Fruit maturity stage and provenance affect seed germination of Passiflora mollissima (banana passion fruit) and P. ligularis (sweet granadilla), two commercially valuable tropical fruit species

Orphan crops, which include many of the tropical fruit species used in the juice industry, lack genomic resources and breeding efforts. Typical of this dilemma is the lack of commercial cultivars of purple passion fruit, Passiflora edulis f. edulis, and of information on the genetic resources of its substantial semiwild gene pool. In this study, we develop single-nucleotide polymorphism (SNP) markers for the species and show that the genetic diversity of this fruit crop has been reduced because of selection for cultivated genotypes compared to the semiwild landraces in its center of diversity. A specific objective of the present study was to determine the genetic diversity of cultivars, genebank accession, and landraces through genotyping by sequencing (GBS) and to conduct molecular evaluation of a broad collection for the species P. edulis from a source country, Colombia. We included control genotypes of yellow passion fruit, P. edulis f. flavicarpa. The goal was to evaluate differences between fruit types and compare landraces and genebank accessions from in situ accessions collected from farmers. In total, 3820 SNPs were identified as informative for this diversity study. However, the majority distinguished yellow and purple passion fruit, with 966 SNPs useful in purple passion fruits alone. In the population structure analysis, purple passion fruits were very distinct from the yellow ones. The results for purple passion fruits alone showed reduced diversity for the commercial cultivars while highlighting the higher diversity found among landraces from wild or semi-wild conditions. These landraces had higher heterozygosity, polymorphism, and overall genetic diversity. The implications for genetics and breeding as well as evolution and ecology of purple passion fruits based on the extant landrace diversity are discussed with consideration of manual or pollinator-assisted hybridization of this species.

Guava (Psidium guajava L.) is a tropical fruit exhibiting rapid post-harvest ripening. However, the physiological basis involved in the ripening process of guava is not totally clear, which makes it difficult to develop technologies to enhance fruit storability. Two experiments were carried out with the objective of determining the ripening behavior of 'Pedro Sato' guavas. In the first experiment, guava fruits at three maturity stages (I - dark green, II - light green and III - yellow-green) were stored at room temperature (23 ± 1°C and 85 ± 5 % RH). The respiratory rate, ethylene production, pulp and skin colours, and firmness were evaluated. In the second experiment, ethylene and 1-methylcyclopropene (1-MCP) were applied to guavas at the light green maturity stage and the ripening behaviour during storage at room temperature was studied. Fruits from all maturity stages showed a gradual increase in the respiratory rate and ethylene production. The intense changes in pulp and skin colours and in firmness preceded the maximum respiratory rate and ethylene production. 1-MCP reduced the rate of ripening, while the application of ethylene did not promote this process. These results do not permit the classification of 'Pedro Sato' guava as a traditional climacteric fruit.

Brazil has a large variety of native and exotic fruit species, including the gabiroba, which can be sources of income for the population. The objective of this study was to evaluate the post-harvest behavior of gabiroba fruit by maturity stage and storage temperature. The fruits were divided into two lots and stored at temperatures of 6, 12, 24 and 30°C. The first batch was used for the physical and chemical analysis of acidity, soluble solids, firmness, percentage of green color, and visual analysis; this batch was composed of green and yellow-green fruit. The second lot was intended for breath analysis, composed of green, yellow-green, and yellow fruits. The experiment was conducted in a completely randomized design with five replications. Data were analyzed using descriptive statistics. Considering the evaluated characteristics, the gabiroba soluble solid/titratable acidity ratio values should be approximately 4.0 for the fruits to be considered acceptable for consumption. For the preservation of the post-harvest quality of gabiroba, it is indicated that the harvest is carried out at the green stage of maturation and that they are stored at a temperature of 6°C. The highest respiratory rates were observed in fruits harvested at the yellow stage, not being recommended its storage.

Brazil has great variability, being one of the centers of origin of the genus Passiflora. Passion fruit has a multiple and diversified uses, being little explored as an ornamental plant in the country. The objective of this work was to characterize four passion fruit genotypes (two hybrid materials from the experimental field of passion fruit breeding program of the University of Brasília, and two passion fruit cultivars - BRS Sertão Forte and BRS Mel do Cerrado, both of them from Brazil), based on official and validated morphoagronomic descriptors for the identification of morphological aspects and ornamental potential, under field conditions, in the Distrito Federal, Brazil. The experiment consisted of a randomized block design, with four treatments, four replications, and six plants per plot. Plants were analyzed weekly regarding the characteristics of the branches, leaf blade, petiole, and flowers, with 24 measurements of each structure in the four treatments, which were recorded using a digital camera. Flowers presented pronounced size and color, colored branches, and leaves of varying sizes and shapes. All materials studied showed commercial ornamental potential to be explored as new options for ornamental climbing plants.

Vasconcellea quercifolia A. St.-Hil. (Caricaceae) is a tropical fruit species native to Brazil, with a great importance in plant breeding programs. The V. quercifolia has a resistance to the main diseases of Caricaceae, Papaya Ringspot Virus (PRSV). Considering its potential, cryopreservation becomes a tool for the conservation of this species. The objective of this paper was to study the cryopreservation of V. quercifolia zygotic embryos through dehydration in silica gel. Excised zygotic embryos were dehydrated in silica at 0, 20, 40, 80 and 100 minutes and then inoculated in MS medium. The percentage of germinated and recovered embryos, and growth analysis were evaluated, besides water content. Subsequently, they were acclimatized in a growth room with temperature controlled. For cryopreservation, the embryos were excised and dehydrated in silica for 0, 20, 40 and 60 minutes, immersed in Liquid Nitrogen (LN) for 1 hour, thawed in Recovery Solution (RS) and inoculated in MS medium. After 30 days, the percentage of germinated and recovered embryos was evaluated. The silica gel promotes a fast dehydrate of embryos. The results showed that embryo dehydration affected seedling development, and dehydration for over 20 minutes showed a reduction in all evaluated parameters. The plantlets regenerated from embryos dehydrate survive the acclimatization. It was possible to cryopreserve the V. quercifolia zygotic embryos when the dehydration time of 20 minutes by silica gel was used.
