Two Cipangopaludina snails were discovered in Harris County, Texas, USA, during routine fieldwork in October 2015. Dissection yielded one male and one female containing 52 offspring in her brood pouch. Phylogenetic analysis of the cytochrome c oxidase subunit I (COI) gene confirmed both individuals to be Cipangopaludina japonica (von Martens, 1861). This is the first distribution record of C. japonica in Texas. Non-native invasive snails, such as C. japonica, compete with native species and may serve as reservoirs for parasites, prompting the need for increased diligence in monitoring public waterways.

Dermanyssus gallinae (the poultry red mite, PRM) is an important ectoparasite in the laying hen industry. PRM can also infest humans, causing gamasoidosis, which is manifested as skin lesions characterized by rash and itching. Recently, there has been an increase in the reported number of human infestation cases with D. gallinae, mostly associated with the proliferation of pigeons in cities where they build their nests. The human form of the disease has not been linked to swallows (Hirundinidae) before. In this report, we describe an incident of human gamasoidosis linked to a nest of swallows built on the window ledge of an apartment in the island of Kefalonia, Greece. Mites were identified as D. gallinae using morphological keys and amplifying the Cytochrome C oxidase subunit I (COI) gene by PCR. Bayesian phylogenetic analysis and median-joining network supported the identification of three PRM haplogroups and the haplotype isolated from swallows was identical to three PRM sequences isolated from hens in Portugal. The patient was treated with topical corticosteroids, while the house was sprayed with deltamethrin. After one week, the mites disappeared and clinical symptoms subsided. The current study is the first report of human gamasoidosis from PRM found in swallows’ nest.

The snakehead fish (Channidae) is widely distributed in inland water of Indonesia. This fish is native species in Suma-tera, Java and Kalimantan, but non-native species in Sulawesi and Papua. Study on molecular identification and phylo-geny of this fish using cytochrome c oxidase subunit I (COI) gene has only been conducted on snakehead fish origin from Tasikmalaya, Ambarawa, Bali, Aceh, Pontianak, and Banjarmasin waters, but none is available in South Sulawesi waters. The objectives of this research are to identify species of snakehead fish from Towuti Lake Sorowako and the first generation (F1) of domesticated snakehead fish from Bantaeng waters of South Sulawesi using COI gene for DNA barcoding, and to analyze the phylogenetic resolution of the fish. Partial sequences of the COI gene of the snakehead fish were aligned with sequences of snakehead fish deposited in GenBank. The phylogenetic tree was constructed using MEGA 7.0.20 program. The result indicated that COI gene nucleotides of snakehead fish from Towuti Lake Sorowako showed 99% homology with Channa striata acc no. KU692418 and KU692421, and showed 98% homology with those of acc no. KU852443. Therefore it can be conclude that Channa striata exist in Towuti Lake Sorowako. Nucleotide sequences of the first generation (F1) of domesticated snakehead fish from Bantaeng waters showed 65% homology with Channa pleurophthalma (acc no. KJ937390) origins from Banjarmasin waters and Channa gachua (acc no. KX389277). Based on this result, it assumed that snakehead fish from Towuti Lake Sorowako are distantly in gene to those from Bantaeng waters, and further analyses are required to identify the population of snakehead fish from Bantaeng waters. Abstrak Populasi ikan gabus (famili Channidae) tersebar luas di wilayah perairan tawar Indonesia. Ikan gabus merupakan ikan asli di Sumatera, Jawa, dan Kalimantan) tetapi merupakan ikan introduksi di Sulawesi dan Papua. Identifikasi berdasar-kan gen cytochrome c oxidase subunit I (COI) telah dilakukan terhadap ikan gabus dari perairan di Tasikmalaya, Amba-rawa, Bali, Aceh, Pontianak dan Banjarmasin, tetapi ikan gabus dari perairan Sulawesi Selatan belum dilakukan. Tuju-an penelitian adalah untuk mengidentifikasi species ikan gabus dari Danau Towuti Sorowako dan ikan gabus generasi I (F1) hasil domestikasi dari induk yang berasal dari perairan tawar Kabupaten Bantaeng Sulawesi Selatan menggunakan gen COI untuk DNA barcoding, dan untuk menganalisis filogeni ikan gabus tersebut. Sekuen gen COI ikan gabus terse-but disejajarkan dengan sekuen nukleotida ikan gabus yang terdeposit di GenBank. Pohon filogenetik dikonstruksi de-ngan menggunakan program MEGA 7.0.20. Hasil penelitian menunjukkan bahwa nukleotida gen COI sampel ikan gabus dari Danau Towuti Sorowako memiliki kemiripan 99% dengan Channa striata nomor aksesi KU692418 dan KU692421 dan 98% dengan Channa striata nomor aksesi KU852443 sehingga dapat disimpulkan bahwa di Danau To-wuti terdapat ikan gabus jenis Channa striata. Tingkat kemiripan sekuen nukleotida ikan gabus F1dari perairan Kabu-paten Bantaeng adalah 65% dengan ikan gabus Channa pleurophthalma (KJ937390) asal perairan Banjarmasin dan ikan gabus Channa gachua (KX389277). Berdasarkan hasil tersebut maka diduga bahwa ikan gabus dari Danau Towu-ti Sorowako berkerabat jauh dengan ikan gabus dari perairan Bantaeng, dan diperlukan analisis yang lebih lanjut untuk menentukan jenis populasi ikan gabus F1 asal perairan tawar Kabupaten Bantaeng tersebut.

Ultrastructural and molecular data are provided from a single adult female pentastomid opportunistically collected from a road-killed rattlesnake in Russell, KS. Ultrastructural data consisted of light and SEM microscopy of the pentastomid and its eggs, while molecular data consisted of partial 18S and 28S ribosomal sequences and a partial cytochrome c oxidase subunit 1 sequence from the same specimen used for SEM. Ultrastructural and molecular data support generic identification of the pentastomid as Porocephalus sp. These molecular data were also used with previously published pentastomid sequence data for a concatenated phylogenetic analysis, which support the current, morphology-based taxonomic placement of the genus.

AbstractThe fall armyworm (FAW), an invasive pest of maize, is an emerging threat in Southern Asia after America and Europe. Recently, this notorious pest has also been found in different areas of Pakistan. To assess its presence in Pakistan, a survey was carried out in the provinces of Punjab, Sindh, and Khyber Pakhtunkhwa during May–October 2019. We observed the highest incidence of FAW in Sindh with maximum impact in districts Tando-Allahyar and Hyderabad. These samples were identified as Spodoptera frugiperda on the morphological and taxonomical bases. However, morphological identification of this pest is very difficult at early larval instars. Here, we use the mitochondrial cytochrome c oxidase I (COI) gene region for the precise identification of larva of this invasive pest at species level. Two different regions of COI gene (COI-5′ and COI-3′) were used as molecular markers for the identification of this species. DNA sequence similarity searches of the obtained COI gene sequences (NCBI GenBank Accession Nos. MW241537, MW241538, MW349515, MW349516, MW349493 and MW349494) revealed that genetically it is more than 99% identical to S. frugiperda. The phylogenetic analysis indicated it as the rice-strain (R-strain). Both 3′- and 5′-fragment tree topologies showed that the collected samples of the FAW species belong to the R-strain. To the best of our knowledge, this is the first report providing molecular evidence for the existence of R-strain of S. frugiperda that was found feeding on maize crop in Sindh, Pakistan, using COI gene sequences as a marker.

En este estudio se analizan dos fragmentos del gen de la citocromo c oxidasa subunidad I (COX1) del ADN mitocondrial de 61 individuos del género Rhynchophorus colectados en la Región de Murcia a fin de determinar su procedencia. El análisis filogenético del fragmento 1 de las muestras de la Región de Murcia conjuntamente con las secuencias disponibles en GenBank indica que los individuos corresponden a la especie Rhynchophorus ferrugineus.Las secuencias de Murcia se colapsan en un único haplotipo (H8 mediterráneo) que aparece dentro del clado de R. ferrugineus. De los análisis filogeográficos se infiere que el origen de los individuos de Murcia es Egipto. Adicionalmente, se examinó una región contigua del COX1 (fragmento 2) en la que las secuencias se colapsaron en dos haplotipos. In this research two fragments of the cytochrome c oxidase subunit I (COX1) gene of the mitochondrial DNA were analyzed in 61 individuals of the genus Rhynchophorus collected in the Region of Murcia with the aim of determining their origin. Phylogenetic analysis of fragment 1 of the samples collected in the Region of Murcia together with the available sequences in GenBank, indicated that these individuals correspond to the species R. ferrugineus. Sequences from Murcia collapsed into the H8 Mediterranean haplotype, which cluster into the R. ferrugineus clade. Phylogeographic analysis shows that the origin of the individuals collected in the Region of Murcia is Egypt. Additionally, a contiguous fragment of COX1 (fragment 2) was analyzed and the sequences collapsed into two haplotypes.

We submitted first results of the DNA studies of the Central Asiatic owlet moths of the genus Euchalcia. Standard cytochrome C oxidase subunit I (COI) gene fragments were sequenced for DNA barcoding of six specimens belonging to Euchalcia herrichi and Euchalcia gyulai. We compared the received sequences between discussed species and with two European Euchalcia species (E. variabilis and E. consona). We found no variability within the COI sequences of the samples collected in the same locality (Alai Mts., Kyrgyzstan), whereas the difference in COI sequences between two populations (Ketmen Mts., Kazakhstan and Alai Mts., Kyrgyzstan) was 0.005.

ABSTRACT Collections of mosquitoes were conducted for the surveillance of species of medical importance in the state of Morelos, Mexico, in June 2017. Species collected included Mansonia (Mansonia) dyari, which was identified using morphological characters and cytochrome c oxidase I DNA barcoding. Although 3 species of genus Mansonia have been previously reported in Mexico, this is the 1st confirmed record of Ma. dyari in Morelos State, where no Mansonia species had been recorded. Historical records of Ma. dyari and Ma. indubitans in Mexico were reviewed. Therefore, this record increases the number of mosquito species occurring in Morelos to 46. The specimens collected in this study were deposited in the Culicidae collection of the Universidad Autónoma Agraria Antonio Narro, Unidad Laguna.

Daerah Modoinding dan Tomohon di Sulawesi Utara, dikenal sebagai  daerah penghasil sayuran kubis di Indonesia. Sayuran kubis memiliki hama utama yaitu Plutella xylostella. Penyebab serangga ini dapat bertahan hingga saat ini karena adanya sifat resistensi akibat pemberian insektisida yang berlebihan. Penelitian ini dilakukan untuk menganalisis variasi pada gen cytochrome C oxidase IPlutella xylostella yang diperoleh dari dua lokasi yang berbeda, yaitu Modoinding dan Tomohon. Analisis sekuens menunjukkan adanya perbedaan pasang basa nukleotida dari sampel yang berbeda lokasi. Selain itu, variasi juga ditunjukkan pada sampel yang diperoleh dari basis data GenBank dengan adanya perbedaan 1-14 pasang basa nukleotida dengan spesimen pada penelitian ini. Hubungan kekerabatan gen COI P. xylostella keseluruhan sampel tergolong dalam variasi intraspesies dengan nilai jarak genetik berkisar antara 0-0,022 (0-2,20%).Modoinding and Tomohon areas in North Sulawesi, are known as regions in Indonesia that produce a cabbage. The main pest of cabbage, Plutella xylostella. This insect can survive due to its resistance resulted from prolonged insecticide application. This study aims to analyze genetic variation of COI genes in P. xylostella from Modoinding and Tomohon areas. Sequence analysis showed there were differences in nucleotide base pairs between these locations. In addition, variations were also shown in samples obtained from the GenBank database with differences in 1-14 nucleotide base pairs with specimens in this study.
