Hydatidosis/cystic echinococcosis (CE) caused by Echinococcus granulosus is a parasitic zoonotic disease worldwide, threatening animal health and production and public health safety. However, it is still unclear that whether E. granulosus infection can result in the alteration of gut microbiota in Tibetan sheep. Therefore, a study was designed to investigate the influences of E. granulosus infection on gut microbiota of Tibetan sheep. A total of 10 ovine small intestinal contents (five from healthy and five from infected) were obtained and subjected to high-throughput sequencing by MiSeq platform. A total of 2,395,641 sequences and 585 operational taxonomic units (OTUs) were identified. Firmicutes and Proteobacteria were the most dominant phyla in all samples. Moreover, the proportions of Armatimonadetes and Firmicutes in the infected Tibetan sheep were significantly decreased, whereas Actinobacteria, Chloroflexi, and Acidobacteria had significantly increased. At the genus level, the Christensenellaceae_R-7_group and Ruminococcaceae_NK4A214_group were the predominant bacterial genera in all the samples. Furthermore, the healthy Tibetan sheep exhibited higher abundances of Intestinimonas, Butyrivibrio, Pseudobutyrivibrio, Ruminococcaceae, Eubacterium_coprostanoligenes_group, Oxobacter, Prevotella_1, Ruminiclostridium_6, Coprococcus_1, Ruminococcus, Lachnospiraceae_UCG-002, Olsenella, and Acetitomaculum, whereas Kocuria, Clostridium_sensu_stricto_1, Slackia, Achromobacter, and Stenotrophomonas levels were lower. In conclusion, our results conveyed an information that E. granulosus infection may cause an increase in pathogenic bacteria and a decrease in beneficial bacteria. Additionally, a significant dynamical change in gut microbiota could be associated with E. granulosus infection.

Abstract Background The species complex of Echinococcus granulosus sensu lato (s.l.) causes cystic echinococcosis distributed worldwide. There is no genotype information from hydatid cysts in the intermediate hosts in Central Iran. Therefore, in this study, we analyzed the hydatid cysts in livestock slaughtered in an abattoir in this region. Six hundred fifty-seven hydatid cysts were isolated from 97 animals, including sheep, cattle, camels, and goats slaughtered in Yazd abattoir from September 2018 to January 2020. The demographic data was collected as well as cyst location, fertility, and viability. Out of 657 samples, 164 samples were genotyped. Then, phylogenetic analysis was performed using MEGAX. Statistical analyses were done using SPSS version 16.0 by chi-square with a significant difference of less than 0.05. Results Out of 164 samples, the G1-G3 complex genotype had the most frequency in samples, with 135 cases recognized. The G6/G7 was observed in 19 isolates and G5 was reported in nine samples. One sample was detected as Taenia hydatigena. Conclusions This study showed that G1-G3 and G6/G7 genotypes were presented in all animals, but G5 was reported only in cattle, goats, and camels. It is the first molecular identification of cystic echinococcosis in Central Iran. Hence, reporting G5 in livestock in this area should be considered due to transmission to humans.

Abstract Background There is ample evidence demonstrating a reverse relationship between helminth infection and immune-mediated diseases. Accordingly, several studies have shown that Echinococcus granulosus infection and hydatid cyst compounds are able to suppress immune responses in allergic airway inflammation. Previous studies have documented the ability of hydatid cysts to suppress aberrant Th2 immune response in a mouse model of allergic asthma. However, there is a paucity of research on the effects of protoscoleces on allergic asthma. Thus, this study was designed to evaluate the effects of somatic antigens of protoscoleces in a murine model of allergic airway inflammation. Methods Ovalbumin (OVA)/aluminum hydroxide (alum) was injected intraperitoneally to sensitize BALB/c mice over a period of 0 to 7 days, followed by challenge with 1% OVA. The treatment group received somatic antigens of protoscoleces emulsified with PBS on these days in each sensitization before being challenged with 1% OVA on days 14, 15, and 16. The effects of somatic antigens of protoscoleces on allergic airway inflammation were evaluated by examining histopathological changes, the recruitment of inflammatory cells in the bronchoalveolar lavage, cytokine production in the homogenized lung tissue (IL-4, IL-5, IL-10, IL-17, and IFN-γ), and total antioxidant capacity in serum. Results Overall, administration of somatic antigens of protoscoleces exacerbated allergic airway inflammation via increased Th2 cytokine levels in the lung homogenate, recruitment of eosinophils into bronchoalveolar lavage fluid, and pathological changes. In addition, total antioxidant capacity and IFN-γ levels declined following the administration of somatic antigens. Conclusions The results revealed that the co-administration of somatic products of protoscoleces with OVA/alum contributed to the exacerbation of allergic airway inflammation in BALB/c mice. Currently, the main cause of allergic-type inflammation exacerbation is unknown, and further research is needed to understand the mechanism of these interactions. Graphical Abstract

This study was conducted to investigate the effect of a number of drugs on the viability of primary protoscolices isolated from sheep infected with the larval stage of E. granulosus. These drugs included Oxfendazole (OFZ) at a concentration of 0.05 mg/ml, mixed with Praziquantel (PZQ) at a concentration of 4 mg/ml and Albendazole (ABZ) at a concentration of 1 mg/ml. The results showed that OFZ was the most effective in influencing the viability of the protoscolices. The percentage of killing after five minutes of treatment was 85.63% and it was 17.21%, 15.10% and 13.09% for OFZ+ABZ, OFZ+PZQ and ABZ+PZQ, respectively. After 30 minutes, the killing percentage was 99.04%, 71.63%, 40.19% and 31.09%, respectively. The percentage of killing reached 100% after 35 minutes, 40 minutes, 2 hours and 3 hours, respectively. The results confirm the use of OFZ as a lethal drug, or a viability-suppressant for protoscolices in the future, after conducting more researches to know the side effects of this drug in laboratory animals for access to the best treatment to those infected with hydatid cysts.

Human cystic echinococcosis (CE) is a zoonotic disease caused by the larval stage of Echinococcus granulosus sensu lato that causes economic losses by affecting livestock and also poses a public health threat worldwide. The present study is the first retrospective report on the seroprevalence of anti-E. granulosus antibodies in humans in Pakistan. The study used data from 93 blood analysis reports of patients suspected of having CE from different medical centers in Lahore, Pakistan. Out of 93 sera samples, 20 (21.5%) were seropositive, and higher seropositivity (17.2%) was recorded with the indirect hemagglutination test (IHA) than with enzyme-linked immunosorbent assay (ELISA). The findings indicated that age, gender, and year had no significant relationship with the seropositivity of CE. The current study provides directions towards the management of the disease in the near future in Pakistan.
