AbstractFor the first time, nuclear microsatellite (nSSRs) primers were developed in the endangered tree species Lecythis ampla (Lecythidaceae) as molecular tools. An enrichment protocol with genomic DNA libraries for nSSRs was used to identify candidate loci. A large number of candidate loci were identified. Consecutively population genetic parameters of these loci were tested in two available populations. Eventually 17 microsatellite loci have been identified that show no or only low evidence for linkage disequilibrium, deviations from Hardy-Weinberg expectations or high levels of null alleles. These markers are apt for future molecular population studies.

Abstract Most initially perfect flowers of Toona ciliata Roem subsequently develop into functionally unisexual flowers and their relative positions in the same inflorescence could enhance the outcrossing system in this species. Here we investigated the mating system of this species. We used eight nuclear microsatellite markers and investigated the progeny of 125 mother trees from six populations naturally distributed in South China, with sample sizes ranging from 64 to 300 seeds. The multilocus outcrossing rate was 0.970 ± 0.063, and the single locus outcrossing rate was 0.859 ± 0.106, indicating the pattern of predominant outcrossing. Selfing was present in one population, but biparental inbreeding occurred in five populations. Inbreeding was absent in maternal parents, and correlations of selfing among families or among loci were generally insignificant. Positive correlation of paternity at multiple loci was significant in four populations, but was not consistent with the results at single loci. Population substructure occurred in male similarity between outcrosses only in one population. Population genetic differentaitaion was significant (Fst = 34.5%) and the effects of isolation-by-distance at the eight loci were significant among the six populations. These results provide evidence that self-comptability and inbreeding naturally occur in T. ciliata and indicate that inbreeding avoidance is necessary during genetic improvement and breeding of this endangered tree species.

Abstract Background Astronium fraxinifolium is an endangered tree species from Brazil. Due to its high importance for environmental reforestation, as well as for the use of its wood, it is necessary to implement management programs for conservation of this species. Simple sequence repeats (SSR) or microsatellite markers have been widely used in population genetic studies across diverse organisms. In this study, we reported for the first time SSR markers for A. fraxinifolium as well as its frequency and distribution from transcriptome data. Results More than 125 thousand RNA-seq sequences derived microsatellites, with predominant distribution of trinucleotides repeats. From the initial screening, we selected 20 microsatellite loci, validated and evaluated genetic indexes in two natural populations. All loci were polymorphic, ranging from four to eleven alleles per locus. The observed and expected heterozygosities ranged from 0 to 1.0 and from 0.533 to 1.0, respectively. Genetic differentiation between populations (= 0.363) showed higher diversity within than among populations. Conclusions The developed SSR loci from RNA-seq consists in a base for future studies of genetic diversity and population structure, mating system and gene flow in A. fraxinifolium populations as well as related species, aiming the conservation and management of the species.

This study was conducted at Pulau Jerejak, Penang to determine the floristic variation of its tree communities. A 0.5-hectare study plot was established and divided into 11 subplots. A total of 587 trees with diameter at breast height (DBH) of 5 cm and above were measured, identified and recorded. The tree communities comprised of 84 species, 63 genera and 32 families. The Myrtaceae was the most speciose family with 10 recorded species while Syzgium glaucum (Myrtaceae) was the most frequent species. The Myrtaceae recorded the highest density of 306 individuals while Syzgium glaucum (Myrtaceae) had the highest species density of 182 individuals. Total tree basal area (BA) was 21.47 m2/ha and family with the highest BA was Myrtaceae with 5.81 m2/ha while at species level, Syzgium glaucum (Myrtaceae) was the species with the highest total BA in the plot with value of 4.95 m2/ha. The Shannon˗Weiner Diversity Index of tree communities showed a value of 3.60 (H'max = 4.43) and Evenness Index of 0.81 which indicates high uniformity of tree species. The Margalef Richness Index (R') revealed that the tree species richness was 13.02. Myrtaceae had the highest Importance Value of 20.4%. The Canonical Correspondence Analysis (CCA) showed that Diospyros buxifolia (Ebenaceae) and Pouteria malaccensis (Sapotaceae) were strongly correlated to low pH. Dysoxylum cauliflorum (Meliaceae) and Eriobotrya bengalensis (Rosaceae) were correlated to phosphorus (P) and calcium ion (Ca2+), respectively. Therefore, the trees species composition at Pulau Jerejak showed that the biodiversity is high and conservation action should be implemented to protect endangered tree species. A invasive rugose spiraling whitefly (RSW) <em>Aleurodicus rugioperculatus</em> Martin (Hemiptera: Aleyrodidae) was found infesting coconut, banana, custard apple and several ornamental plants in Tamil Nadu, Andhra Pradesh and Kerala for the first time in India. The identity of the pest species was determined through morphological and molecular tools. Furthermore cytochrome c oxidase-I gene (658 bp) of RSW was sequenced (GenBank accession number KY209909) which would serve as an ideal molecular diagnostic marker for its identification irrespective of its phenotypic plasticity. During the survey, several natural enemies were recorded and maximum parasitism was recorded by <em>Encarsia guadeloupae</em> Viggiani (Hymenoptera: Aphelinidae) and its COI gene was sequenced and deposited as <em>Encarsia</em> sp. (GenBank accession number KY223606). Per cent parasitism ranged from 20.0 to 60.0 % in different collection locations, highest parasitism being recorded in Kerala as compared to other states. The predators recorded were <em>Mallada</em> sp., few coccinellids and predatory mites. This communication is the first report of the rugose spiraling whitefly, its host plant range and associated natural enemies in India.

Increased rate of soil salinity caused changes in macro and micro nutrients uptake, which may lead to decline in photosynthesis capacity and respiration in the plant Syzgium travancoricum Gamble. is a critically endangered species located in damp forest with marshy land, where high rate of salt accumulation has been happening. An optimum level of salinity (0.05 - 0.07) is inevitable for the proper growth and survivability of this species in their natural habitat and also for natural regeneration. A range of reduction in the number of existing plants of this species can be seen in their natural habitat due to the salinity stress.
