ABSTRACTSix experiments involving the artificial insemination (AI) of a total of 300 female red deer (hinds) with frozen-thawed red deer semen (collected by electro-ejaculation) were conducted over 3 years. Insemination took place at fixed times following various oestrous synchronization procedures using progesterone withdrawal and treatment with pregnant mare's serum gonadotropin (PMSG). In the 1st year, the experiments evaluated basic AI techniques in which pregnancy rates were 45% in 20 hinds receiving two inseminations per vaginam (PV) and 56% in 27 hinds inseminated by the laparoscopic intrauterine method (IU). In the 2nd year, the experiments involved comparisons of the progesterone regime, one or two PV inseminations, and the timing of a single PV insemination. There was no effect of replacement of the progesterone device after 9 days and withdrawal 3 days later compared with the use of one device for the whole period in two experiments. The pregnancy rate for a double PV insemination was significantly higher than for a single PV insemination (58 and 34%; P < 0·05) and there was also a small effect of timing of insemination relative to the synchronization treatment. In the 3rd year all hinds were inseminated by the IU method. The experiments involved a comparison of various times of AI following progesterone withdrawal and a comparison of the progesterone regime. The overall pregnancy rate for 63 hinds inseminated was 56% with no difference between three times of insemination (48, 52 and 55 h). In the second experiment, the difference in pregnancy rate between treatment with progesterone for 15 days and 12 days (44 and 72% for 18 hinds per group) was not significant, but the interaction between the length of progesterone treatment and insemination time was significant (P < 0·05), with the 12-day progesterone/55 h insemination giving a much higher pregnancy rate than the 15-day/55 h insemination (89 and 20% respectively). Although no experiments involved direct comparisons of the routes of insemination, overall pregnancy rates were 56% for IU, 53% for double PV and 35% for single PV inseminations.

The aim was to estimate the effective pharmacological method of the estrous cycle synchronization by checking the effects of synchronization by measurement of progesterone (P4) and 17-beta estradiol (E2) concentration by RIA and artificial insemination. The experiment was performed at the red deer farm in Rudzie (North-East Poland; 3 year’s old). The herd (N = 14) was kept away from bulls and was divided in two groups of seven animals. In the Group I, CIDR insert (0.3 g of P4) was applicated intravaginally for 12 days; a second insert replaced the first one for the next 12 days, and next 200 IU of equine chorionic gonadotropin (eCG) was injected intramuscularly (Folligon). Estrus was expected 48 h after eCG injection. In the Group II, Chronogest sponge (20 mg of flugestone acetate) was applicated intravaginally and after 7 days replaced with second chronogest sponge for 7 days. After removing the sponge, on the same day eCG was injected and estrus was expected after 48 h. Artificial insemination was provided with frozen-thawed semen twice: 12 and 24 h after expected estrus. The peripheral blood from the jugular vein was collected each time when the inserts or sponge were applicated and 40 days after insemination. The concentration of P4 and E2 in plasma was measured by RIA. The effectiveness of insemination was monitored by pregnancy-associated glycoproteins determination and observed by the number of calves born. Two pregnancies were confirmed in Group I and five in Group II based on PAG concentration. One newborn was observed in Group I and five in Group II. Both methods of synchronization are effective in hinds based on the received profile of steroids. Although the sponge shape in case of chronogest is better comparing with CIDR, which was not completely deposited in the vagina of hind, potentially leads to bacteria inflammation, and it disturbs the rightful endocrine regulation. Moreover, pregnancy rate and hormone responsiveness were better in Group II.

Abstract Three male F1 hybrids between Père David's deer and red deer were mated to red deer to produce 143 backcross calves. The pedigrees are a rare example of a fertile hybrid between evolutionarily divergent species. We examined the use of these families for genetic mapping of evolutionarily conserved (Type I) loci by testing for genetic linkage between five species-specific protein variants and 12 conserved DNA probes. Two probes were homologous, and the remainder syntenic, to the protein coding loci in cattle or humans. Using six restriction enzymes, each DNA probe detected one or more restriction fragments specific to Père David's deer. Linkage analyses among the species-specific variants placed the loci into four linkage groups within which linkage between adjacent loci and gene order was supported by a LOD &gt; 3. The linkage groups were (HPX, HBB)-FSHB-ACP2, LDHA-CD5-IGF2, BMP3-(GC, ALB)-(KIT, PDGFRA) and LDLR-C3-FGF1. Southern and protein analysis of LDHA and ALB provided identical segregation data. These linkage groups were consistent with the cattle gene map and provide new information for comparing the gene maps of ruminants, humans and mice. The deer hybrids are an important new resource that can contribute to the comparative analysis of the mammalian genome.

The prostaglandin F2 alpha analogue cloprostenol stimulates ovarian secretion of oxytocin in red deer hinds and Pere David's deer hinds, as in cattle and sheep, but the response of the uterus to administered oxytocin has not been studied in deer. In the present experiment, oxytocin administered intravenously caused an increase in circulating concentrations of 13,14-dihydro-15-keto prostaglandin F2 alpha from 186 +/- 35 to 404 +/- 34 pmol L-1 within 5 min; concentrations in saline-treated hinds were unchanged (150 +/- 12 and 164 +/- 12 pmol L-1 before and after treatment respectively). This suggest that in red deer as in other ruminants, a positive feedback relationship between the corpus luteum and the uterus may operate to stimulate luteolytic episodes of prostaglandin F2 alpha.
