ABSTRACT The study was divided into three experiments. In the first one, broilers were distributed into six groups and vaccinated against infectious bursal disease at 14 days of age: T1-not vaccinated, T2-Lukert1 (intermediate), T3-Lukert2 (intermediate plus), T4-228E, T5-V877 and T6-Winterfield 2512 (“hot” strains). Bursas of Fabricius (BF) were collected at 17, 21, 28 and 35 days to measure BF relative weight (BFRW), diameter, histological examination and image processing analysis (IPA). At one, 14, 21, 28 and 35 days of age, samples of blood taken from eight birds from each group for serology analysis by ELISA test. Hot strains vaccines induced reduction of BFRW and BF diameter, higher histological score lesion degree, more lymphocyte depletion on the BF follicles and higher IBD antibody titer. In the second experiment, 16 birds from groups T1 to T6 were isolated and challenged with a very virulent strain of IBDV (vvIBDV) at 25 days of age. Only groups T4, T5 and T6 were totally protected against vvIBDV challenge. In the third experiment, the immunosuppressive potential of each vaccine was determined by examining the ability of IBDVvaccinated birds to respond to Newcastle disease (ND) vaccination and challenge. None of the vaccines was found to be immunodepressive.

Two different vaccination programs against chicken infectious bursal disease (IBD) were compared. Results showed the first program including IBD vaccination at 8 and 16th day old as well as ND vaccination(live attenuated and inactivated types ) give high antibody titers at 21 days post vaccination with significantly difference (P≤0.0005) in comparison with the second program including only IBD vaccine . serological antibodies titer were determined to study the correlation between two different programs using ELISA test. pathological changes in vaccinated groups reveals mild & moderate lesions appear in liver, spleen and bursa of fabriciusKey word : IBD,ND , vaccination , ELISA test ,Broiler , histopathology .

. Using the method of shifting assay of standardized indirect agglutination (SSIA), the prevalence of Newcastle disease viruses (NDV) and infectious bursal disease viruses (IBDV) in chickens reared in several districts of Thua Thien Hue province in the Spring-Summer and Fall-Winter seasons was determined. In the Spring-Summer season of 2011, about 22.3% of the chickens were infected with NDV, in which A Luoi  accounted for the highest percentage of 25% of the infected chickens and Huong Thuy  the lowest  of 18.2%. Meanwhile, 36% of the same chickens were infected with IBDV, with the highest percentage (46.66%) also in A Luoi and the lowest (30.3%) also in Huong Thuy. The intensity of NDV infection in the Spring-Summer season in A Luoi and Phu Vang was highest (GMT = 1.45), and in Huong Thuy lowest (GMT = 1.31). In addition, in the Fall-Winter season, about 46% of the chickens were infected with NDV and 46.3% with IBDV in Huong Thuy and Phu Vang – two neighbouring districts of Hue City, in which NDV was detected in 54.4% of the chickens in Huong Thuy and 33.9% in Phu Vang. In contrast, IBDV was detected in 41.9% and 52.7% of the chickens respectively in the two districts. The infection was not inter-dependent. Methodically, although the differences in the infection rates were insignificant with the accuracy of 95%, faecal samples showed higher sensitivity in SSIA analyses for both cases of NDV and IBDV infection in comparision with mouth exudates. By SSIA method, results could be read clearly with unaided eyes for a long time after the performance, and it was also proven applicable for cases of haemagglutinating viruses if proper treatments for depletion of animal RBCs’ surface agglutinins could be applied.

Infectious bursal disease virus (IBDV) is a bi-segmented RNA virus, which belongs to the genus Avibirnavirus of the family Birnaviridae. Two serotypes, 1 and 2, exist in IBDV. The serotype 1 IBDVs are the causative agents of infectious bursal disease (IBD) in chickens worldwide and lead to immunosuppression in young birds. Genome re-assortment has been speculated to occur and contribute to the emergence of new IBDV strains. However, evidence was lacking until recently when two re-assortant viruses were detected in China. In this study, we determined the complete nucleotide sequence of an IBDV, designated KZC-104, from a confirmed natural IBD outbreak in Lusaka, Zambia in 2004. The genome consisted of 3074 and 2651 nucleotides in the coding regions of segments A and B, respectively. Alignment of both nucleotide and deduced amino acid sequences, and phylogenetic analysis revealed that the genome segment A of KZC-104 was derived from a very virulent strain, whereas its segment B was derived from a classical attenuated strain. On BLAST search, the full-length segments A and B sequences showed 98% closest nucleotide homology to the very virulent strain D6948 and 99.8% closest nucleotide homology to the classical attenuated strain D78, respectively. This is a unique IBDV reassortant strain, which has emerged in nature involving segment B of a live attenuated vaccine. This observation provides direct evidence for the involvement of vaccine strains in the emergence of reassortant IBDV in the field. Taken together, these findings suggest an additional risk of using live IBDV vaccines, which may act as genetic donors for genome re-assortment. Further studies are required to investigate the epidemiology and biological characteristics of reassortant strains so that the appropriate and safe IBDV vaccines can be recommended.

This study was designed with a view to investigate the prevalence of poultry diseases in Gazipur district of Bangladesh. A total of 679 poultry birds (313 layers, 338 broilers and 28 cockrels) either dead or live were brought for diagnosis of diseases at Gazipur Sadar Upazilla Veterinary Hospital. The diseases were diagnosed on the basis of history, clinical signs and post-mortem findings. The diseases encountered in layers were bacterial diseases 52.29% (salmonellosis 38.56%, colibacillosis 6.7%, fowl cholera 4.79% and necrotic enteritis 1.60%), viral diseases 23.95% (avian influenza 2.56%, Newcastle disease 16.61%, infectious bronchitis 3.19% and avian leucosis 0.64%), mycoplasmal disease (mycoplasmosis 14.70%) and protozoal disease (coccidiosis 5.75%). Salmonellosis was most prevalent disease in age group of >20 weeks, while Newcastle disease most common in 8 to 20 weeks of age group. In case of broiler, bacterial diseases 28.99% (salmonellosis 21.30% and colibacillosis 7.69%), viral diseases 53.24% (infectious bursal disease 28.99%, Newcastle disease 8.87% and infectious bronchitis 15.38%), mycoplasmal disease (mycoplasmosis 7.1%) and protozoal disease (coccidiosis 6.5%). In cockrels, the most prevalent disease was colibacillosis 35.71% followed by salmonellosis 28.57%, Newcastle disease 14.28% and mycoplasmosis14.28%. So among the diseases, salmonellosis is most prevalent disease followed by infectious bursal disease and mycoplasmosis in different kinds of poultry of Gazipur district of Bangladesh.Asian J. Med. ABSTRACTObjective: The study of Mycoplasma gallisepticum (MG) infection is needed, not only to understand the disease process but also to understand theinterference with the evaluation of some live viral poultry vaccines. This study aims to investigate the titration and potency of some live attenuatedpoultry viral vaccines; Newcastle disease, infectious bronchitis, infectious bursal disease, and Reo in both specific pathogen-free (SPF) embryonatedchicken eggs (ECEs) and chickens.Methods: Titration of live attenuated viral poultry vaccines in ECEs was carried out by dividing the inoculated eggs into four groups; the pre-,simultaneously-, post-, and non-MG contaminated. MG effect on the potency test was carried out using seventeen groups of SPF chickens (25 chicken/group) placed into separate isolators. Each live attenuated viral poultry vaccine was inoculated into 4 groups.Results: The highest titer of these vaccines that appeared in MG pre- contaminated ECEs were 1011, 107.5, 107.9, and 10, respectively. The lowest vaccinetiters that appeared in non-MG contaminated ECEs were 108, 106, 106.8, and 1067.5, respectively. Although the potency of these previous vaccines indicated thatthe highest antibodies titer that appeared in MG pre-infected vaccinated chickens were 7.5 log, 36 enzyme-linked immunosorbent assay unit (EU), and42 EU, respectively; the lowest antibodies titer that appeared in non-MG infected vaccinated chickens were 6.5 log22, 12 EU, 17 EU, and 10 EU, respectively.Conclusion: The present study findings underline the importance of using Mycoplasma -free eggs or chicken for the production of virus vaccines.Keywords: Mycoplasma gallisepticum, Newcastle disease virus, Infectious bronchitis virus, Infectious bursal disease virus, Reo virus, Chicken, Specificpathogen-free eggs.
