AbstractThe consumption of energy-dense diets has contributed to an increase in the prevalence of obesity and its comorbidities worldwide. The adoption of unhealthy feeding habits often occurs at early age, prompting the early onset of metabolic disease with unknown consequences for reproductive function later in life. Recently, evidence has emerged regarding the intergenerational and transgenerational effects of high-fat diets (HFD) on sperm parameters and testicular metabolism. Hereby, we study the impact of high-fat feeding male mice (F0) on the testicular metabolome and function of their sons (F1) and grandsons (F2). Testicular content of metabolites related to insulin resistance, cell membrane remodeling, nutritional support and antioxidative stress (leucine, acetate, glycine, glutamine, inosine) were altered in sons and grandsons of mice fed with HFD, comparing to descendants of chow-fed mice. Sperm counts were lower in the grandsons of mice fed with HFD, even if transient. Sperm quality was correlated to testicular metabolite content in all generations. Principal Component Analysis of sperm parameters and testicular metabolites revealed an HFD-related phenotype, especially in the diet-challenged generation and their grandsons. Ancestral HFD, even if transient, causes transgenerational “inherited metabolic memory” in the testicular tissue, characterized by changes in testicular metabolome and function.

In intense-light systems, the traditional discrete optical components lead to high complexity and high cost. Metasurfaces, which have received increasing attention due to the ability to locally manipulate the amplitude, phase, and polarization of light, are promising for addressing this issue. In the study, a metasurface-based reflective deflector is investigated which is composed of silicon nanohole arrays that confine the strongest electric field in the air zone. Subsequently, the in-air electric field does not interact with the silicon material directly, attenuating the optothermal effect that causes laser damage. The highest reflectance of nanoholes can be above 99% while the strongest electric fields are tuned into the air zone. One presentative deflector is designed based on these nanoholes with in-air-hole field confinement and anti-damage potential. The 1st order of the meta-deflector has the highest reflectance of 55.74%, and the reflectance sum of all the orders of the meta-deflector is 92.38%. The optothermal simulations show that the meta-deflector can theoretically handle a maximum laser density of 0.24 W/µm2. The study provides an approach to improving the anti-damage property of the reflective phase-control metasurfaces for intense-light systems, which can be exploited in many applications, such as laser scalpels, laser cutting devices, etc.

Apoptosis is a crucial process in spermatogenesis, responsible for the elimination of abnormal sperm cells and testicular regression out of breeding season. The aim of this study was to assess if the expression of apoptosis-related genes in testicular tissue of domestic cats differed: (1) between normozoospermic and teratozoospermic donors, and (2) between reproductive and non-reproductive season. The expression of genes: BCL2L1, BCL2, BAX, BAD, FAS, FASLG, and caspases (CASP3, CASP8, CASP9, and CASP10) was analyzed by qRT-PCR in testicular tissue samples. During non-reproductive season significantly higher expression of two anti-apoptotic genes (BCL2L1 and BCL2) was observed. Additionally, there was a significant higher expression of CASP10 in teratozoospermic cats during non-reproductive than during reproductive season. No differences were noted between normozoospermic and teratozoospermic groups. Upregulation of some genes during the non-reproductive season indicates engagement of apoptotic mechanisms in the seasonal changes of semen quality in cats, however further studies on protein levels and analysis of changes on distinct testicular germinal layers are required. At the same time, teratozoospermia in the general population of cats seems to be not connected with dysregulation of apoptosis in the testes.
