Components and Antioxidant Activity of the Polysaccharide from Streptomyces virginia H03

A polysaccharide was isolated from the broth of cultured Streptomyces virginia H03 which was treated by ethanol deposition and savage method to remove the protein, and was purified using Sephadex G-150 column chromatography. The components of the polysaccharide were determined by gas chromatography. The purified polysaccharide was made up of mannose, glucose and galactose, in a 2:1:1 proportion. Its average apparent molecular weight was 3.76 · 104 Da which was determined by gel permeation chromatography. In addition, several antioxidant assays were adopted to investigate the antioxidant activity of the polysaccharide in vitro. The results indicated that the purified polysaccharide showed significant antioxidant activity against superoxide anion, hydrogen peroxide and 1,1-diphenyl-2-picrylhydrazyl radical, and lipid peroxidation as with standard antioxidants such as vitamin C. Furthermore, the polysaccharide had a better heat stability than vitamin C, which suggested that the polysaccharide might be a potent useful antioxidant

<p>Assessment of antioxidant activity was imperative in the screening of medicinal plants for potential health benefits. In present study methanol extract of <em>Polygonum glabrum</em> <em> </em>(polygonaceae) was screened for its in vitro antioxidant activity using biologically relevant methodologies which scavenge radicals such as  1,1 diphenyl 2 picryl hydrazyl (DPPH), nitric oxide, hydrogen peroxide, hydroxyl,  superoxide anion and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS). Total reducing ability by conversion of ferric (III) to ferrous (II) and molybdenum (VI) to molybdenum (V), metal ion chelating capacity and anti lipid peroxidation activities were also examined. The antioxidant ability of <em>Polygonum glabrum</em> whole plant extract was found to be in a dose dependent manner. The IC<sub>50</sub> values for scavenging of DPPH<sup>●</sup> and ABTS<sup>●+</sup> free radicals were 13.18 μg/ml and 20.46 μg/ml. For scavenging of nitric oxide, hydrogen peroxide, hydroxyl and superoxide anion radicals, the IC<sub>50</sub> values were found to be 80.22 μg/ml, 33.06 μg/ml, 52.26μg/ml and 36.98 μg/ml respectively. Further, addition of 120μg/ml of plant extract to the reaction mixture produced 50% lipid peroxidation inhibition activity. Commercial antioxidants such as vitamin E, quercetin, butylated hydroxytoluene and ascorbic acid were used as reference compounds. The strong antioxidant activity of <em>Polygonum glabrum</em> may be credited to the presence of triterpenes [beta-hydroxyfriedalanol], phenols        [3-hydroxy-5-methoxystilbene], flavonoids [pinocembrin and pinocembrin-5-methylether], steroids [sitosterol - (6-O-palmitoyl)- 3-O-β-D glucopyranoside and sitosterol-3-O-β-D glucopyranoside], sesqueterpenes [2,3-dihydroxy isodrimeninol] and pigments etc in methanol extract.</p>

A protein of apparent molecular weight 280,000 (syncolin), which is immunoreactive with antibodies to hog brain microtubule-associated protein (MAP) 2, was purified from chicken erythrocytes. Immunofluorescence microscopy of bone marrow cells revealed the presence of syncolin in cells at all stages of erythrocyte differentiation. In early erythroblasts syncolin was diffusely distributed throughout the cytoplasm. At later stages it was found along microtubules of the marginal band, as confirmed by immunoelectron microscopy. The association of syncolin with the marginal band was dependent on the integrity of microtubules, as demonstrated by temperature-dependent de- and repolymerization or marginal band microtubules. Syncolin cosedimented in a saturable manner with microtubules assembled in vitro, and it was displaced from the polymer by salt. Brain as well as erythrocyte microtubules, reconstituted with taxol from MAP-free tubulin and purified syncolin, were aggregated into dense bundles containing up to 15 microtubules, as determined by electron microscopy. On the ultrastructural level, syncolin molecules were visualized as globular or ringlike structures, in contrast to the thin, threadlike appearance of filamentous MAPs, such as brain MAP 2. According to ultrastructural measurements and gel permeation chromatography, syncolin's molecular weight was approximately 1 x 10(6). It is suggested that syncolin's specific function is the cross-linking of microtubules in the marginal band and, by implication, the stabilization of this structure typical for nucleated (chicken) erythrocytes.

This study examines the antioxidant activity of crude phlorotannins from the brown algae Sargassum hystrix v. buxifolium (Chauvin) J. Agardh, through the inhibition of a lipid peroxidation reaction that is induced by the UV radiation. The antioxidant activity during the UV exposure was investigated using the laser-based photoacoustic method for the detection of the ethylene as indicator for lipid peroxidation. This involves an experiment that isolated crude phlorotannins from the ethyl acetate fraction of the Sargassum hystrix methanol extract, hereafter referred to as PFSH. It results in the antioxidant activity as a potent lipid peroxidation inhibitor. Statistically, such antioxidant activity is not significantly different than the commercial antioxidant, which is vitamin C (p > 0.05). The amount of the total phlorotannins, using the Folin-Ciocalteu method, was measured to be approximately 0.13% w/w. The radical copolymerization of resorcinol (RSC) and 3,5-dihydroxyl benzoic (DHBA) was carried out in water by the initiator of horseradish (HRP)/H2O2. It was discussed that the effects of monomer composition on the properties of the copolymer. The best monomer mass ratio of RSC:DHBA was 60:40 and the shrink temperature can reached to 88.5oC. The tanning result indicated that the copolymer of RSC and DHPA has particularly excellent tanning properties and can be served as leather tannage substitute for chrome tanning materials. And also the mechanism of the HRP initiated copolymerization was proposed. The structure and molecular weight of the copolymer was characterized by Fourier Transform Infrared spectroscopy (FTIR), Nuclear Magnetic Resonance (NMR) and Gel Permeation Chromatography (GPC). The copolymer can be as tannage and retannage in making leather process. The results showed that it has excellent tanning properties and retanning effects.

Gel permeation columns of Bio Beads S-X8 have been used to provide separation of oligomers and other small organic molecules. Results show successful separations up to molecular weight c. 600. The retention times of compounds have been correlated with the largest molecular dimension of the molecules and also with molar volumes.

Dialdehyde starches with different aldehyde content from native corn starch were prepared by sodium periodate oxidation to be used as a tanning agent in leather making. For this purpose, native corn starch was oxidized with sodium metaperiodate in different molar ratios. After oxidation processes, the yields, solubility in water and aldehyde contents of the obtained dialdehyde starches were determined as well as structure characterizations by Proton Nuclear Magnetic Resonance Spectroscopy, Fourier Transform Infrared Spectroscopy and Gel Permeation Chromatography. Evaluating the gel permeation chromatography data, the dialdehyde starch samples which were thought to be in appropriate molecular weight/size to penetrate into skin fibers were selected to be used in the tanning process. Their tanning abilities were evaluated by investigating hydrothermal stabilities, filling and fiber isolation characteristics and physical properties determined by mechanical tests and organoleptically. From the evaluation of the results, it was revealed that sodium metaperiodate oxidized starches which have appropriate molecular weight and adequate aldehyde content has a remarkable tanning effect and can be utilized as a tanning agent with the advantages of not necessitating pickling process which means saving time and simplifying the production but more importantly offering an important advantage from an environmental point of view.
