavenging assays of 2,2-diphenyl-1-picrylhydrazyl (DPPH ) and hydroxyl radicals (OH ), Trolox equivalent antioxidant capacity (TEAC ), and ferric-reducing antioxidant capacity (FRAC ). Total phenolic content ranged from 40.3 to 193.0 mg gallic acid equivalents (GAE )/100 g; the flavonoid content varied from 1.4 to 7.5 mg quercetin equivalents (QE)/100 g. Eucalyptus honeys exhibited significantly higher phenolic content and free radical-scavenging activity than clover honey samples (p<0.05 for all). Principal component analysis explained 73% of the differences observed in antiradical activity with respect to floral origin. Total phenolic content may be more useful than the radical-scavenging assay for detecting antioxidant capacity in honey; it also represents the variable that most appropriately discriminated among these honeys.

All parts of N. nucifera are used as oriental medicine for various medicinal purposes. The aimed of this study was to determine phenolic compound and antioxidant activity of different parts of three lotus (N. nucifera including of Roseum Plenum (RP), Album Plenum (AP), and Hindu Lotus (HL)). Total phenolic content using Folin-Ciocalteu reagent method and free radical scavenging activities using DPPH assay were determined. Three deferent parts of lotus (leaves, stem, and flower) were determined. The result indicated that Roseum Plenum leaf showed the highest total phenolic contents and Roseum Plenum flower showed the highest antioxidant activities. Total phenolic content of different part of different of N. nucifera showed a significant difference (p < 0.05). It also was not uniform when compare between different of N. Nucifera. The results found that the total phenolic content was the opposite of antioxidant activity. This result may be attributed to antioxidant activity may not be from phenolic content. Therefore, it is important to research and develop the potential of lotus extraction in the future.

Penelitian ini bertujuan untuk menentukan aktivitas antioksidan dan anti glikasi ekstraks fenolik bebas dan ekstrak fenolik terikat dari tongkol jagung. Tongkol jagung diekstraksi secara sekuensial berbantu ultrasonikasi dengan pelarut etanol dan aseton selama 60 menit yang untuk mendapatkan ekstrak fenolik bebas. Residu dari ekstraksi fenolik bebas dihidrolisis dengan NaOH 2 M dan dinetralkan serta diekstraksi dengan etil asetat untuk mendapatkan ekstrak fenolik terikat. Ketiga ekstrak tersebut dilakukan analisis kandungan total fenolik dan pengujian aktivitas antioksidan dan anti glikasi. Kandungan total fenolik ekstrak  ekstrak fenolik terikat (EFT), ekstrak fenolik bebas aseton (EFBA) dan ekstrak fenolik bebas etanol (EFBE) berturut-turut adalah 288,39; 100,32 dan 92,95 μg/mL yang dinyatakan sebagai ekuivalen asam galat. Ekstrak EFT menunjukkan aktivitas penangkalan radikal bebas DPPH dan total antioksidan lebih tinggi daripada ekstrak EFBA dan ekstrak EFBE. Sejalan dengan itu, aktivitas anti glikasi dari EFT, EFBA dan EFBE berturut-turut adalah 61,93; 64,42 dan 66,31%. Hasil penelitian ini menyimpulkan bahwa ekstrak EFT dari tongkol jagung mengandung senyawa yang memilki sifat antioksidan dan berpotensi sebagai anti glikasi. ABSTRACTThis objective of this research was to determine the antioxidant activity and inhibition of AGEs formation of free phenolic and bound phenolic extract from corn cobs. Corn cobs were extracted sequentially assisted by ultrasonication with ethanol and acetone solvents for 60 minutes to obtain free phenolic extract. The residue from free phenolic extraction was hydrolyzed with 2 M NaOH and neutralized and extracted with ethyl acetate to obtain bound phenolic extract. The three extracts were analyzed for total phenolic content and evaluated for antioxidant activity and inhibitory activity of AGEs formation. The total phenolic content of bound phenolic extract (EFT), acetone-free phenolic extract (EFBA) and ethanol-free phenolic extract (EFBE) were 288,39; 100,32 and 92,95 μg/mL expressed as gallic acid equivalents, respectively. EFT extract showed higher DPPH free radical scavenging activity and total antioxidants more than EFBA extract and EFBE extract. Anti-glication of EFT, EFBA and EFBE were 61,93; 64,42 dan 66,31%, respectively. These result concluded that the EFT extract from corn cobs contains compounds having antioxidant properties and potential as anti-glication.

The purpose of this study was to determine antioxidant activity of polyphenol and phlorotannin extracts from brown algae Sargassum polycystum. Polyphenol was extracted with ethanol 96% then tested total phenolic content, while phlorotannin was extracted with methanol, chloroform, aquabidest, and ethyl acetate then tested total phlorotannin content. After crude polyphenol and crude phlorotannin extracts were obtained, the studied compounds were extracted using column chromatography. The extracts were then analyzed with HPLC (High Performance Liquid Chromatograpys). Antioxidant activity was analyzed by radical scavenging activity (RSA) and ferrous ion chelating ability (FIC) for four samples, crude polyphenol, crude phlorotannin, polyphenol, and phlorotannin. The result of total phenolic content was 1.18±0.67 mg GAE/g dry sample and total phlorotannin content was 0.61±0.27 mg PGE/g dry sample. Based on RSA and FIC analysis, phlorotannin (IC50 1.12±0.02 mg/mL and 1.34±0.01 mg/mL) showed the highest activity followed by crude phlorotannin (IC50 1.20±0.01 mg/mL and 1.47±0.10 mg/mL), polyphenol (IC50 1.23±0.01 and 1.55±0.02 mg/mL), and crude polyphenol (IC50 1.27±0.01 mg/mL and 1.63±0.02 mg/mL). Setelah memperoleh ekstrak polifenol dan florotanin, kedua senyawa tersebut dipurifikasi dengan kromatografi kolom. Hasil kromatografi kolom dianalisis menggunakan HPLC (High Performance Liquid Chromatography). Uji aktivitas antioksidan dilakukan dengan metode radical scavenging activity (RSA) dan ferrous ion chelating ability (FIC) terhadap empat sampel, yaitu ekstrak polifenol, ekstrak florotanin, polifenol, dan florotanin. Kandungan total fenolik yang diperoleh adalah 1,18±0,67 mg GAE/g ekstrak kering, sedangkan kandungan total florotanin diperoleh 0,61±0,27 mg PGE/g ekstrak kering. Berdasarkan uji RSA dan FIC, florotanin (IC50 1,12±0,02 mg/ mL dan 1,34±0,01 mg/mL) memiliki aktivitas tertinggi diikuti ekstrak florotanin (IC50 1,20±0,01 mg/mL dan 1,47±0,10 mg/mL), polifenol (IC50 1,23±0,01 dan 1,55±0,02 mg/mL), dan ekstrak polifenol (IC50 1,27±0,01 mg/mL dan 1,63±0,02 mg/mL). Hasil analisis kromatografi menunjukkan ekstrak S. polycystum mengandung senyawa fenolik relatif seperti senyawa floroglusinol.Kata kunci: Sargassum polycystum, antioksidan, polifenol, florotanin

In this study highlights on the optimization of extraction process parameters of total phenolic content and antioxidant activity from Spirulina platensis, through ultrasonication (probe) technology. Especially, optimization of ultrasonication parameters was carried out employing Box-Behnken Design (BBD) and response surface methodology. Alongside the three levels of extraction parameters i.e. solvent volume, extraction time and frequency have been fixed. As responses, the total yield of extract, total phenolic content and DPPH radical scavenging activity (IC50) of the extracts were determined. The variability of each response variable was determined by developing multiple linear regressions. The statistical analysis (ANOVA) of developed mathematical models allowed the prediction of the behaviour of the responses, as a function of the variables involved in the process. The optimized extraction conditions were obtained at 58.76 kHz frequency, 10 mL of solvent volume for 6 min. Simultaneously this extract exhibited the highest content of total phenolic content (30.89 mg GAE/g powder), IC50 for DPPH activity (151.27 µg/mL). The extract exhibited an important antioxidant i.e. caffeic acid, which was quantified as 660.72±41.05 µg/g of dry algal powder using HPLC. In spite of fact that, this study offers an alternative method for obtaining natural antioxidants from algae for food and pharmaceutical applications. Keywords: Microalgae; Probe sonication extraction; Total phenolic content; Antioxidant activity
