Biodiversitas 19: 1035-1043. A molecular study was carriedout to investigate the potential of the cytochrome c oxidase subunit I (COI) gene as a molecular marker for the genetic identification ofshark fin samples that have gone through various preservation processes. A total number of 17 shark fin samples were collected fromlocal markets in Sabah and Sarawak (Malaysian Borneo). The DNA sequences of the 17 samples were amplified by using polymerasechain reaction. The results from the analysis showed that, in the 17 sequences, there were 16 haplotypes present, with 244 sites from 688bp of the sequences. For phylogeny analysis, tree topologies were reconstructed using the neighbor-joining (NJ) and maximumparsimony (MP) methods. DNA barcoding technique successfully identifies shark fins collected in local markets in Malaysian Borneo atspecies level employed during this study. Phylogenetic analysis showed that there were four clades that distinguish the four differentorders present in the sample species. These clades had bootstrap values higher than 80. In addition, results indicated that 88.2% of theindividuals are listed as endangered (Lamiopsis tephrodes, Sphyrna mokarran, and Sphyrna lewini), vulnerable (Alopias pelagicus andRhynchobatus australiae), and near threatened (Carcharhinus limbatus, Chiloscyllium griseum, Carcharhinus sorrah, and Carcharhinusbrevipinna), in the International Union for Conservation of Nature (IUCN) Red Data List.

A new species of the synallactid sea cucumber genus Pseudostichopus is described, P. aemulatus sp. nov., based on genetic (DNA sequences of the mitochondrial gene Cytochrome Oxidase I [COI] gene) and morphological characters. A comparative molecular study with two other species of the same genus (P. villosus and P. mollis) and from a different family (Isostichopus fuscus) was carried out in order to clarify its taxonomic identity. The nucleotide distance between P. aemulatus sp. nov. and P. villosus and P. mollis is sufficient to support distinct species status. The estimated difference in the number of amino acids, coded for by a partially sequenced COI gene, within the species of the family Synallactidae ranged from 4 to 18. The phylogenetic analysis clearly supports separate species status of these sympatric morphotypes, as indicated by the morphological analysis.

Abstract Bats are mammals of great ecological and medical importance, which have associations with different pathogenic microorganisms. DNA barcoding is a tool that can expedite species identification using short DNA sequences. In this study, we assess the DNA barcoding methodology in bats from the Colombian Northern region, specifically in the Córdoba department. Cytochrome oxidase subunit I (COI) gene sequences of nine bat species were typified, and their comparison with other Neotropic samples revealed that this marker is suitable for individual species identification, with ranges of intra-species variation from 0.1 to 0.9%. Bat species clusters are well supported and differentiated, showing average genetic distances ranging from 3% between Artibeus lituratus and Artibeus planirostris, up to 27% between Carollia castanea and Molossus molossus. C. castanea and Glossophaga soricina show geographical structuring in the Neotropic. The findings reported in this study confirm DNA barcoding usefulness for fast species identification of bats in the region.

Peacock bass or the cichlids are known locally as top predator fishes which are invasive in Malaysia freshwater system. Detection probabilities for these fishes are typically low, especially using conventional capture-survey method due to the fish’s behaviour of hiding beneath the water’s surface. Hence, the environmental DNA (eDNA) monitoring is a relatively new approach that can be used to assess the distribution of these invasive fishes. Here, we report the strategy to develop small fragment (280- 400 bp) specific-specific primers for three selected invasive Cichla species namely, C. ocellaris, C. monoculus, and C. kelberi based on mitochondrial DNA (mtDNA) sequences. Current research showed that the developed species-specific primers from cytochrome oxidase I (COI) gene has high resolution at species level. Species-specific amplification tests also proved the specificity of the developed primers, securing the high- level species identification potential which may help in controlling the spread of alien invasive fish species.

The wild betta fish is a potential ornamental fish export commodity normally caught by traders or hobbyists in the wild. However, the population of wild betta in nature has declined and become a threat for their sustainability. This research was conducted to analyze the genetic diversity, phylogenetic relationships, and molecular identification through DNA COI gene sequence of Indonesian wild betta fish. A total of 92 wild betta fish specimens were collected in this study. Amplification of COI genes was carried out using Fish F1, Fish R1, Fish F2, and Fish R2 primers. The genetic diversity and phylogenetic relationships were analyzed using MEGA version 5 software program. Species identification of the specimen was conducted using BLAST program with 98-100% similarity value of the DNA sequences to indicate the same species. Phylogenetic tree construction showed seven monophyletic clades and showed that Betta smaragdina was the ancestral species of genus Betta in Indonesian waters. Genetic distance among species ranged from 0.02 to 0.30, whereas intra-species genetic distance ranged from 0 to 6.54.

Omalonyx unguis (d’Orbigny, 1837) is a semi-slug inhabiting the Paraná river basin. This species belongs to Succineidae, a family comprising a few representatives in South America. In this work, we provide the first record for the species from Misiones Province, Argentina. Previous records available for Omalonyx in Misiones were identified to the genus level. We examined morphological characteristics of the reproductive system and used DNA sequences from cytochrome oxidase subunit I (COI) gene for species-specific identification. These new distributional data contribute to consolidate the knowledge of the molluscan fauna in northeastern Argentina.
