Abstract: Malnutrition because of protein deficiency on diet caused antioxidant deficiency state and oxidative stress on brain that marked by increased MDA level. The aim of this study was to analyze the antioxidant effects of saluang (Rasbora spp.) on brain MDA level in malnourished rats. This was an experimental study with a Posttest Only with Control Group Design, consisted of malnutrition control group (M), group that feed with saluang (S), group that given standard feeding (P), and group that given standard feeding added with DHA supplement (D). Brain MDA level measured with TBARS method in spectrophotometer. Mean brain MDA level for each group in a row is M = 210,750 μΜ, S = 194,125 μΜ, P = 202,625 μΜ, and D = 200,875 μΜ. Kruskal-Wallis and Mann Whitney test showed that there were significant differences between the group that feed with saluang and the other groups (p<0.05). Based on this research, concluded that the administration of saluang (Rasbora spp.) significantly affects the decrease of brain MDA level in malnourished rats. Keywords: antioxidant, saluang fish, MDA, malnutrition Abstrak: Malnutrisi karena defisiensi protein akan menimbulkan keadaan kekurangan antioksidan dan menimbulkan stres oksidatif pada otak yang ditandai dengan peningkatan kadar MDA. Tujuan penelitian ini untuk menganalisis efek antioksidan ikan saluang terhadap kadar MDA otak tikus putih malnutrisi. Penelitian ini merupakan penelitian eksperimental dengan Posttest Only with Control Group Design, terdiri dari kelompok kontrol malnutrisi (M), kelompok pemberian pakan saluang (S), kelompok pemberian pakan standar (P), dan kelompok pemberian pakan standar yang ditambah suplemen DHA (D). Kadar MDA otak diukur dengan menggunakan metode TBARS secara spektrofotometer. Rerata kadar MDA otak masing-masing kelompok perlakuan berturut-turut adalah M = 210,750 μΜ, S = 194,125 μΜ, P = 202,625 μΜ, dan D = 200,875 μΜ. Uji Kruskal-Wallis dan uji Mann Whitney menunjukkan bahwa terdapat perbedaan bermakna antara kelompok yang diberi pakan saluang dengan kelompok lainnya (p<0,05). Berdasarkan penelitian yang dilakukan dapat diambil kesimpulan bahwa pemberian ikan saluang (Rasbora spp.) berpengaruh secara bermakna terhadap penurunan kadar MDA otak tikus putih malnutrisi. Kata-kata kunci: antioksidan, ikan saluang, MDA, malnutrisi

Background:While the pathophysiology of chronic disorders varies there are three basic mechanisms - inflammation, oxidative stress and endothelial dysfunction – that are common in many chronic diseases. These mechanisms, which have a dynamic structure, are key to homeostasis. However, the failure of these mechanisms to work synchronously can lead to morbidity complicating the course of many chronic diseases.Objectives:To evaluate the effect of anti-atherosclerotic liquid (AAL), anti-inflammatory capsules (AIC) and anti-oxidant liquid (AOL) supplementation on the flow-mediated dilatation (FMD), inflammatory, oxidative stress and endothelial dysfunction markers in patients with selected chronic diseasesMethods:We analyzed data of 178 patients from cohorts with selected chronic diseases (Rheumatoid arthritis, familial Mediterranean fever, DM type-2, Hypertension, Multiple sclerosis, Chronic obstructive pulmonary disease, Alzheimer disease and Cancer) in this quasi-experimental study. Endothelial dysfunction was determined by FMD and serum asymmetric dimethylarginine (ADMA) levels. Serum ADMA, high sensitive C-reactive protein (hs-CRP), serum PTX3, malondialdehyde (MDA), Cu/Zn-superoxide dismutase (Cu/Zn-SOD), glutathione peroxidase (GSH-Px) levels and FMD were studied in baseline and after 12 weeks of Morinda citrifolia (AAL, 3 ml once per day), omega-3 (AIC, 3 capsules once per day) and extract with Alaskan blueberry and 21 different red purple fruit vegetables (AOL, 30 ml once per day). Stepwise multivariate regression analysis evaluated the association of FMD with clinical and serologic parameters.Results:Serum ADMA, MDA, PTX3, hsCRP and albumin levels, and proteinuria were significantly decreased and CuZn-SOD, GSH-Px and FMD levels were significantly increased following AAL, AIC and AOL therapies. FMD was negatively correlated with serum ADMA, MDA, PTX3, hsCRP levels, SBP and DBP and positively correlated to CuZn-SOD and eGFR levels both at baseline and after the 12-weeks treatment period. Multivariate regression analysis revealed that ADMA and PTX3 levels were independently related to FMD both before and after AAL, AIC and AOL therapies (Table 1, Figure 1).Conclusion:Our study shows that serum ADMA, MDA, PTX3 levels are associated with endothelial dysfunction in patients with selected chronic diseases. Short-term AAL, AIC and AOL therapies significantly improves FMD and normalizes ADMA, PTX3, hsCRP and MDA. AbstractThe aim of this experimental study was to assess the effects of the acute administration of L-arginine alone and in combination with L-NAME (a non-selective NO synthase inhibitor) on the coronary flow and oxidative stress markers in isolated rat hearts. The experimental study was performed on hearts isolated from Wistar albino rats (n=12, male, 8 weeks old, body mass of 180-200 g). Retrograde perfusion of the isolated preparations was performed using a modified method according to the Langendorff technique with a gradual increase in the perfusion pressure (40–120 cmH2O). The following values were measured in the collected coronary effluents: coronary flow, released nitrites (NO production marker), superoxide anion radical and the index of lipid peroxidation (measured as thiobarbiturate reactive substances). The experimental protocol was performed under controlled conditions, followed by the administration of L-arginine alone (1 mmol) and L-arginine (1 mmol) + L-NAME (30 μmol). The results indicated that L-arginine did not significantly increase the coronary flow or the release of NO, TBARS and the superoxide anion radical. These effects were partially blocked by the joint administration of L-arginine + L-NAME, which indicated their competitive effect. Hence, the results of our study do not demonstrate significant effects of L-arginine administration on the coronary flow and oxidative stress markers in isolated rat hearts.
