An efficient and eco-friendly cetrimonium bromide promoted one-pot synthesis of 1-amidoalkyl-2-naphthols by the multicomponent condensation of aldehydes, β-naphthol and amides in water is reported. The wide range of substrate scope, excellent yield, short reaction rate, operational simplicity and environmentally friendly water as medium are the prominent feature of this protocol.

Abstract 3-Cyanopiperidin-2,6-diones are key intermediates in the synthesis of several drugs and natural products. We developed  one-pot synthesis of 3-cyanopiperidin-2,6-diones by Knoevenagel condensation of cyanoacetic acid and aldehydes, followed by addition of cyanoacetic acid, Achmatowicz reaction and decarboxylation. We tested a wide range of aliphatic and aromatic aldehydes. Products were obtained under simple, environmentally friendly conditions and in yields up to 99%. Substrates were stirred in toluene at reflux, and filtered through short pad of silica gel, and the product was purified by recrystallization.

Numerous O (oxa)- and S (thia)-glycosyl esters and their analogous glycosyl acids have been accomplished through stereoselective glycosylation of various peracetylated bromo sugar with benzyl glycolate using InBr3 as a glycosyl promotor followed by in situ hydrogenolysis of resulting glycosyl ester. A tandem glycosylating and hydrogenolytic activity of InBr3 has been successfully investigated in a one-pot procedure. The resulting synthetically valuable and virtually unexplored class of β-CMGL (glycosyl acids) could serve as an excellent potential chiral auxiliary in the asymmetric synthesis of a wide range of enantiomerically pure medicinally prevalent β-lactams and other bioactive molecules of diverse medicinal interest.

An efficient and practical protocol for the geminal heterodihalogenation of methyl ketones by using readily available dimethyl sulfoxide and a combination of HCl and HBr is reported. Control experiments suggested that the acidity of the solution, as well as the oxidizing ability and nucleophilicity of the dimethyl sulfoxide might work cooperatively in ensuring the success of the tandem substitution. Its operational simplicity, easy accessibility, and mild oxidative conditions suggest that the present strategy might be useful for the assembly of bromochloromethyl functional groups in drug discovery.

ZnO nanoparticles as a worthwhile and recyclable catalyst have been used for the one-pot synthesis of 1,8-dioxo-decahydroacridines and 1,8-dioxooctahydro-xanthenes via multi-component reactions under solvent-free conditions. The presented method is mild, environmentally friendly, inexpensive and highly effective to give the products in good to excellent yields.

An ethylenediamine-promoted three-component synthesis of 3-(thiomethyl)indoles from indoles, thiophenols, and paraformaldehyde has been developed. Water is used as the green solvent in a simple and environmentally friendly procedure. Stable and low-toxicity paraformaldehyde is used as a carbon source.
