On detecting interference from endogenous antibodies in immunoassays by doubling dilutions test

AbstractThe clinical laboratory may perform additional test or tests to ascertain the integrity of immunoassay analyses when the results are in doubt. Doubling serial dilutions is one of these confirmatory tests. Doubling dilutions is a useful and informative test for detecting inaccuracy and potential interference from endogenous antibodies in immunoassays. Despite its technical simplicity, the science underpinning this test is highly complex. Appreciating such complexities could help in understanding the various outcomes of such tests from misleadingly inaccurate but virtually linear/parallel results, despite the presence of interfering antibodies in some immunoassays, to an array of non-linearity/non-parallelism synonymous with interference in others. A lack of parallelism/linearity could identify some 60% of samples with inaccurate and potentially misleading results caused by endogenous interfering antibodies. Assessing real-time binding kinetics using techniques such as non-linear regression analysis or linearised transformations such as the Scatchard plot could help in identifying more samples in which the doubling dilutions test remains linear and parallel.Clin Chem Lab Med 2007;45:851–4.

Purpose. To examine the association between fracture risk assessment tool (FRAX) scores and coronary artery calcification (CAC) score in adults. Methods. The medical records of 81 adults who underwent both coronary computed tomography and bone mineral density (BMD) studies in a package during their health exams were reviewed at a regional hospital in Southern Taiwan. Data collected included health history, anthropomorphic characteristics, clinical laboratory results, and BMD. Fracture risk was determined using FRAX. Univariate and multivariate linear regression analysis were used to assess the association between CAC score and 10-year probability of hip fracture (HF) and major osteoporotic fracture (MOF) determined by FRAX. Results. The mean age of the patients was 55.8 years, and 63.0% were male. Univariate linear regression analysis showed that increases in MOF and HF risks, as measured by FRAX, were significantly and positively associated with CAC score. Multiple linear regression analysis adjusting for potential confounders showed that CAC score remained significantly associated with four FRAX indicators, including right MOF (r=0.45, P<0.001), left MOF (r=0.31, P=0.021), right HF (r=0.38, P=0.001), and left HF (r=0.23, P=0.049). Conclusions. Increased risks of MOF and HF as determined by FRAX were significantly and independently associated with CAC score.

This paper proposed a new method to describe, compare, and classify the traffic congestion points in Beijing, China, by using the online map data and further revealed the relationship between traffic congestion and land use. The data of the point of interest (POI) and the real-time traffic was extracted from an electronic map of the area in the fourth ring road of Beijing. The POIs were quantified based on the architectural area of the land use; the congestion points were identified based on real-time traffic. Then, the cluster analysis using the attributes of congestion time was conducted to identify the main traffic congestion areas. The result of a linear regression analysis between the congestion time and the land use showed that the influence of the high proportion of commercial land use on the traffic congestion was significant. Also, we considered five types of land use through performing a linear regression analysis between the congestion time and the ratio of four types of land use. The results showed that the reasonable ratio of land use types could efficiently reduce congestion time. This study makes contributions to the policy-making of urban land use.

AbstractAs it is common practice for laboratories to store patient samples for a predefined period, allowing clinicians to request additional tests on previously collected samples, knowledge about sample stability is indispensable for the laboratorian. A common approach to estimating the maximum storage time is to use a discrete study design, measuring the analyte of interest at various time-points and then checking for significant differences with the help of a statistical test, such as Student’s t-test, Wilcoxon’s test or an analysis of variance (ANOVA) test. Because only discrete time intervals are considered, stability data can just be approximated. Alternatively, a continuous study design, as described by the Clinical and Laboratory Standards Institute (CLSI) for performing stability experiments for in vitro diagnostic reagents, can also be adopted by the clinical laboratory to evaluate the stability of biological samples. The major advantage of this approach is that it allows laboratories to define individual stability limits for different medical situations and offers more flexibility when choosing time-points for measurements. The intent of this paper is to demonstrate the evaluation of sample stability in the clinical laboratory with a continuous study design implemented with linear or non-linear regression analysis. Appropriate statistical modeling and acceptance criteria are presented, stability functions are described briefly, and checking the overall validity of the results is discussed.

Abstract Background Since the COVID-19 pandemic, several therapeutic agents have been used in COVID-19 management. However, the results were controversial. Here, we aimed to evaluate the efficacy and safety of hydroxychloroquine (HCQ)/chloroquine (CQ) in COVID-19. Methods We retrospectively reviewed the medical charts of patients with COVID-19 admitted to an inpatient ward in Wuhan from 2020/Feb/08 to 2020/Mar/05. Patients with HCQ/CQ and age, gender, disease severity matched ones without HCQ/CQ were selected at a 1:2 ratio. The clinical, laboratory and imaging findings were compared between these two groups. The multivariate linear regression analysis was performed to identify the factors that might influence patients’ virus shedding periods (VSPs). Results A total of 14 patients with HCQ/CQ and 21 matched ones were analyzed. The HCQ/CQ treatment lasted for an average of 10.36 ± 3.12 days. The mean VSPs were longer in the HCQ/CQ treatment group (26.57 ± 10.35 days vs. 19.10 ± 7.80 days, P = 0.020). There were 3 patients deceased during inpatient period, two patients were with HCQ/CQ treatment (P = 0.551). In the multivariate linear regression analysis, disease durations at admission (t = 3.643, P = 0.001) and HCQ/CQ treatment (t = 2.637, P = 0.013) were independent parameters for patients’ VSPs. One patient with CQ had recurrent first-degree atrioventricular block (AVB) and obvious QTc elongation, another one complained about dizziness and blurred vision which disappeared after CQ discontinuation. One patient with HCQ had transient AVB. Conclusions In summary, we identify that the HCQ/CQ administration is not related to less mortality cases at later phase of COVID-19. More studies are needed to explore whether HCQ/CQ treatment would lead to SARS-Cov-2 RNA clearance delay or not.

This study aims to determine the effect of Inflation, Exchange Rate and Composite Stock Price Index (IHSG) to Return of PT Nikko Securities Indonesia Stock Fund period 2014-2017. The study used secondary data obtained through documentation in the form of PT Nikko Securities Indonesia Monthly Net Asset (NAB) report. Data analysis is used with quantitative analysis, multiple linear regression analysis using eviews 9. Population and sample in this research are PT Nikko Securities Indonesia. The result of multiple linear regression analysis was the coefficient of determination (R2) showed the result of 0.123819 or 12%. This means that the Inflation, Exchange Rate and Composite Stock Price Index (IHSG) variables can influence the return of PT Nikko Securities Indonesia's equity fund of 12% and 88% is influenced by other variables. Based on the result of the research, the variables of inflation and exchange rate have a negative and significant effect toward the return of PT Nikko Securities Indonesia's equity fund. While the variable of Composite Stock Price Index (IHSG) has a negative but not significant effect toward Return of Equity Fund of PT Nikko Securities Indonesia

XX in Jakarta and to identify variables that have a dominant influence on the productivity of employees. The research method used is using multiple linear regression analysis. The results show that the all variables simultaneously and partially change the organizational structure, job design, and organizational culture has a significant impact on employee productivity at PT. XXin Jakarta.

Some students perceive lesbian, gay, bisexual, and transgender (LGBT) positively, even though LGBT is a sexual deviation that is not appropriate with values and norms. There are several factors that influence an individual's perception of LGBT, including sexual identity. This study aims at looking at the contribution of sexual identity to student perceptions about LGBT. This research used a quantitative approach with a descriptive method and a simple linear regression analysis. The sample of this research was 385 taken from 15.752 undergraduate students of Universitas Negeri Padang which the sample was drawn by using the Slovin formula and continued with a Proportional Random Sampling technique. The instrument used was the Guttman model's sexual identity scale and the scale of students' perceptions of the LGBT Likert model. After analyzing the data with the descriptive technique and the simple linear regression analysis, the results showed that sexual identity significantly contributed to the students' perceptions of LGBT. This research has implications as a basis for counselors to help students avoid sexual identity mismatches and prevent the emergence of positive perceptions of LGBT.

The problem in this study is whether there is an influence of leadership and work discipline on the employees’ performance of PT.Sucofindo Palembang City. This research includes associative research. The sample in this study were 88 respondents, with propotionate random sampling analysis technique. The data used were primary data and secondary data. Data collection method through questionnare. Analysis techniques using multiple linear regression analysis, F test (Simultaneoys) and t test (partial) and determination coeffiecient. The results show that there is influence of leadership and work discipline on the performance of PT.Sucofindo Palembang City.
