Abstract Background The dry body of the Tokay Gecko (Gekko gecko) is the source of a valuable traditional Chinese medicine, it is therefore listed as a Class II protected animal species in China. Due to increasing market demand and a declining supply of the species, a considerable number of adulterants have emerged in the market. Thus, it is necessary to establish an accurate and rapid method of identification for distinguishing G. gecko from its adulterants and for separating it from highly processed products. Methods A total of 274 COI sequences were analyzed by using MEGA 5.0 software. Several specific primers were designed to amplify mini-barcode regions and identify G. gecko from its counterfeits and products. Results 274 COI sequences of G. gecko and 15 adulterants species were analyzed. G. gecko could be distinguished from its adulterants through BLAST analysis, intra- and inter-specific distance analyses, and an NJ tree based on COI sequences. Two pairs of specific primers designed for this study, COISF2/COISR2 and COISF3/COISR3, amplified 200- and 133-bp fragments of the COI region, respectively, both of which were suitable for the identification of G. gecko and its adulterants. Furthermore, COISF3/COISR3 detected G. gecko in 15 batches of products. Conclusion Therefore, the specific DNA mini-barcoding method developed here may be a powerful tool for the identification of G. gecko and counterfeits, and may also be used to distinguish G. gecko from its highly processed by-products.

Arrowroot is an alternative food substitute that can be used as processed flour or starch. This arrowroot can also produce several processed products such as arrowroot chips. The number of arrowroot requests from various regions causes the need for accurate calculations related to the volume of harvest from the arrowroot. Fuzzy logic is a method that can be used to predict arrowroot yields every period to meet market demand. The parameters used in this system are based on environmental data (temperature humidity, climate, altitude), genetic data (age and variety), and cultivation technique data (seed quality, fertilizing, planting media). The results of this study are in the form of an application to predict the volume of arrowroot crop yields based on these parameters. From the results of MAPE, get a percentage of 11.7% which indicates that the level of accuracy using the fuzzy cheng time series model is said to be useful for forecasting on arrowroot plants.

Abstract Introduction: Leptospirosis affects a wide range of mammals, humans, and even a few poikilothermic animal species. In Pakistan, serological studies of equine leptospirosis have reported a prevalence of over 40%, but no study has ever been conducted towards molecular detection of Leptospira in horses. Material and Methods: Blood samples from 128 horses were screened using ELISA and 41 positive samples were examined for the presence of leptospiral DNA using specific primers for 16S rRNA gene. Results: Out of 41 tested samples, 20 samples were found to be PCR-positive, revealing a fragment of 306 bp after gel electrophoresis. Sequencing and phylogenetic analysis of positive samples revealed circulation of pathogenic Leptospira spp. in Pakistani horses. No evidence of circulation of intermediate species was found in this study. Conclusion: This study reports the first molecular evidence of equine leptospirosis in Pakistan and lays ground for further research in this area. It also confirms the efficiency of 16S rRNA for the diagnosis of equine leptospirosis.

Due to higher market demand of buffalo milk paneer, lower price cow milk is often adulterated with higher cost buffalo milk for preparation of paneer. Till date no rapid technique is available in market to ensure that paneer is made from buffalo milk. Currently a PCR based method has been developed to authenticate the buffalo milk paneer. DNA was isolated from paneer by DNeasy Mericon food kit. A set of bovine specific primers (P1) targeting D-loop (displacement loop) of mt- DNA was selected and standardized to amplify cow DNA resulted 126bp amplicon. Using this PCR based approach even upto 1% level of cow milk adulteration in buffalo milk paneer could be detected.

ABSTRACT Garlic is a vegetable rich in starch and in aromatic substances; it has high value as a spice and is consumed as a fresh vegetable or in processed products. The garlic market demand has been growing in the last years, and virus-free garlic seed is a technology that can result in better quality products. The objective of this work was to evaluate the quality of virus-free garlic cultivars (VFGC) grown under high altitude conditions in the Semiarid region of the Northeast of Brazil. Three experiments were conducted in 2018 in Portalegre, state of Rio Grande do Norte, Brazil: the first experiment with first-generation VFGC (G1), the second with second-generation VFGC (G2), and the third with infected cultivars (without clonal cleaning). The experiments were conducted in randomized block experimental design, with four replications. The treatments consisted of five garlic cultivars (Amarante, Branco-Mossoró, Cateto-Roxo, Gravatá, and Hozan). The use of virus-free garlic seeds results in bulbs with larger diameter and higher soluble solid and total solid contents, pungency, and industrial yield, showing that the seed health is important for the quality of garlics for fresh consumption and industrial processing. The cultivars Amarante, Cateto -Roxo, and Hozan had the highest total solid contents; and the cultivar Hozan had the largest bulb diameter and the highest soluble solid contents, pungency, and industrial index. The cultivar Hozan is the most indicated for dehydration due to its higher industrial index. Bulb diameter, pungency, and industrial index presented positive correlations with soluble solids, pungency, and industrial index.

In line with the staggering number of about 14 percent of India's GDP relying on the food processing industry. Our area of research aims at exploring the wide spread applications of the use of ozone a chemical compound that has the property of being a strong oxidant along with its potent capabilities of being a disinfecting agent in the food processing industry. Specifically focusing on the implication of the ozonized based processing techniques on the food and animal related processed products which include goat skin and manufacturing of starch. During the phase of the research we compared the impacts, advantages on using Ozone based processing technique over the existing methods like salt curing, and starch manufacturing from tapioca starch involves peeling, washing, rasping, screening, dewatering, drying, pulverization, dry screening and packaging. Ozonised skins kept exposed to atmospheric air were more impervious to microbial activity is in existence. This opens up the likelihood of applying Ozonisation innovation to protect goatskins so as to take out earth unsafe salt curing. And improving the whiteness property of sago starch without creating any harmful by products using Ozonisation based techniques.
